    This work proposes advanced emulation of the physical layer behavior of NB-PLC channels and the application of a channel emulator for the evaluation of NB-PLC systems. In addition, test procedures and reference channels are proposed to improve efficiency and accuracy in the system evaluation and classification. This work shows that the channel emulator-based solution opens new ways toward flexible, reliable and technology-independent performance assessment of PLC modems

    The development of an on-chip-metering solution

    Includes bibliographical references.Energy Measurements Ltd (EML), a joint venture partnership between Siemens and Spescom, manufacture prepaid electricity utility meters for both the local and international markets. Under the brand name CASHPOWER 2000, EML produces single and polyphase prepayment utility meters. Currently, these meters currently utilise a separate module for the measuring of electrical energy. In order to reduce component costs, EML proposed the energy measurement be conducted by the onboard Microcontroller Unit (MCU), a term known as On-Chip-Metering (OCM). It is envisioned that this would quickly translate in an increase in revenue. However, a major concern regarding this has been the increase in the required processor overhead. The CASHPOWER 2000 embedded MCU would be required to conduct all the present metering functionality in addition to the energy measurement. This, together with the cost analysis and compliance with the stipulated IEC1036 regulations, constitute the key criteria in determining the projects viability. This dissertation represents the investigative and development stages of a prototype algorithm and accompanying peripheral hardware as a possible solution for OCM. As part of the preliminary research, several examples of digital power and energy-measurement techniques were investigated. A comparative analysis of these was performed to facilitate the development of a unique solution based on the research conducted. This completed, a prototype was developed and preliminary testing was conducted to determine its compliance with the stipulated regulations for a class 2 meter, as per IEC1O36 specifications

    Dynamic properties of noise and Her6 levels are optimized by miR-9, allowing the decoding of the Her6 oscillator

    This work was supported by a Wellcome Trust Senior Research Fellowship to NP (106185/Z/14/Z).Noise is prevalent in biology and has been widely quantified using snapshot measurements. This static view obscures our understanding of dynamic noise properties and how these affect gene expression and cell state transitions. Using a CRISPR/Cas9 Zebrafish her6::Venus reporter combined with mathematical and in vivo experimentation, we explore how noise affects the protein dynamics of Her6, a basic helix‐loop‐helix transcriptional repressor. During neurogenesis, Her6 expression transitions from fluctuating to oscillatory at single‐cell level. We identify that absence of miR‐9 input generates high‐frequency noise in Her6 traces, inhibits the transition to oscillatory protein expression and prevents the downregulation of Her6. Together, these impair the upregulation of downstream targets and cells accumulate in a normally transitory state where progenitor and early differentiation markers are co‐expressed. Computational modelling and double smFISH of her6 and the early neurogenesis marker, elavl3, suggest that the change in Her6 dynamics precedes the downregulation in Her6 levels. This sheds light onto the order of events at the moment of cell state transition and how this is influenced by the dynamic properties of noise. Our results suggest that Her/Hes oscillations, facilitated by dynamic noise optimization by miR‐9, endow progenitor cells with the ability to make a cell state transition.Publisher PDFPeer reviewe

    Inferring kinetic parameters of oscillatory gene regulation from single cell time-series data

M.R.’s work was supported by a Wellcome Trust Investigator Award (204832/B/16/Z).Gene expression dynamics, such as stochastic oscillations and aperiodic fluctuations, have been associated with cell fate changes in multiple contexts, including development and cancer. Single cell live imaging of protein expression with endogenous reporters is widely used to observe such gene expression dynamics. However, the experimental investigation of regulatory mechanisms underlying the observed dynamics is challenging, since these mechanisms include complex interactions of multiple processes, including transcription, translation and protein degradation. Here, we present a Bayesian method to infer kinetic parameters of oscillatory gene expression regulation using an auto-negative feedback motif with delay. Specifically, we use a delay-adapted nonlinear Kalman filter within a Metropolis-adjusted Langevin algorithm to identify posterior probability distributions. Our method can be applied to time-series data on gene expression from single cells and is able to infer multiple parameters simultaneously. We apply it to published data on murine neural progenitor cells and show that it outperforms alternative methods. We further analyse how parameter uncertainty depends on the duration and time resolution of an imaging experiment, to make experimental design recommendations. This work demonstrates the utility of parameter inference on time course data from single cells and enables new studies on cell fate changes and population heterogeneity.Publisher PDFPeer reviewe

    Study of spread spectrum multiple access systems for satellite communications with overlay on current services

    The feasibility of using spread spectrum techniques to provide a low-cost multiple access system for a very large number of low data terminals was investigated. Two applications of spread spectrum technology to very small aperture terminal (VSAT) satellite communication networks are presented. Two spread spectrum multiple access systems which use a form of noncoherent M-ary FSK (MFSK) as the primary modulation are described and the throughput analyzed. The analysis considers such factors as satellite power constraints and adjacent satellite interference. Also considered is the effect of on-board processing on the multiple access efficiency and the feasibility of overlaying low data rate spread spectrum signals on existing satellite traffic as a form of frequency reuse is investigated. The use of chirp is examined for spread spectrum communications. In a chirp communication system, each data bit is converted into one or more up or down sweeps of frequency, which spread the RF energy across a broad range of frequencies. Several different forms of chirp communication systems are considered, and a multiple-chirp coded system is proposed for overlay service. The mutual interference problem is examined in detail and a performance analysis undertaken for the case of a chirp data channel overlaid on a video channel
