A Decision Support System for Economic Viability and Environmental Impact Assessment of Vertical Farms
Vertical farming (VF) is the practice of growing crops or animals using the vertical dimension via multi-tier racks or vertically inclined surfaces. In this thesis, I focus on the emerging industry of plant-specific VF. Vertical plant farming (VPF) is a promising and relatively novel practice that can be conducted in buildings with environmental control and artificial lighting. However, the nascent sector has experienced challenges in economic viability, standardisation, and environmental sustainability. Practitioners and academics call for a comprehensive financial analysis of VPF, but efforts are stifled by a lack of valid and available data.
A review of economic estimation and horticultural software identifies a need for a decision support system (DSS) that facilitates risk-empowered business planning for vertical farmers. This thesis proposes an open-source DSS framework to evaluate business sustainability through financial risk and environmental impact assessments. Data from the literature, alongside lessons learned from industry practitioners, would be centralised in the proposed DSS using imprecise data techniques. These techniques have been applied in engineering but are seldom used in financial forecasting. This could benefit complex sectors which only have scarce data to predict business viability.
To begin the execution of the DSS framework, VPF practitioners were interviewed using a mixed-methods approach. Learnings from over 19 shuttered and operational VPF projects provide insights into the barriers inhibiting scalability and identifying risks to form a risk taxonomy. Labour was the most commonly reported top challenge. Therefore, research was conducted to explore lean principles to improve productivity.
A probabilistic model representing a spectrum of variables and their associated uncertainty was built according to the DSS framework to evaluate the financial risk for VF projects. This enabled flexible computation without precise production or financial data to improve economic estimation accuracy. The model assessed two VPF cases (one in the UK and another in Japan), demonstrating the first risk and uncertainty quantification of VPF business models in the literature. The results highlighted measures to improve economic viability and the viability of the UK and Japan case.
The environmental impact assessment model was developed, allowing VPF operators to evaluate their carbon footprint compared to traditional agriculture using life-cycle assessment. I explore strategies for net-zero carbon production through sensitivity analysis. Renewable energies, especially solar, geothermal, and tidal power, show promise for reducing the carbon emissions of indoor VPF. Results show that renewably-powered VPF can reduce carbon emissions compared to field-based agriculture when considering the land-use change.
The drivers for DSS adoption have been researched, showing a pathway of compliance and design thinking to overcome the ‘problem of implementation’ and enable commercialisation. Further work is suggested to standardise VF equipment, collect benchmarking data, and characterise risks. This work will reduce risk and uncertainty and accelerate the sector’s emergence
A direct-laser-written heart-on-a-chip platform for generation and stimulation of engineered heart tissues
In this dissertation, we first develop a versatile microfluidic heart-on-a-chip model to generate 3D-engineered human cardiac microtissues in highly-controlled microenvironments. The platform, which is enabled by direct laser writing (DLW), has tailor-made attachment sites for cardiac microtissues and comes with integrated strain actuators and force sensors. Application of external pressure waves to the platform results in controllable time-dependent forces on the microtissues. Conversely, oscillatory forces generated by the microtissues are transduced into measurable electrical outputs. After characterization of the responsivity of the transducers, we demonstrate the capabilities of this platform by studying the response of cardiac microtissues to prescribed mechanical loading and pacing.
Next, we tune the geometry and mechanical properties of the platform to enable parametric studies on engineered heart tissues. We explore two geometries: a rectangular seeding well with two attachment sites, and a stadium-like seeding well with six attachment sites. The attachment sites are placed symmetrically in the longitudinal direction. The former geometry promotes uniaxial contraction of the tissues; the latter additionally induces diagonal
fiber alignment. We systematically increase the length for both configurations and observe a positive correlation between fiber alignment at the center of the microtissues and tissue length. However, progressive thinning and “necking” is also observed, leading to the failure of longer tissues over time. We use the DLW technique to improve the platform, softening
the mechanical environment and optimizing the attachment sites for generation of stable microtissues at each length and geometry. Furthermore, electrical pacing is incorporated into the platform to evaluate the functional dynamics of stable microtissues over the entire range of physiological heart rates. Here, we typically observe a decrease in active force and contraction duration as a function of frequency.
Lastly, we use a more traditional ?TUG platform to demonstrate the effects of subthreshold electrical pacing on the rhythm of the spontaneously contracting cardiac microtissues. Here, we observe periodic M:N patterns, in which there are ? cycles of stimulation for every ? tissue contractions. Using electric field amplitude, pacing frequency, and homeostatic beating frequencies of the tissues, we provide an empirical map for predicting the emergence of these rhythms
[EN] Currently, regional and local decision-makers lack of tools to achieve greenhouse gases (GHG) emissions inventories with enough rigor, accuracy and completeness in order to prioritize available resources efficiently against climate change. Thus, in this thesis the development of a territorial and sectoral information system (SITE) to monitor GHG emissions as a local and regional climate governance tool is exposed. This system combines the advantages of both, top-down and bottom-up approaches, to achieve an innovative hybrid approach to account and manage efficiently GHG emissions. Furthermore, this thesis defines the methodologies developed, a computer proposal for the back-end of the system and seven implementations as representative case studies at different scales (local and regional level), with the different methodological approaches and applied to different sectors.
