Investigating the effects of palmitoylation on the dopamine 1 receptor (D1)
The dopamine D1 receptor (D1) is a G protein-coupled receptor (GPCR) which regulates various key brain functions like attention, movement, reward, and memory. Understanding D1 signalling may open the horizon for novel treatments for neurological disorders. Upon agonist activation, the heterotrimeric G proteins Gαs activate adenylyl cyclase to increase cAMP/PKA signalling. D1 also engages β-arrestin proteins leading to β-arrestin dependent signalling. The D1 has two palmitoylation sites on cysteines 347&351 in its C-tail domain. However, the distinct roles and implications of palmitoylation on the D1 signalling, trafficking and β-arrestins recruitment are still largely unexplored. A palmitoylation D1 mutant was generated and luminescent based techniques such as BRET and split-Nanoluc complementation assay were employed, to delineate D1 palmitoylation effects on its pharmacology and signalling. The D1 agonists induced 50% less cAMP production in the mutant compared to wildtype (WT) and WT showed a more efficient dissociation of its Gαs. Moreover, the mutant receptor failed to recruit β-arrestin1&2, induced less ERK1/2 activation and internalises in an agonist-independent process while showing an altered intracellular Golgi trafficking. Also, in β-arrestin 1&2 KO HEK 293 cells similar cAMP production levels were reported for D1 WT and palmitoylation mutant. β-arrestin 1&2 KO blocked agonist-induced WT D1 plasma membrane trafficking, indicating that these β-arrestins are driving the differences between WT and the palmitoylation mutant D1. Taken together, our studies indicate that Gαs is the main transducer for D1 cAMP and ERK1/2 signalling and that palmitoylation is essential for its β-arrestin 1&2 interactions and modulating D1 signalling cascades in a drug-dependant process
Measuring the Health and Development of School-age Zimbabwean Children
Health, growth and development during mid-childhood (from 5 to 14 years) are poorly characterised, and this period has been termed the ‘missing middle’. This thesis describes the piloting and application of the School-Age Health, Activity, Resilience, Anthropometry and Neurocognitive (SAHARAN) toolbox to measure growth, cognitive and physical function amongst the SHINE cohort in rural Zimbabwe. The SHINE cluster-randomised trial tested the effects of a household WASH intervention and/or infant and young child feeding (IYCF) on child stunting and anaemia at age 18 months in rural Zimbabwe. SHINE showed that IYCF modestly increased linear growth and reduced stunting by age 18 months, while WASH had no effects. The SAHARAN toolbox was used to measure 1000 HIV-unexposed children (250 in each intervention arm), and 275 HIV-exposed children within the SHINE cohort to evaluate long-term outcomes. Children were re-enrolled at age seven years to evaluate growth, body composition, cognitive and physical function. Four main findings are presented from the SAHARAN toolbox measurements of this cohort. Firstly, child sex, growth and contemporary environmental conditions are associated with school-age physical and cognitive function at seven years. Secondly, early-life growth and baseline environmental conditions suggest the impact of early-life trajectories on multiple aspects of school-age growth, physical and cognitive function. Thirdly, the long-term impact of HIV-exposure in pregnancy is explored, which indicate reduced cognitive function, cardiovascular fitness and head circumference by age 7 years. Finally, associations with the SHINE trial early life interventions are explored, demonstrating that the SHINE early-life nutrition intervention has minimal impact by 7 years of age, except marginally stronger handgrip strength. The public health implications advocate that child interventions need to be earlier (including antenatal), broader (incorporating nurturing care), deeper (providing transformational WASH) and longer (supporting throughout childhood), as well as targeting particularly vulnerable groups such as children born HIV-free

