    Organizing sustainable development

    The role and meaning of sustainable development have been recognized in the scientific literature for decades. However, there has recently been a dynamic increase in interest in the subject, which results in numerous, in-depth scientific research and publications with an interdisciplinary dimension. This edited volume is a compendium of theoretical knowledge on sustainable development. The context analysed in the publication includes a multi-level and multi-aspect analysis starting from the historical and legal conditions, through elements of the macro level and the micro level, inside the organization.     Scientific Railway Signalling Symposium 2023 - Einfach Fahren! – Digitale Transformation im Spannungsfeld Automatisierung, europäische Standardisierung und schneller Rollout

    Im Rahmen des Programms „Digitale Schiene Deutschland“ wird aktuell an der digitalen Transformation der Infrastruktur gearbeitet. Ziel ist es unter anderem, eine höhere Kapazität sowie optimale Auslastung des Schienennetzes zu erreichen, ohne im größeren Umfang zusätzliche Gleise zu verlegen. Für dieses Ziel bedarf es eines harmonisierten und reibungslosen Zusammenspiels zukunftsweisender Technologien im Bereich der Leit- und Sicherungstechnik, die zunächst umfassend erforscht und anschließend im Feld eingesetzt werden müssen. Mit dieser spannenden Thematik beschäftigte sich das Scientific Railway Signalling Symposium (SRSS) 2023 am 14. Juni, welches vom Institut für Bahnsysteme und Bahntechnik der TU Darmstadt zusammen mit der DB Netz AG sowie erstmals dem Fachgebiet Bahnbetrieb und Infrastruktur am Institut für Land- und Seeverkehr der TU Berlin organisiert wurde. Ca. 100 Fachexpert:innen aus Forschung und Praxis verfolgten die unterschiedlichen Vorträge unter dem Tagungstitel „Einfach Fahren! – Digitale Transformation im Spannungsfeld Automatisierung, europäische Standardisierung und schneller Rollout“ und brachten ihre Expertise in Diskussionsrunden ein. Zu Beginn der Tagung stellte Volker Hentschel von der DB Netz AG in seiner Keynote das volle Potenzial des digitalen Bahnsystems vor. Dabei gab er einen Überblick über die Innovationen, deren Umsetzung in den kommenden Jahren im Rahmen der Digitalen Schiene zu erwarten sind. Daran anschließend diskutierten die Kleingruppen, welche Innovationen der digitalen LST schnell zu Verbesserungen im Feld führen können. Bevor Markus Montigel, RAILvelation GmbH, zum Abschluss des Tages durch einen interaktiven Vortrag das Motto des Tages „Einfach fahren“ noch einmal aus etwas Distanz reflektierte, boten zahlreiche Beiträge detaillierte Einblicke zu unterschiedlichen LST-bezogenen Themen aus Wissenschaft und Praxis. Aufgrund der gestiegenen Anzahl der Vorträge im Vergleich zu den Vorjahren wurde die Tagung im Nachmittagsblock erstmals in parallele Sessions aufgeteilt. In dieser Publikation sind diejenigen Beiträge enthalten, die erfolgreich den Review-Prozess durchlaufen haben. Eine wesentliche Innovation ist das Automatisierte Fahren, dessen Sicherheit vor Einführung geprüft und sichergestellt werden muss. Marco Kinas et al. beschreiben daher in ihrem Beitrag Aspekte des im Rahmen des ATO-RISK-Projekts verfolgten Ansatzes, so z.B. den Einsatz von Regelwerken als Risikoakzeptanzprinzip gemäß der CSM-Verordnung und die Analyse menschlicher Faktoren im Vergleich zum derzeitigen bemannten Betrieb. Neben der funktionalen Sicherheit spielt auch die Sicherheit vor Angriffen eine wichtige Rolle. Simon Unger et al. erläutern daher in ihrem Beitrag das Zusammenspiel zwischen Safety und Security und arbeiten insbesondere die Relevanz von Security in Safety-kritischen Systemen anhand des RaSTA-Protokolls heraus. Im Gegensatz zu Safety handelt es sich bei Security um einen dynamischen Aspekt, denn die Bedrohungslage durch Angreifer kann sich stetig wandeln. Matthias Drodt von der DB Netz AG und Frank Weber vom Eisenbahn-Bundesamt beschreiben in ihrem Beitrag daher Lösungsansätze zur Zulassung der sehr dynamischen IT-Security in Eisenbahnsystemen. Für eine schnelle Zulassung und einen schnellen Rollout ist auch eine Beschleunigung des Entwicklungs- und Sicherheitsnachweisprozesses notwendig. Frederic Reiter et al. beschreiben in ihrem Paper eine vollständig virtualisierte Evaluierungsumgebung für digitale Stellwerke, die die Komplexität reduziert und Tests für Ausführung von Funktionen vor der Integration mit physischen Komponenten ermöglicht. Gunnar Smith von Prover Technology aus Stockholm stellt in seinem Beitrag die Methoden vor, die zur Reduzierung des Aufwandes in der Entwicklungsphase der LST-Systeme und dadurch auch zum schnellen Rollout beitragen werden. Julian Lucas von der TU Darmstadt und Markus Rothkehl von der