Thus, these implementations demonstrate the potential of the system as decision-making tool against climate change at the regional and local level as climate governance tool. The different implementations in representative pilot cases, both at the regional level in the Valencian Community and at the local level in large (Valencia) and medium-sized municipalities (Quart de Poblet and Llíria) demonstrate the potential for territorial and sectoral adaptation of the system developed. The methodologies developed for the specific sectors of road transport, building and forestry, offer quantifications with a spatial resolution with a great capacity to optimize local and regional policies. Therefore, the tool has a great potential for scalability and a great capacity for continuous improvement through the inclusion of new methodological approaches, adapting the methodologies to the availability of data, specific methodologies for key sectors, and updating to the best methodologies available in the scientific community.Lorenzo Sáez, E. Full stack development toward a trapped ion logical qubit
Quantum error correction is a key step toward the construction of a large-scale quantum computer, by preventing small infidelities in quantum gates from accumulating over the course of an algorithm. Detecting and correcting errors is achieved by using multiple physical qubits to form a smaller number of robust logical
qubits. The physical implementation of a logical qubit requires multiple qubits, on which high fidelity gates
can be performed.
The project aims to realize a logical qubit based on ions confined on a microfabricated surface trap. Each
physical qubit will be a microwave dressed state qubit based on 171Yb+ ions. Gates are intended to be realized through RF and microwave radiation in combination with magnetic field gradients. The project vertically integrates software down to hardware compilation layers in order to deliver, in the near future, a fully functional small device demonstrator.
This thesis presents novel results on multiple layers of a full stack quantum computer model. On the hardware level a robust quantum gate is studied and ion displacement over the X-junction geometry is demonstrated.
The experimental organization is optimized through automation and compressed waveform data transmission. A new quantum assembly language purely dedicated to trapped ion quantum computers is introduced. The demonstrator is aimed at testing implementation of quantum error correction codes while preparing for larger
scale iterations.Open Acces
Novel Cardiac Mapping Approaches and Multimodal Techniques to Unravel Multidomain Dynamics of Complex Arrhythmias Towards a Framework for Translational Mechanistic-Based Therapeutic Strategies
[EN] Cardiac arrhythmias are a major problem for health systems in the developed world due to their high incidence and prevalence as the population ages. Atrial fibrillation (AF) and ventricular fibrillation (VF), are amongst the most complex arrhythmias seen in the clinical practice. Clinical consequences of such arrhythmic disturbances include developing complex cardio-embolic events in AF, and dramatic repercussions due to sustained life-threatening fibrillatory processes with subsequent neurological damage under VF, leading to cardiac arrest and sudden cardiac death (SCD). However, despite the technological advances in the last decades, their intrinsic mechanisms are incompletely understood, and, to date, therapeutic strategies lack of sufficient mechanistic basis and have low success rates.
Most of the progress for developing optimal biomarkers and novel therapeutic strategies in this field has come from valuable techniques in the research of arrhythmia mechanisms. Amongst the mechanisms involved in the induction and perpetuation of cardiac arrhythmias such AF, dynamic high-frequency re-entrant and focal sources, in its different modalities, are thought to be the primary sources underlying the arrhythmia. However, little is known about the attractors and spatiotemporal dynamics of such fibrillatory primary sources, specifically dominant rotational or focal sources maintaining the arrhythmia. Therefore, a computational platform for understanding active, passive and structural determinants, and modulators of such dynamics was developed. This allowed stablishing a framework for understanding the complex multidomain dynamics of rotors with enphasis in their deterministic properties to develop mechanistic approaches for diagnostic aid and therapy.
Understanding fibrillatory processes is key to develop physiologically and clinically relevant scores and tools for early diagnostic aid. Specifically, spectral and time-frequency properties of fibrillatory processes have shown to highlight major deterministic behaviour of intrinsic mechanisms underlying the arrhythmias and the impact of such arrhythmic events. Using prior knowledge, signal processing, machine learning techniques and data analytics, we aimed at developing a reliable mechanistic risk-score for comatose survivors of cardiac arrest due to VF.
Cardiac optical mapping and electrophysiological mapping techniques have shown to be unvaluable resources to shape new hypotheses and develop novel mechanistic approaches and therapeutic strategies. This technology has allowed for many years testing new pharmacological or ablative therapeutic strategies, and developing multidomain methods to accurately track arrhymia dynamics identigying dominant sources and attractors. Even though, panoramic mapping is the primary method for simultaneously tracking electrophysiological parameters, its adoption by the multidisciplinary cardiovascular research community is limited mainly due to the cost of the technology. Taking advantage of recent technological advances, we focus on developing and validating low-cost optical mapping systems for panoramic imaging using clinically relevant models for basic research and bioengineering.Calvo Saiz, CJ. Development of a light-sheet fluorescence microscope employing an ALPAO deformable mirror to achieve video-rate remote refocusing and volumetric imaging.
