    Neural Processes Supporting Young and Older Adults' Emotional Memories

    Young and older adults are more likely to remember emotional information than neutral information. The present functional magnetic resonance imaging study examined the neural processes supporting young (ages 18-35) and older (ages 62-79) adults' successful encoding of positive, negative, and neutral objects (e.g., a sundae, a grenade, a canoe). The results revealed general preservation of the emotional memory network across the age groups. Both groups recruited the amygdala and the orbito-frontal cortex during the successful encoding of positive and negative information. Both ages also showed valence-specific recruitment: right fusiform activity was greatest during the successful encoding of negative information, whereas left prefrontal and temporal activity was greatest during the successful encoding of positive information. These valence-specific processes are consistent with behavioral evidence that negative information is processed with perceptual detail, whereas positive information is processed at a conceptual or schematic level. The only age differences in emotional memory emerged during the successful encoding of positive items: Older adults showed more activity in the medial prefrontal cortex and along the cingulate gyrus than young adults. Because these regions often are associated with self-referential processing, these results suggest that older adults' mnemonic boost for positive information may stem from an increased tendency to process this information in relation to themselves.Psycholog

    Mental sleep activity and disturbing dreams in the lifespan

    Sleep significantly changes across the lifespan, and several studies underline its crucial role in cognitive functioning. Similarly, mental activity during sleep tends to covary with age. This review aims to analyze the characteristics of dreaming and disturbing dreams at dierent age brackets. On the one hand, dreams may be considered an expression of brain maturation and cognitive development, showing relations with memory and visuo-spatial abilities. Some investigations reveal that specific electrophysiological patterns, such as frontal theta oscillations, underlie dreams during sleep, as well as episodic memories in the waking state, both in young and older adults. On the other hand, considering the role of dreaming in emotional processing and regulation, the available literature suggests that mental sleep activity could have a beneficial role when stressful events occur at dierent age ranges. We highlight that nightmares and bad dreams might represent an attempt to cope the adverse events, and the degrees of cognitive-brain maturation could impact on these mechanisms across the lifespan. Future investigations are necessary to clarify these relations. Clinical protocols could be designed to improve cognitive functioning and emotional regulation by modifying the dream contents or the ability to recall/non-recall them

    The Role of Autobiographical Memory Recall in Reappraisal Efficacy and Effort Across Age

    Socioemotional theories posit that the experience of overcoming unique life challenges over a lifetime enhances self-efficacy and emotional resilience among older adults. Older adults demonstrate greater emotional well-being and motivation to regulate emotions than younger adults, but specific regulatory mechanisms supporting late-life emotional resilience remain unclear. Cognitive reappraisal is an effective but cognitively demanding emotion regulation strategy and shows mixed efficacy in later-life. While a growing repertoire of autobiographical memories may be a resource with age, the role of autobiographical recall in momentary reappraisal has never been tested empirically. In this online study, older and younger adults were trained to reappraise the meaning of negative images as more positive by associating them either to any relevant autobiographical memory from the past (n=153), a specific cued autobiographical memory (reappraisal, n=118), or without autobiographical reference (n=156). Results revealed all strategies to be effective in regulating negative image intensity across age. While older adults outperformed younger adults in the non-AM condition, the opposite was observed for uncued AM reappraisals, and no age differences were found for cued AM reappraisals. Non-AM reappraisal was easiest to employ for all participants, and older adults reported all reappraisal strategies as easier than younger adults. While older adults found their AMs more helpful and more similar to reappraised images than younger adults, AM- image similarity was surprisingly associated with lower reappraisal efficacy. Findings suggests that AM reappraisal benefits are mixed for older adults, likely due to efficacy but higher associated cognitive burden of AM recall. We posit that older adults may sacrifice immediate hedonic relief, to engage with challenging but helpful emotion regulation strategies.     Functional Magnetic Resonance Imaging of Semantic Memory as a Presymptomatic Biomarker of Alzheimer’s Disease Risk

    Extensive research efforts have been directed toward strategies for predicting risk of developing Alzheimer\u27s disease (AD) prior to the appearance of observable symptoms. Existing approaches for early detection of AD vary in terms of their efficacy, invasiveness, and ease of implementation. Several non-invasive magnetic resonance imaging strategies have been developed for predicting decline in cognitively healthy older adults. This review will survey a number of studies, beginning with the development of a famous name discrimination task used to identify neural regions that participate in semantic memory retrieval and to test predictions of several key theories of the role of the hippocampus in memory. This task has revealed medial temporal and neocortical contributions to recent and remote memory retrieval, and it has been used to demonstrate compensatory neural recruitment in older adults, apolipoprotein E ε4 carriers, and amnestic mild cognitive impairment patients. Recently, we have also found that the famous name discrimination task provides predictive value for forecasting episodic memory decline among asymptomatic older adults. Other studies investigating the predictive value of semantic memory tasks will also be presented. We suggest several advantages associated with the use of semantic processing tasks, particularly those based on person identification, in comparison to episodic memory tasks to study AD risk. Future directions for research and potential clinical uses of semantic memory paradigms are also discussed.     Age and the Neural Network of Personal Familiarity