DB Netz beschreiben ein Konzept für ein modulares Nachweisverfahren für verschiedene Komponenten der digitalen Sicherungstechnik. Auch im Falle eines beschleunigten Rollouts wird jedoch eine längere Übergangsphase zur digitalen LST erforderlich sein, in der es mehrere Migrationsschritte geben wird. In seinem Beitrag erläutert Melvin Zinngrebe von SBB AG zwei Strategien, die von der SBB AG zur Bewältigung der Migrationsherausforderungen angewendet werden. Eine davon ist die Lebensverlängerung der ersten Generation elektronischer Stellwerke, um Investitionen in eine Übergangstechnologie vor der Führerstandssignalisierung (FSS)-Migration zu vermeiden. Die andere Strategie ist eine Nachrüstung mit FSS. Zuverlässige Infrastrukturdaten sind eine unerlässliche Voraussetzung für die erfolgreiche Durchführung der digitalen Transformation. Benedikt Wenzel et al. haben in ihrem Beitrag die Auswirkungen auf die Qualität von Infrastrukturdaten und geeignete Strategien zur Bewältigung der Herausforderungen, die mit der Erstellung und Pflege zuverlässiger Infrastrukturdaten verbunden sind, herausgearbeitet. Um Pünktlichkeit und den reibungslosen Zugverkehr sicherstellen zu können, ist die Gewährleistung der Verfügbarkeit von LST-Systemen ein wichtiges Thema. In ihrem Paper beleuchten Simon Hofer und Martin Müller von team Technology Management GmbH verschiedene Strategien, um die Anforderungen an die hohe Verfügbarkeit beim Zugsicherungssystem ETCS gewährleisten zu können. Die Potenziale für Kapazitätssteigerungen und Leistungsverbesserungen, die durch Digitale Stellwerke, ETCS und andere Technologien erzielt werden können, sind in Deutschland nach wie vor enorm. Peter Reinhart von der DB Netz AG fasst in seinem Paper den bisherigen Erkenntnisstand hierzu anhand des Digitalen Knoten Stuttgart zusammen. Die breite Themenvielfalt der in diesem Tagungsband enthaltenen Artikel zeigt, wie dynamisch das Umfeld in unserer Branche derzeit ist und wie viele spannende Innovationen wir in den nächsten Jahren erwarten dürfen

    Spatial control of angiogenesis by engineered patterns of Notch ligands

    Spatial control of angiogenesis by engineered patterns of Notch In tissue engineering and regenerative medicine, proper vascularization of engineered tissues is imperative, as diffusion of nutrients, oxygen and waste is limited in constructs larger than a few cells thick. Therefore, tissue vascularization has been extensively researched. However, most approaches rely on preset structural support for the cells provided by scaffolds and micro fluidic chips to instruct vascular organization, which limit the integration into engineered and native tissues. Self-assembly of vascular constructs by combinations of support material, cells and growth factors shows promise but is not capable of dictating structure and organization of the engineered vasculatures. Therefore, spatial control over vascularization is required for tissue engineered constructs to remain alive and functioning. The growth of vasculature from existing vessels towards the oxygen gradient, is a process called angiogenesis. During angiogenesis, migrating tip cells emerge from the vascular endothelium and sprout towards the oxygen gradient. Proliferating stalk cells follow their path and form a vascular tube. The selection of tip and stalk cell phenotypes is regulated via several cell signaling pathways, among which the Notch signaling pathwayhas a predominant role. The Notch signaling pathway is a highly conserved cell-cell signaling pathway involved in numerous cell fate decisions and the patterning of tissues. The Notch receptor Notch1 and ligands Dll4 and Jag1 are involved in angiogenesis. Here, Dll4/Notch1 signaling is responsible for tip and stalk cell selection and Jag1 modulates Dll4/Notch1 signaling, though the role of Jag1 in angiogenesis needs to be further elucidated. Notch signaling is involved in several (genetic) cardiovascular diseases as well as cancer. For therapeutic targeting of Notch, mostly soluble molecules have been used to inhibit signaling. However, systemic administration of these molecules gives rise to adverse effects and toxicity. Local activation of Notch signaling, especially to target angiogenesis has not been widely addressed. In this thesis, we have developed an in vitro method where we used spatial patterns of parallel lines of Notch signaling ligands to locally modulate endothelial tip/stalk cell selection and thereby gained spatial control over endothelial sprouting. The technical details of the protocol, optimization, important considerations and alternative approaches are also provided. Using the developed technology we have found that line patterns of Dll4 induced controlled unidirectional and