There are numerous situations in microscopy where it is desirable to remotely refocus a microscope employing a high numerical aperture (NA) objective lens. This thesis describes the characterisation,
development and implementation of an Alpao membrane deformable mirror-based system to achieve this goal for a light-sheet fluorescence microscope (LSFM).
The Alpao deformable mirror (DM) DM97-15 used is this work has 97 actuators and was sufficiently
fast to perform refocus sweeps at 25 Hz and faster. However, a known issue with using Alpao
deformable mirrors in open-loop mode is that they exhibit viscoelastic creep and temperature-
dependent variations in the mirror response. The effect of visco-elastic creep was reduced by
ensuring that the mirror profile was on average constant on timescales shorter than the characteristic time of the visco-elastic creep. The thermal effect was managed by ensuring that the electrical power delivered to the actuators was constant prior to optimisation and use. This was achieved by ensuring that the frequency and amplitude of oscillation of the mirror was constant prior to optimisation, so that it reached a thermal steady state, was approximately constant during optimisation and constant during use.
The image-based optimisation procedure employed used an estimate of the Strehl ratio of the optical system calculated from an image of an array of 1 μm diameter holes. The optimisation procedure
included optimising the amount of high-NA defocus and the Zernike modes from Noll indices 4 to 24. The system was tested at 26.3 refocus sweeps per second over a refocus range of -50 to 50 μm with a 40x/0.85 air objective and a 40x/0.80 water immersion objective. The air objective enabled a mean Strehl metric of more than 0.6 over a lateral field of view of 200x200 microns and for a refocus range of 45 microns. The water objective achieved a mean Strehl metric of more than 0.6 over a lateral field of view of 200x200 microns over a larger refocus range of 77 microns.
The DM-based refocusing system was then incorporated into a LSFM setup. The spatial resolution of the system was characterised using fluorescent beads imaged volumetrically at 26.3 volumes per second. The performance of the system was also demonstrated for imaging fluorescence pollen grain samples.Open Acces
Where Should Traffic Sensors Be Placed on Highways?
This paper investigates the practical engineering problem of traffic sensors
placement on stretched highways with ramps. Since it is virtually impossible to
install bulky traffic sensors on each highway segment, it is crucial to find
placements that result in optimized network-wide, traffic observability.
Consequently, this results in accurate traffic density estimates on segments
where sensors are not installed. The substantial contribution of this paper is
the utilization of control-theoretic observability analysis -- jointly with
integer programming -- to determine traffic sensor locations based on the
nonlinear dynamics and parameters of traffic networks. In particular, the
celebrated asymmetric cell transmission model is used to guide the placement
strategy jointly with observability analysis of nonlinear dynamic systems
through Gramians. Thorough numerical case studies are presented to corroborate
the proposed theoretical methods and various computational research questions
are posed and addressed. The presented approach can also be extended to other
models of traffic dynamics

The European Spallation Source neutrino super-beam conceptual design report
Artículo escrito por un elevado número de autores, solo se referencian el que aparece en primer lugar, el nombre del grupo de colaboración, si le hubiere, y los autores pertenecientes a la UAMA design study, named ESS νSB for European Spallation Source neutrino Super Beam, has been carried out during the years 2018–2022 of how the 5 MW proton linear accelerator of the European Spallation Source under construction in Lund, Sweden, can be used to produce the world’s most intense long-baseline neutrino beam. The high beam intensity will allow for measuring the neutrino oscillations near the second oscillation maximum at which the CP violation signal is close to three times higher than at the first maximum, where other experiments measure. This will enable CP violation discovery in the leptonic sector for a wider range of values of the CP violating phase δCP and, in particular, a higher precision measurement of δCP. The present Conceptual Design Report describes the results of the design study of the required upgrade of the ESS linac, of the accumulator ring used to compress the linac pulses from 2.86 ms to 1.2 μs, and of the target station, where the 5 MW proton beam is used to produce the intense neutrino beam. It also presents the design of the near detector, which is used to monitor the neutrino beam as well as to measure neutrino cross sections, and of the large underground far detector located 360 km from ESS, where the magnitude of the oscillation appearance of νe from νμ is measured. The physics performance of the ESS νSB research facility has been evaluated demonstrating that after 10 years of data-taking, leptonic CP violation can be detected with more than 5 standard deviation significance over 70% of the range of values that the CP violation phase angle δCP can take and that δCP can be measured with a standard error less than 8° irrespective of the measured value of δCP. These results demonstrate the uniquely high physics performance of the proposed ESS νSB research facilit
