    Nuclear RNA Surveillance in \u3cem\u3eSaccharomyces cerevisiae\u3c/em\u3e: Trf4p-dependent Polyadenylation of Nascent Hypomethylated tRNA and an Aberrant Form of 5S rRNA

    1-Methyladenosine modification at position 58 of tRNA is catalyzed by a two-subunit methyltransferase composed of Trm6p and Trm61p in Saccharomyces cerevisiae. Initiator tRNA (tRNAiMet) lacking m1A58 (hypomethylated) is rendered unstable through the cooperative function of the poly(A) polymerases, Trf4p/Trf5p, and the nuclear exosome. We provide evidence that a catalytically active Trf4p poly(A) polymerase is required for polyadenylation of hypomethylated tRNAiMet in vivo. DNA sequence analysis of tRNAiMet cDNAs and Northern hybridizations of poly(A)+ RNA provide evidence that nascent pre-tRNAiMet transcripts are targeted for polyadenylation and degradation. We determined that a mutant U6 snRNA and an aberrant form of 5S rRNA are stabilized in the absence of Trf4p, supporting that Trf4p facilitated RNA surveillance is a global process that stretches beyond hypomethylated tRNAiMet. We conclude that an array of RNA polymerase III transcripts are targeted for Trf4p/ Trf5p-dependent polyadenylation and turnover to eliminate mutant and variant forms of normally stable RNAs

    USING OTOLITH MICROCHEMISTRY TO CLASSIFY YELLOW PERCH AS STOCKED OR NATURALLY PRODUCED

    Fisheries managers routinely use stocking to supplement fish populations (Schramm and Piper 1995, Fisher 1996). Stocking eyed-eggs offers substantial cost savings compared to stocking fry and fingerlings (PFBC 2011); however, traditional stocking evaluation using oxytetracycline (OTC) marking of otoliths is ineffective for eyed-eggs of some species (e.g., yellow perch, [Perca fla- vescens]). Thus, there is a need for additional approaches to be able to classify fish as stocked or naturally produced. Fish otoliths are paired calcified structures in the inner ear that permanently deposit trace elements in proportion to water column concentrations (Campana 1999, Campana et al. 2000). Coupled with otolith growth increments (i.e., annuli), elemental accumulation permits retrospective evaluation of environmental history (e.g., natal origins, movement) if water chemistry is spatially heterogeneous and temporally constant (Elsdon et al. 2008). Otolith microchemistry can be used to evaluate stocking contributions (Pracheil et al. 2014) and in the context of eyed-egg stockings, may be useful for classifying fish as stocked or naturally produced. Yellow perch is a popular sport fish species in South Dakota (Gigliotti 2007) and is routinely stocked by fisheries managers to supplement weak year classes (Schoene- beck et al. 2010). The South Dakota Department of Game, Fish and Parks (SDGFP) propagates yellow perch for stocking (e.g., eyed-eggs, fry, fingerlings) and also stocks adult perch through trap and transfer operations (Lott 1991, Fisher 1996). However, the contributions of yellow perch stockings in South Dakota are largely unknown because it is difficult to differentiate stocked fish from resident individuals (Brown and St. Sauver 2002).     Ludwig Meidner (1884-1966), who belonged to the mystical wing of German Expressionism, developed his existing interest in Blake when he was forced to leave Germany in 1939. It is during his exile years that Meidner’s new style matured, and this was partly due to his new appreciation of Blake. The article examines the British context of Meidner’s engagement with Blake and outlines how he understood Blake in a cultural setting dominated by Neo-Romanticism as well as the existential fear of exile experience

    Identification of decomposition volatile organic compounds from surface deposited and submerged porcine remains

    Cadaver dogs are routinely used internationally by police and civilian search organisations to locate human remains on land and in water, yet little is currently known about the volatile organic compounds (VOCs) that are released by a cadaver underwater; how this compares to those given off by a cadaver deposited on land; and ultimately, how this affects the detection of drowned victims by dogs. The aim of this study was to identify the VOCs released by whole porcine (Sus scrofa domesticus) cadavers deposited on the surface and submerged in water using solid phase microextraction gas chromatography mass spectrometry (SPME GC–MS) to ascertain if there are notable differences in decomposition odour depending on the deposition location. For the first time in the UK, the volatile organic compounds (VOCs) from the headspace of decomposing porcine cadavers deposited in both terrestrial and water environments have been detected and identified using SPME-GCMS, including thirteen new VOCs not previously detected from porcine cadavers. Distinct differences were found between the VOCs emitted by porcine cadavers in terrestrial and water environments. In total, seventy-four VOCs were identified from a variety of different chemical classes; carboxylic acids, alcohols, aromatics, aldehydes, ketones, hydrocarbons, esters, ethers, nitrogen compounds and sulphur compounds. Only forty-one VOCs were detected in the headspace of the submerged pigs with seventy detected in the headspace of the surface-deposited pigs. These deposition-dependent differences have important implications for the training of cadaver dogs in the UK. If dog training does not account for these depositional differences, there is potential for human remains to be missed. Whilst the specific odours that elicit a trained response from cadaver dogs remain unknown, this research means that recommendations can be made for the training of cadaver dogs to incorporate different depositions, to account for odour differences and mitigate the possibility of missed human remains operationally

    In his article Arabic Music and Burroughs\u27s The Ticket That Exploded David Holzer discusses how the experience of hearing Arabic music in Tangier and being exposed to the healing music of the Master Musicians of Joujouka, a remote village in the foothills of the Ahl Srif mountain range in Northern Morocco, significantly influenced both the writing of William Burroughs and his multi-media experiments. This essay considers what Arabic music and specifically that of Joujouka meant to Burroughs, with particular reference to The Ticket That Exploded (1962). Drawing on The Ticket, Burroughs’s letters, critical studies and biographical material, it demonstrates that his understanding of what the music was and did was fundamental to his creative mission

    Selective tidal stream transport in silver European eel (<i>Anguilla anguilla</i> L.) – Migration behaviour in a dynamic estuary

    Different fish species use selective tidal stream transport (STST) to efficiently migrate through strong tidal systems to complete their life cycle, but the use of STST by silver European eels is still controversial. In this study, we found strong evidence that silver European eels apply STST. The results illustrate that eels can distinguish between ebb and flood and suggest that tides play a role in orientation, either directly or indirectly. The general migration speed was higher in the downstream part of the estuary compared to the upstream part, while tidal migration speed was equal in both parts, indicating that eels migrated more consistently in the downstream part. The results of this study give insight in how a diadromous species migrates through an estuary and underline the importance of the tides
