    Addressing Uncertainties of Polychlorinated Biphenyls in Schools

    Despite the potential neurological health deficits resulting from polychlorinated biphenyls, there is a lack of inhalation studies and regulations to protect students and teachers who are widely exposed to PCBs in older (1950 to 1979) schools. To estimate the extent of PCB detections in Los Angeles County schools, I applied a mathematical model to estimate the range of PCB detections in schools built between 1950 and 1984. I estimate the range of 17-34% PCB detections of open schools within Los Angeles County. Next, I reviewed exposure tools provided by the US EPA to bridge the uncertainty gaps between ingestion and inhalation studies available. I identified the following uncertainties: toxicity values derived from animal studies, data gaps related to weathered PCB congeners and Aroclors, and using extrapolated measures to minimize uncertainty gaps. Lastly, I completed an extensive literature review of toxicological health effects associated with PCBs that revealed liver toxicity and its relative harm to other organs and systems. Based on my findings, I have formulated six management recommendations: 1) quantifying PCB detections in LA County; 2) mitigating secondary sources; 3) identifying helpful inclusions for the upcoming EPA Integrated Risk Information System (IRIS) PCB assessment; 4) bridging the data gap for IRIS toxicity values; 5) complementing the data gap associated with weathered Aroclors; and 6) proposing a cleanup grant. By addressing these uncertainties, the general public, scientific community, and regulatory agencies may pursue in minimizing the adverse health risks associated with the lack of inhalation toxicity values and updated regulations

    Alternative methods for regulatory toxicology – a state-of-the-art review

    This state-of-the art review is based on the final report of a project carried out by the European Commission’s Joint Research Centre (JRC) for the European Chemicals Agency (ECHA). The aim of the project was to review the state of the science of non-standard methods that are available for assessing the toxicological and ecotoxicological properties of chemicals. Non-standard methods refer to alternatives to animal experiments, such as in vitro tests and computational models, as well as animal methods that are not covered by current regulatory guidelines. This report therefore reviews the current scientific status of non-standard methods for a range of human health and ecotoxicological endpoints, and provides a commentary on the mechanistic basis and regulatory applicability of these methods. For completeness, and to provide context, currently accepted (standard) methods are also summarised. In particular, the following human health endpoints are covered: a) skin irritation and corrosion; b) serious eye damage and eye irritation; c) skin sensitisation; d) acute systemic toxicity; e) repeat dose toxicity; f) genotoxicity and mutagenicity; g) carcinogenicity; h) reproductive toxicity (including effects on development and fertility); i) endocrine disruption relevant to human health; and j) toxicokinetics. In relation to ecotoxicological endpoints, the report focuses on non-standard methods for acute and chronic fish toxicity. While specific reference is made to the information needs of REACH, the Biocidal Products Regulation and the Classification, Labelling and Packaging Regulation, this review is also expected to be informative in relation to the possible use of alternative and non-standard methods in other sectors, such as cosmetics and plant protection products.JRC.I.5-Systems Toxicolog

    Diesel exhaust: current knowledge of adverse effects and underlying cellular mechanisms

    Diesel engine emissions are among the most prevalent anthropogenic pollutants worldwide, and with the growing popularity of diesel-fueled engines in the private transportation sector, they are becoming increasingly widespread in densely populated urban regions. However, a large number of toxicological studies clearly show that diesel engine emissions profoundly affect human health. Thus the interest in the molecular and cellular mechanisms underlying these effects is large, especially concerning the nature of the components of diesel exhaust responsible for the effects and how they could be eliminated from the exhaust. This review describes the fundamental properties of diesel exhaust as well as the human respiratory tract and concludes that adverse health effects of diesel exhaust not only emerge from its chemical composition, but also from the interplay between its physical properties, the physiological and cellular properties, and function of the human respiratory tract. Furthermore, the primary molecular and cellular mechanisms triggered by diesel exhaust exposure, as well as the fundamentals of the methods for toxicological testing of diesel exhaust toxicity, are described. The key aspects of adverse effects induced by diesel exhaust exposure described herein will be important for regulators to support or ban certain technologies or to legitimate incentives for the development of promising new technologies such as catalytic diesel particle filters
