    Envelope glycoproteins of vesiculoviruses: characteristics of antibody interactions and immunogenicity

    Vesicular stomatitis virus Indiana strain is the prototype envelope glycoprotein for the genus Vesiculovirus. While the wild-type virus, VSVind, has developed into a potent and versatile oncolytic virotherapy and vaccine vector delivery platform, the G protein (VSVind.G) is ubiquitously used to pseudotype lentiviral vectors (LVs) for experimental and clinical applications. Recently, G proteins derived from other vesiculoviruses (VesG), for example, Cocal, Piry, and Chandipura viruses have been proposed as alternative LV envelopes with possible advantages compared to VSVind.G. However, vesiculovirus research has not developed extensively, and there is a gap in knowledge regarding the antigenic and immunogenic characteristic of VesG. In this work, I investigated two anti-VSVind.G monoclonal antibodies for their ability to cross-react with other VesG, identified the epitopes they recognise, and explored the mechanisms behind their neutralisation activity. Furthermore, these G proteins were characterised for their sensitivity to inactivation by fresh mammalian sera. Using some mix-and-match constructs, I identified that the hypervariable PH domain of VSVind.G confers sensitivity to otherwise serum resistant Cocal G. I further examined VesG regarding their immunogenicity, explored the humoral immune response triggered by systemic administration of LVs and investigated the inhibitory effects of the induced anti-G neutralising response on subsequent LV administrations. However, this could be alleviated using a heterogeneous panel of envelopes sequentially. Taken together, this work will broaden the use of VesG pseudotyped lentiviral vectors in clinical gene therapy by providing the proof-of-concept to circumvent anti-envelope immunity where repeated systemic vector administration is necessary and the opportunity to modify the VesG and improve G protein-containing advanced therapy medicinal products and vaccine vectors

    Oncolytic Virus Immunotherapy

    Dear Readers, Oncolytic Viruses (OV) are self-propagating agents that can selectively induce the lysis of cancer cells while sparing normal tissues. OV-mediated cancer cell death is often immunogenic and triggers robust anticancer immune responses and immunoconversion of tumor microenvironments. This makes oncolytic virotherapy a promising new form of immunotherapy and OVs ideal candidates for combination therapy with other anticancer agents, including other immunotherapeutics. There are more than 40 OVs from nine different families in clinical development and many more at the preclinical stage. Each OV has its own unique characteristics, its pros and cons. Although herpes simplex virus is currently the lead clinical agent, a real champion among the OVs has not yet emerged, justifying the continuous development and optimization of these agents. This book, “Oncolytic Virus Immunotherapy”, summarizes the state-of-the-art and gives a comprehensive overview of the OV arena with a particular focus on new trends, directions, challenges, and opportunities

    Immune modulation in gene therapy studies

    Summary Host immune responses play a major role in clearance of viral infections from the body, and may limit long-term expression and clinical efficacy of viral vectors. Methods to prevent these immune responses may also increase the risk for infections, recombination with wild type virus and affect biodistribution, persistence, shedding and transmission. The study described in this report was initiated to assess possible environmental risks associated with the use of immune modulation in combination with gene therapy and set up as a literature study, by performing PubMed searches for certain keywords, by interviewing experts and by attending selected meetings. Lack of availability of clinical data combining gene therapy and immune modulation and limited animal data warranted additional exploration of relevant non-gene therapy studies from closely related fields such as stem cell and organ transplantation, and vaccination studies with live attenuated vaccines. ...... Finally, we propose the use of a checklist to assess current environmental risks in the use of immune modulation during gene therapy. This report is expected to provide guidance to risk assessors and regulatory officers as well as to applicants for a gene therapy licence

    Viral infections in 47 CVID patients in allergy and immunology department of Rasool E Akram hospital in Tehran

    Background: CVID is a heterogeneous primary immune deficiency with infectious, autoimmune and autoinflamatory features. It is most common symptomatic PID in Iran, with prevalence of 1 in 25000 to 50000 people. CVID has been divided into some phenotypes to produce more homogenized subpopulations. CVID is not a pure Ab deficiency .and because of both abnormalities in Tcell and innate immunity in combination with B cell dysfunction these patients are predisposed to viral and opportunistic infections. Method: prevalence of viral infections is reported in 47 CVID patients registered in Rasool E Akram hospital in Tehran. Patients have been diagnosed as CVID with the PAGID-ESID diagnostic criteria in our department or referred from other clinics for follow up and treatment. Diagnosis of viral germs has been made by clinical signs, pathological significances and in some cases by PCR. Cases: 9 patients (19%) had problems with viral infections. Infections occurred befor diagnosis of CVID in some cases or after that. Four patients (8.5 %) had problems with wart. Sever mucocutaneus HSV infection has occurred in 3 (6 %), recurrent zona in one (2 %) and CMV infection as colitis or pneumonitis in 3(6 %) patients. Sever progressive lethal CNS infection with JC virus occurred in one patient. Conclusion: evidences show that CVID is not a pure B cell defect, and we should be aware of opportunistic and viral infections that in some cases may be fatal

    This work was supported by the National Institute of General Medical Sciences [GM131919].In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide.     Drug development progress in duchenne muscular dystrophy

    Duchenne muscular dystrophy (DMD) is a severe, progressive, and incurable X-linked disorder caused by mutations in the dystrophin gene. Patients with DMD have an absence of functional dystrophin protein, which results in chronic damage of muscle fibers during contraction, thus leading to deterioration of muscle quality and loss of muscle mass over time. Although there is currently no cure for DMD, improvements in treatment care and management could delay disease progression and improve quality of life, thereby prolonging life expectancy for these patients. Furthermore, active research efforts are ongoing to develop therapeutic strategies that target dystrophin deficiency, such as gene replacement therapies, exon skipping, and readthrough therapy, as well as strategies that target secondary pathology of DMD, such as novel anti-inflammatory compounds, myostatin inhibitors, and cardioprotective compounds. Furthermore, longitudinal modeling approaches have been used to characterize the progression of MRI and functional endpoints for predictive purposes to inform Go/No Go decisions in drug development. This review showcases approved drugs or drug candidates along their development paths and also provides information on primary endpoints and enrollment size of Ph2/3 and Ph3 trials in the DMD space