Deep Learning Techniques for Electroencephalography Analysis
In this thesis we design deep learning techniques for training deep neural networks on electroencephalography (EEG) data and in particular on two problems, namely EEG-based motor imagery decoding and EEG-based affect recognition, addressing challenges associated with them. Regarding the problem of motor imagery (MI) decoding, we first consider the various kinds of domain shifts in the EEG signals, caused by inter-individual differences (e.g. brain anatomy, personality and cognitive profile). These domain shifts render multi-subject training a challenging task and impede robust cross-subject generalization. We build a two-stage model ensemble architecture and propose two objectives to train it, combining the strengths of curriculum learning and collaborative training. Our subject-independent experiments on the large datasets of Physionet and OpenBMI, verify the effectiveness of our approach. Next, we explore the utilization of the spatial covariance of EEG signals through alignment techniques, with the goal of learning domain-invariant representations. We introduce a Riemannian framework that concurrently performs covariance-based signal alignment and data augmentation, while training a convolutional neural network (CNN) on EEG time-series. Experiments on the BCI IV-2a dataset show that our method performs superiorly over traditional alignment, by inducing regularization to the weights of the CNN. We also study the problem of EEG-based affect recognition, inspired by works suggesting that emotions can be expressed in relative terms, i.e. through ordinal comparisons between different affective state levels. We propose treating data samples in a pairwise manner to infer the ordinal relation between their corresponding affective state labels, as an auxiliary training objective. We incorporate our objective in a deep network architecture which we jointly train on the tasks of sample-wise classification and pairwise ordinal ranking. We evaluate our method on the affective datasets of DEAP and SEED and obtain performance improvements over deep networks trained without the additional ranking objective
A Simple and Effective Method of Cross-Lingual Plagiarism Detection
We present a simple cross-lingual plagiarism detection method applicable to a
large number of languages. The presented approach leverages open multilingual
thesauri for candidate retrieval task and pre-trained multilingual BERT-based
language models for detailed analysis. The method does not rely on machine
translation and word sense disambiguation when in use, and therefore is
suitable for a large number of languages, including under-resourced languages.
The effectiveness of the proposed approach is demonstrated for several existing
and new benchmarks, achieving state-of-the-art results for French, Russian, and
Armenian languages

A Tale of Two Trees: A Comparative Study on the Effects of Scale and Biodiversity Efforts in Ghana’s Cocoa and Shea Production Networks
This thesis presents two case studies on Ghana’s cocoa and shea production networks, focusing on the effects of scale on biodiversity sustainability dissemination. Analysis is done through my unique holistic framework for action-based production network analysis, which provides a robust and multiscalar analysis to answer my main research question How does scale affect biodiversity sustainability throughout Ghana’s cocoa and shea production networks? The two case studies focus on Ghana’s cocoa and shea production network coordination and the action taken up by different levels of actors within scale, their considerations of, approaches to, and outcomes of biodiversity sustainability dissemination throughout their Ghanaian cocoa production networks. These case studies focus on the history and context of the cocoa and shea sectors as they function within Ghana’s agricultural industry, the influencers, and barriers to biodiversity sustainability dissemination throughout the studied production networks, and the effects of scale on this sustainability attainment. My research is support through primary data collected in Ghana and secondary data. The two case studies are then cross-analysed to draw out the commonalities in context, issues faced, and effects of scale on the studied sustainability aspects. The findings of this research show that in order to achieve biodiversity sustainability, social sustainability must be incorporated into production network coordination and that the level of actors’ scale and scalar approach to network coordination significantly impact achievement of biodiversity sustainability dissemination. The results of this thesis are novel in the fact that it combines several streams of analytical consideration into a holistic framework and presents clear and applicable results that can significantly impact the approach to sustainability dissemination throughout global production networks in an equitable manner that is fit to the context within which production takes place

Constructing a profile for proactive career self-management in public higher education institutions in Ghana
Text in English with abstracts and keywords in English, isiXhosa, and isiZuluThe principal focus of this research study was to investigate the relationship among psychosocial career pre-occupation, social connectedness, and organisational commitment, and to determine whether an overall proactive career management profile can be constructed to inform career self-management practices in public higher education institutions (HEIs) in Ghana. Again, the research study intended to provide a background for testing hypotheses and theories as well as moderating the effect of socio-demographic characteristics (age, gender, marital status and job level) on the relationship between psychosocial career pre-occupations, social connectedness and organisational commitment.
A cross-sectional quantitative approach was conducted on a purposively selected sample of senior staff comprising academic and administrative staff (n = 288), from a single public higher educational institution in Ghana. The measuring instruments include a career pre-occupations inventory workplace friendship scale and an organisational commitment scale. Descriptive statistics (mean, standard deviation, skewness and kurtosis), bi-variate correlation analysis (Pearson product moment correlation coefficient), and inferential and multivariate statistics (SEM analysis, moderated regression analysis, ANOVAs and independent sample t-test) were used for the study.
Descriptive, bi-variate correlation and inferential statistics revealed that individual psychosocial career pre-occupations, social connectedness and organisational commitment can be used as elements within a proactive career self-management framework within the Ghanaian higher education institutions. The results of the moderated analysis showed that respondents’ gender and job level moderated employees’ level of psychosocial career pre-occupations in predicting organisational commitment. Furthermore, the test for significant mean differences revealed that gender, marital status and job level differed marginally in their psychosocial career pre-occupations, social connectedness and commitment to the organisation.
Theoretically and empirically, the results advanced the career construction theory by empirically validating the core elements of career self-management.