restricted sprouting on the lines, resulting in localization of endothelial sprouts between the lines (negative patterning). Next, we used our method to investigate the role of Jag1 in endothelial patterning. In contrast to Dll4, we have found that Jag1 patterns did not exert control over endothelial sprouting location and direction. Gene expression profiles did not provide significant difference upon stimulation with Dll4 and Jag1 to explain the difference in control observed.Computational modeling of Notch signaling endothelial cells on patterns of ligand, revealed that underlying differences in signaling dynamics between Jag1 and Dll4 are likely the cause for the lack of control over endothelial sprouting exerted by Jag1. To study the potential of ligand patterning further, alternative techniques for complex patterning of ligands were considered and investigated, though further optimization is needed for their application. In summary, we have shown that the use of local presentation of the Notch ligand Dll4 results in spatial control over endothelial sprouting. Additionally, the potential of engineered patterns of ligands to control endothelial sprouting during angiogenesis is valuable for the design of biomaterials and engineering of organized vasculature for tissue engineering as well as for regenerative medicine. Furthermore, the method developed in this thesis allows for the screening of functional engineered ligands and could aid in revealing disease phenotypes by screening for mutations in ligands.Spatial kontroll av angiogenes genom konstruerade mönster av Notch-ligander I vävnadsteknik och regenerativ medicin är vaskularisering av konstruerade vävnader nödvändigt, eftersom spridningen av näringsämnen, syre och avfall är begränsad i konstruktioner som är större än några celler i tjocklek. Därför har vävnadsvaskularisering i omfattande grad utforskats. De flesta infallsvinklarna är dock beroende av förbestämda stödstrukturer och mikofluidiska chip för att instruera vaskulär organisation, vilket begränsar integrationen i levande vävnader. Självkonstruktion av vaskulaturen genom kombinationer av celler och tillväxtfaktorer är lovande, men kan inte diktera strukturen och organisationen. Därför är spatial kontroll av vaskularisering nödvändig för att vävnadstekniska konstruktioner skall vara funktionella. Tillväxten av vaskulatur utgående från existerande kärl mot syregradienten är en process benämnd angiogenes. Under angiogenes framträder migrerande spetsceller från det vaskulära endotelet och rör sig mot syregradienten. Snabbväxande stjälkceller följer deras bana och bildar en vaskulär tub. Urvalet av spets- och stjälkceller styrs genom ett flertal cellsignaleringsvägar, av vilka cellsignaleringsvägen Notch har en framträdande roll. Notchsignaleringsvägen är en i hög grad konserverad cell-cell signaleringsväg inblandad i ett flertal cellödesbeslut och vävnadsmönstring. Notchreceptorn Notch1 och liganderna Dll4 och Jag1 är viktiga för angiogenes. Här är Dll4/Notch1-signalering ansvarig för spets-och stjälkcellsurval och Jag1 modulerar Dll4/Notch1-signalering. Dock bör rollen Jag1 spelar i angiogenes vidare utforskas. Notchsignalering är inblandad i flera (genetiska) hjärt- och kärlsjukdomar samt cancer. För terapeutiska interventioner inriktade på Notch har till största delen lösliga molekyler använts för att hämma signalering. Systemisk administrering av dessa molekyler ger dock upphov till skadliga effekter och toxicitet. Lokal aktivering av Notchsignalering, framförallt inriktad på angiogenes har inte i omfattande grad studerats. I denna avhandling har vi utvecklat en in vitro-metod där vi använde spatiala mönster av parallella linjer av Notchligander för att lokalt modulera urvalet av endotela spets- och stjälkceller och därmed få kontroll över endoteltillväxten. Vi har funnit att linjemönster av Dll4 inducerade kontrollerad enkelriktad och begränsad tllväxt vilket resulterade i endoteltillväxt mellan linjerna (negativ mönstring). Vi använde även vår metod för att utforska vilken roll Jag1 hade i endotelmönstring. Till skillnad från Dll4 påverkade Jag1 inte endoteltillväxtens plats och riktning. Genutrycksprofiler visade ingen signifikant skillnad efter stimulering med Dll4 och Jag1 som skulle förklara de observerade skillnaderna. Datamodellering av Notchsignalerande endotelceller på ligandmönster visade att underliggande skillnader i signaleringsdynamik mellan Jag1 och Dll4 sannolikt är orsaken till bristen på kontroll över endoteltillväxt hos Jag1. För att studera potentialen hos ligandmönstring vidare utforskades alternativa tekniker för komplex mönstring av ligander, dock krävs vidare optimering innan de kan tas i bruk I sammanfattning har vi visat att användningen av lokal presentation av Notchliganden Dll4 resulterar i spatial kontroll över endoteltillväxt. Mönster av ligander för att kontrollera endoteltillväxt vid angiogenes är värdefullt för skapandet av biomaterial och konstruktion av organiserad vaskulatur för vävnadskonstruktion samt regenerativ medicin.     A Software Vulnerabilities Odysseus: Analysis, Detection, and Mitigation