    BACKGROUND: Accessing information that defines personally familiar context in real-world situations is essential for the social interactions and the independent functioning of an individual. Personal familiarity is associated with the availability of semantic and episodic information as well as the emotional meaningfulness surrounding a stimulus. These features are known to be associated with neural activity in distinct brain regions across different stimulus conditions (e.g., when perceiving faces, voices, places, objects), which may reflect a shared neural basis. Although perceiving context-rich personal familiarity may appear unchanged in aging on the behavioral level, it has not yet been studied whether this can be supported by neuroimaging data. METHODOLOGY/PRINCIPAL FINDINGS: We used functional magnetic resonance imaging to investigate the neural network associated with personal familiarity during the perception of personally familiar faces and places. Twelve young and twelve elderly cognitively healthy subjects participated in the study. Both age groups showed a similar activation pattern underlying personal familiarity, predominantly in anterior cingulate and posterior cingulate cortices, irrespective of the stimulus type. The young subjects, but not the elderly subjects demonstrated an additional anterior cingulate deactivation when perceiving unfamiliar stimuli. CONCLUSIONS/SIGNIFICANCE: Although we found evidence for an age-dependent reduction in frontal cortical deactivation, our data show that there is a stimulus-independent neural network associated with personal familiarity of faces and places, which is less susceptible to aging-related changes

    Functional and neural mechanisms of human fear conditioning: studies in healthy and brain-damaged individuals

    Fear conditioning represents the learning process by which a stimulus, after repeated pairing with an aversive event, comes to evoke fear and becomes intrinsically aversive. This learning is essential to organisms throughout the animal kingdom and represents one the most successful laboratory paradigm to reveal the psychological processes that govern the expression of emotional memory and explore its neurobiological underpinnings. Although a large amount of research has been conducted on the behavioural or neural correlates of fear conditioning, some key questions remain unanswered. Accordingly, this thesis aims to respond to some unsolved theoretic and methodological issues, thus furthering our understanding of the neurofunctional basis of human fear conditioning both in healthy and brain-damaged individuals. Specifically, in this thesis, behavioural, psychophysiological, lesion and non-invasive brain stimulation studies were reported. Study 1 examined the influence of normal aging on context-dependent recall of extinction of fear conditioned stimulus. Study 2 aimed to determine the causal role of the ventromedial PFC (vmPFC) in the acquisition of fear conditioning by systematically test the effect of bilateral vmPFC brain-lesion. Study 3 aimed to interfere with the reconsolidation process of fear memory by the means of non-invasive brain stimulation (i.e. TMS) disrupting PFC neural activity. Finally, Study 4 aimed to investigate whether the parasympathetic – vagal – modulation of heart rate might reflect the anticipation of fearful, as compared to neutral, events during classical fear conditioning paradigm. Evidence reported in this PhD thesis might therefore provide key insights and deeper understanding of critical issues concerning the neurofunctional mechanisms underlying the acquisition, the extinction and the reconsolidation of fear memories in humans

    Sleep quality influences subsequent motor skill acquisition

    While the influence of sleep on motor memory consolidation has been extensively investigated, its relation to initial skill acquisition is less well understood. The purpose of the present study was to investigate the influence of sleep quality and quantity on subsequent motor skill acquisition in young adults without sleep disorders. Fifty-five healthy adults (mean age = 23.8 years; 34 women) wore actigraph wristbands for 4 nights, which provided data on sleep patterns before the experiment, and then returned to the laboratory to engage in a motor sequence learning task (explicit 5-item finger sequence tapping task). Indicators of sleep quality and quantity were then regressed on a measure of motor skill acquisition (Gains Within Training, GWT). Wake After Sleep Onset (WASO; i.e., the total amount of time the participants spent awake after falling asleep) was significantly and negatively related to GWT. This effect was not because of general arousal level, which was measured immediately before the motor task. Conversely, there was no relationship between GWT and sleep duration or self-reported sleep quality. These results indicate that sleep quality, as assessed by WASO and objectively measured with actigraphy before the motor task, significantly impacts motor skill acquisition in young healthy adults without sleep disorders.     Age-Based Positivity Effects in Imagining and Recalling Future Positive and Negative Autobiographical Events

    Thoughts about the future reflect personal goals, and projections into the future enrich our emotional life. Researchers have taken an interest in determining whether the tendency to remember more positive than negative emotional events observed when recalling past events also appears when remembering imagined future events. The objective of this study was to examine the age-based positivity effect of recall for future positive and negative autobiographical events in young and older adults. Representative future events were first established to develop the cues used to prompt personal future events. In the production task, the participants were presented with eight positive and eight negative random future events of young or older adults as a model and the corresponding cues to generate their own positive and negative future autobiographical events. In the recall task, the participants recovered as many experiences as they could of the model and the positive and negative events produced by themselves. The participants correctly recalled more positive than negative events and committed more errors for negative than positive events, showing a clear tendency in both young and older adults to recall future imagined events as positive. Regarding age, the young adults recalled more events than the older participants whilst the older participants in particular showed better recall of their own imagined future events than the model's events, and committed more errors when recalling the model's events than their own imagined events. Regarding the positivity effect in incorrect recall, more than half of the errors were valence changes, most of these being from negative to positive events, and these valence changes were more pronounced in the older than in the younger adults. In general, there were fewer differences between young and older adults in the recall of positive events in comparison with negative events.
