    Stem cell based therapy retards the progression of osteoarthritis and promotes repair of meniscus injury of sheep model knee joint

    ABSTRACT The aim of this study to determine if intra-articular injection of autologous bone marrow mesenchymal stem cells (ABMSCs) could repair surgically induced osteoarthritis in sheep model. Eighteen male healthy sheep (weighed 18-20kg) were divided into two test groups and one control group. The control groups were not different from the test groups with respect to age and weight, but the test animals underwent a bone marrow aspira- tion for cell preparation in the same time of osteoarthritis induction. ABMSCs were isolated from sheep bone marrow and divided into two groups, namely test group A; ABMSCs cultured in FD medium supplemented with 10% fetal bovine serum (FBS), Test group B; ABMSCs were cultured in FD medium supplimented with 1% FBS and 10 ng/ml TGFβ-3 for three weeks. OA was induced by complete excision of the medial meniscus and resection of the anterior cruciate ligament (ACL). Sheep were subjected to exercise for three weeks post OA induction. After 6 weeks post-operation, test groups received direct intra-articular injection of a single dose 10x106 cells suspended in basal medium into injured sheep knee joint. Con- trol animals received basal medium alone. Six weeks post- cel injection, the femoral condyle and the tibial plateau from test and control groups were removed,fixed,photographed, and assessed by two blinded evaluators based on ICRS grading system, decalcified. Specimens were sectioned into 5 µm and stained with H & E and Safranin O. The result demonstrated that Gross observation of femoral condyle and tibia plaetue of the operated knee joint had OA. The severe OA was clearly observed in in control group knee joints. Test group received intra-articular injection of ABMSCs alone showed moderate OA. Interestingly test group B that received intra-articularinjection of TGF-β3 induced ABMSCs showed mild OA. The histological examination showed clear evidence of articular cartilage and menicscus regeneration in test group B of sheep injured knee joint that received TGF-β3 induced ABMSCs when compared with other groups

    Vision is the most important sense in higher mammals. The retina is the first step in visual processing and the window to the brain. It is not surprising that problems arising in the retina lead to moderate to severe visual impairments. We offer here a collection of reviews as well as original papers dealing with various aspects of retinal function as well as dysfunction. New approaches in retinal research are described, such as the expression and localization of the endocannabinoid system in the normal retina and the role of cannabinoid receptors that could offer new avenues of research in the development of potential treatments for retinal diseases. Moreover, new insights are offered in advancing knowledge towards the prevention and cure of visual pathologies, mainly AMD, RP, and diabetic retinopathy

    This is the proceeding of the 3rd Java International Nursing Conference 2015 organized by School of Nursing, Faculty of Medicine, Diponegoro University, in collaboration with STIKES Kendal. The conference was held on 20-21 August 2015 in Semarang, Indonesia.     Antioxidant and DPPH-Scavenging Activities of Compounds and Ethanolic Extract of the Leaf and Twigs of Caesalpinia bonduc L. Roxb.

    Antioxidant effects of ethanolic extract of Caesalpinia bonduc and its isolated bioactive compounds were evaluated in vitro. The compounds included two new cassanediterpenes, 1α,7α-diacetoxy-5α,6β-dihydroxyl-cass-14(15)-epoxy-16,12-olide (1)and 12α-ethoxyl-1α,14β-diacetoxy-2α,5α-dihydroxyl cass-13(15)-en-16,12-olide(2); and others, bonducellin (3), 7,4’-dihydroxy-3,11-dehydrohomoisoflavanone (4), daucosterol (5), luteolin (6), quercetin-3-methyl ether (7) and kaempferol-3-O-α-L-rhamnopyranosyl-(1Ç2)-β-D-xylopyranoside (8). The antioxidant properties of the extract and compounds were assessed by the measurement of the total phenolic content, ascorbic acid content, total antioxidant capacity and 1-1-diphenyl-2-picryl hydrazyl (DPPH) and hydrogen peroxide radicals scavenging activities.Compounds 3, 6, 7 and ethanolic extract had DPPH scavenging activities with IC50 values of 186, 75, 17 and 102 μg/ml respectively when compared to vitamin C with 15 μg/ml. On the other hand, no significant results were obtained for hydrogen peroxide radical. In addition, compound 7 has the highest phenolic content of 0.81±0.01 mg/ml of gallic acid equivalent while compound 8 showed the highest total antioxidant capacity with 254.31±3.54 and 199.82±2.78 μg/ml gallic and ascorbic acid equivalent respectively. Compound 4 and ethanolic extract showed a high ascorbic acid content of 2.26±0.01 and 6.78±0.03 mg/ml respectively.The results obtained showed the antioxidant activity of the ethanolic extract of C. bonduc and deduced that this activity was mediated by its isolated bioactive compounds

    COMMUNITY NURSING PROBLEMS RELATED NON COMMUNICABLE DISEASES (CASE STUDY AT RW 02, KELURAHAN SENTUL, BLITAR CITY)

    Background: Individuals aged 15 years and over as people at risk of suffering noncommunicable diseases are the target of community nursing care. Aim of this study is formulating community nursing problems related to non-communicable diseases as the case study. Methods: This case study used documentary study of nursing practice report. The subjects used were all peoples over the age of 15 years living in RW 02, Kelurahan Sentul, Blitar City as much as 283 people (117 families). Time study conducted February 20 to March 1, 2017. The assessment using questionnaires. Data analysis using critical thinking. After formulating nursing problems continued prioritization shared between subjects, community leaders, and health center nurse. Results: Nursing problems are risky health behaviors with a score of 208 and deficiency community health with a score of 168. The community nursing interventions were done of health education by methods of lectures, discussions, and simulations and aimed to community empowerment will have an autonomy to negotiate the nursing problem. Conclusions: Non-communicable diseases can be used as a target of community nursing
