    Type 2 Diabetes Mellitus and its comorbidity, Alzheimer’s disease: Identifying critical microRNA using machine learning

    MicroRNAs (miRNAs) are critical regulators of gene expression in healthy and diseased states, and numerous studies have established their tremendous potential as a tool for improving the diagnosis of Type 2 Diabetes Mellitus (T2D) and its comorbidities. In this regard, we computationally identify novel top-ranked hub miRNAs that might be involved in T2D. We accomplish this via two strategies: 1) by ranking miRNAs based on the number of T2D differentially expressed genes (DEGs) they target, and 2) using only the common DEGs between T2D and its comorbidity, Alzheimer’s disease (AD) to predict and rank miRNA. Then classifier models are built using the DEGs targeted by each miRNA as features. Here, we show the T2D DEGs targeted by hsa-mir-1-3p, hsa-mir-16-5p, hsa-mir-124-3p, hsa-mir-34a-5p, hsa-let-7b-5p, hsa-mir-155-5p, hsa-mir-107, hsa-mir-27a-3p, hsa-mir-129-2-3p, and hsa-mir-146a-5p are capable of distinguishing T2D samples from the controls, which serves as a measure of confidence in the miRNAs’ potential role in T2D progression. Moreover, for the second strategy, we show other critical miRNAs can be made apparent through the disease’s comorbidities, and in this case, overall, the hsa-mir-103a-3p models work well for all the datasets, especially in T2D, while the hsa-mir-124-3p models achieved the best scores for the AD datasets. To the best of our knowledge, this is the first study that used predicted miRNAs to determine the features that can separate the diseased samples (T2D or AD) from the normal ones, instead of using conventional non-biology-based feature selection methods

    Perceptions of surveillance: exploring feelings held by Black community leaders in Boston toward camera enforcement of roadway infractions

    Roadway camera enforcement programs have been found to effectively reduce vehicle travel speeds, as well as decrease the number and severity of collisions. Despite a wealth of evaluative research confirming this enforcement approach's aptitude at promoting safer roadway behavior, fewer than 50 % of US states currently host camera-based programs. Public opposition is frequently cited as the cause for the slow proliferation of this enforcement strategy. However, with public demand for police reform having an increasing presence on the national political stage, how might feelings toward camera technology currently stand among groups most marginalized by existing enforcement systems, and how might those feelings vary by type of enforcement application? Through a series of focus groups, this work centers Black voices on matters of surveillance and roadway enforcement by discussing sentiment toward camera programs with Black community leaders. This discussion is contextually situated in Boston, Massachusetts, where legislation that would allow for camera enforcement of roadway infractions is actively being deliberated in the State Senate. Findings culminate in a list of right-sizing and procedural recommendations for policy makers hoping to gain support for camera enforcement, improve roadway safety, and advance racial equity in our systems of policing and governance

    Klarnetin staccato tekniğini güçlendirme aşamaları eser çalışmalarıyla uygulanmıştır. Staccato geçişlerini hızlandıracak ritim ve nüans çalışmalarına yer verilmiştir. Çalışmanın en önemli amacı sadece staccato çalışması değil parmak-dilin eş zamanlı uyumunun hassasiyeti üzerinde de durulmasıdır. Staccato çalışmalarını daha verimli hale getirmek için eser çalışmasının içinde etüt çalışmasına da yer verilmiştir. Çalışmaların üzerinde titizlikle durulması staccato çalışmasının ilham verici etkisi ile müzikal kimliğe yeni bir boyut kazandırmıştır. Sekiz özgün eser çalışmasının her aşaması anlatılmıştır. Her aşamanın bir sonraki performans ve tekniği güçlendirmesi esas alınmıştır. Bu çalışmada staccato tekniğinin hangi alanlarda kullanıldığı, nasıl sonuçlar elde edildiği bilgisine yer verilmiştir. Notaların parmak ve dil uyumu ile nasıl şekilleneceği ve nasıl bir çalışma disiplini içinde gerçekleşeceği planlanmıştır. Kamış-nota-diyafram-parmak-dil-nüans ve disiplin kavramlarının staccato tekniğinde ayrılmaz bir bütün olduğu saptanmıştır. Araştırmada literatür taraması yapılarak staccato ile ilgili çalışmalar taranmıştır. Tarama sonucunda klarnet tekniğin de kullanılan staccato eser çalışmasının az olduğu tespit edilmiştir. Metot taramasında da etüt çalışmasının daha çok olduğu saptanmıştır. Böylelikle klarnetin staccato tekniğini hızlandırma ve güçlendirme çalışmaları sunulmuştur. Staccato etüt çalışmaları yapılırken, araya eser çalışmasının girmesi beyni rahatlattığı ve istekliliği daha arttırdığı gözlemlenmiştir. Staccato çalışmasını yaparken doğru bir kamış seçimi üzerinde de durulmuştur. Staccato tekniğini doğru çalışmak için doğru bir kamışın dil hızını arttırdığı saptanmıştır. Doğru bir kamış seçimi kamıştan rahat ses çıkmasına bağlıdır. Kamış, dil atma gücünü vermiyorsa daha doğru bir kamış seçiminin yapılması gerekliliği vurgulanmıştır. Staccato çalışmalarında baştan sona bir eseri yorumlamak zor olabilir. Bu açıdan çalışma, verilen müzikal nüanslara uymanın, dil atış performansını rahatlattığını ortaya koymuştur. Gelecek nesillere edinilen bilgi ve birikimlerin aktarılması ve geliştirici olması teşvik edilmiştir. Çıkacak eserlerin nasıl çözüleceği, staccato tekniğinin nasıl üstesinden gelinebileceği anlatılmıştır. Staccato tekniğinin daha kısa sürede çözüme kavuşturulması amaç edinilmiştir. Parmakların yerlerini öğrettiğimiz kadar belleğimize de çalışmaların kaydedilmesi önemlidir. Gösterilen azmin ve sabrın sonucu olarak ortaya çıkan yapıt başarıyı daha da yukarı seviyelere çıkaracaktır