    Programming has become central in the development of human activities while not being immune to defaults, or bugs. Developers have developed specific methods and sequences of tests that they implement to prevent these bugs from being deployed in releases. Nonetheless, not all cases can be thought through beforehand, and automation presents limits the community attempts to overcome. As a consequence, not all bugs can be caught. These defaults are causing particular concerns in case bugs can be exploited to breach the program’s security policy. They are then called vulnerabilities and provide specific actors with undesired access to the resources a program manages. It damages the trust in the program and in its developers, and may eventually impact the adoption of the program. Hence, to attribute a specific attention to vulnerabilities appears as a natural outcome. In this regard, this PhD work targets the following three challenges: (1) The research community references those vulnerabilities, categorises them, reports and ranks their impact. As a result, analysts can learn from past vulnerabilities in specific programs and figure out new ideas to counter them. Nonetheless, the resulting quality of the lessons and the usefulness of ensuing solutions depend on the quality and the consistency of the information provided in the reports. (2) New methods to detect vulnerabilities can emerge among the teachings this monitoring provides. With responsible reporting, these detection methods can provide hardening of the programs we rely on. Additionally, in a context of computer perfor- mance gain, machine learning algorithms are increasingly adopted, providing engaging promises. (3) If some of these promises can be fulfilled, not all are not reachable today. Therefore a complementary strategy needs to be adopted while vulnerabilities evade detection up to public releases. Instead of preventing their introduction, programs can be hardened to scale down their exploitability. Increasing the complexity to exploit or lowering the impact below specific thresholds makes the presence of vulnerabilities an affordable risk for the feature provided. The history of programming development encloses the experimentation and the adoption of so-called defence mechanisms. Their goals and performances can be diverse, but their implementation in worldwide adopted programs and systems (such as the Android Open Source Project) acknowledges their pivotal position. To face these challenges, we provide the following contributions: • We provide a manual categorisation of the vulnerabilities of the worldwide adopted Android Open Source Project up to June 2020. Clarifying to adopt a vulnera- bility analysis provides consistency in the resulting data set. It facilitates the explainability of the analyses and sets up for the updatability of the resulting set of vulnerabilities. Based on this analysis, we study the evolution of AOSP’s vulnerabilities. We explore the different temporal evolutions of the vulnerabilities affecting the system for their severity, the type of vulnerability, and we provide a focus on memory corruption-related vulnerabilities. • We undertake the replication of a machine-learning based detection algorithms that, besides being part of the state-of-the-art and referenced to by ensuing works, was not available. Named VCCFinder, this algorithm implements a Support- Vector Machine and bases its training on Vulnerability-Contributing Commits and related patches for C and C++ code. Not in capacity to achieve analogous performances to the original article, we explore parameters and algorithms, and attempt to overcome the challenge provided by the over-population of unlabeled entries in the data set. We provide the community with our code and results as a replicable baseline for further improvement. • We eventually list the defence mechanisms that the Android Open Source Project incrementally implements, and we discuss how it sometimes answers comments the community addressed to the project’s developers. We further verify the extent to which specific memory corruption defence mechanisms were implemented in the binaries of different versions of Android (from API-level 10 to 28). We eventually confront the evolution of memory corruption-related vulnerabilities with the implementation timeline of related defence mechanisms