    Discovering the hidden structure of financial markets through bayesian modelling

    Understanding what is driving the price of a financial asset is a question that is currently mostly unanswered. In this work we go beyond the classic one step ahead prediction and instead construct models that create new information on the behaviour of these time series. Our aim is to get a better understanding of the hidden structures that drive the moves of each financial time series and thus the market as a whole. We propose a tool to decompose multiple time series into economically-meaningful variables to explain the endogenous and exogenous factors driving their underlying variability. The methodology we introduce goes beyond the direct model forecast. Indeed, since our model continuously adapts its variables and coefficients, we can study the time series of coefficients and selected variables. We also present a model to construct the causal graph of relations between these time series and include them in the exogenous factors. Hence, we obtain a model able to explain what is driving the move of both each specific time series and the market as a whole. In addition, the obtained graph of the time series provides new information on the underlying risk structure of this environment. With this deeper understanding of the hidden structure we propose novel ways to detect and forecast risks in the market. We investigate our results with inferences up to one month into the future using stocks, FX futures and ETF futures, demonstrating its superior performance according to accuracy of large moves, longer-term prediction and consistency over time. We also go in more details on the economic interpretation of the new variables and discuss the created graph structure of the market.Open Acces

    Cultivating Agrobiodiversity in the U.S.: Barriers and Bridges at Multiple Scales

    The diversity of crops grown in the United States (U.S.) is declining, causing agricultural landscapes to become more and more simplified. This trend is concerning for the loss of important plant, insect, and animal species, as well as the pollution and degradation of our environment. Through three separate but related studies, this dissertation addresses the need to increase the diversity of these agricultural landscapes in the U.S., particularly through diversifying the type and number of crops grown. The first study uses multiple, openly accessible datasets related to agricultural land use and policies to document and visualize change over recent decades. Through this, I show that U.S. agriculture has gradually become more specialized in the crops grown, crop production is heavily concentrated in certain areas, and crop diversity is continuing to decline. Meanwhile, federal agricultural policy, while having become more influential over how U.S. agriculture operates, incentivizes this specialization. The second study uses nonlinear statistical modeling to identify and compare social, political, and ecological factors that best predict crop diversity across nine regions in the U.S. Factors of climate, prior land use, and farm inputs best predict diversity across regions, but regions show key differences in how factors are important, indicating that patterns at the regional scale constrain and enable further diversification. Finally, the third study relied on interviews with farmers and key informants in southern Idaho’s Magic Valley – a cluster of eight counties that is known to be agriculturally diverse. Interviews gauge what farmers are currently doing to manage crop diversity (the present) and how they imagine alternative landscapes (the imaginary). We found that farmers in the Magic Valley manage current diversity mainly through cover cropping and diverse crop rotations, but daily struggles and political barriers make experimenting with and imagining alternative landscapes difficult and unlikely to occur. Together, these three studies provide an integrated view of how and why U.S. agriculture landscapes simplify or diversify, as well as the barriers and bridges such pathways of diversification
