    Spatially and temporally distinct encoding of muscle and kinematic information in rostral and caudal primary motor cortex

    The organising principle of human motor cortex does not follow an anatomical body map, but rather a distributed representational structure in which motor primitives are com- bined to produce motor outputs. Electrophysiological recordings in primates and human imaging data suggest that M1 encodes kinematic features of movements, such as joint position and velocity. However, M1 exhibits well-documented sensory responses to cu- taneous and proprioceptive stimuli, raising questions regarding the origins of kinematic motor representations: are they relevant in top-down motor control, or are they an epiphe- nomenon of bottom-up sensory feedback during movement? Here we provide evidence for spatially and temporally distinct encoding of kinematic and muscle information in human M1 during the production of a wide variety of naturalistic hand movements. Using a powerful combination of high-field fMRI and MEG, a spatial and temporal multivariate representational similarity analysis revealed encoding of kinematic information in more caudal regions of M1, over 200 ms before movement onset. In contrast, patterns of muscle activity were encoded in more rostral motor regions much later after movements began. We provide compelling evidence that top-down control of dexterous movement engages kinematic representations in caudal regions of M1 prior to movement production

    Associations of thalamocortical networks with reduced mindfulness in alcohol use disorder.

    BACKGROUND Increased mindfulness is associated with reduced alcohol consumption in patients with alcohol use disorder (AUD) after residential treatment. However, the underlying neurobiological mechanism of mindfulness in AUD is unclear. Therefore, we investigate the structural and functional alterations of the thalamocortical system with a focus on the mediodorsal thalamic nucleus (MD-TN), the default mode and the salience network (DMN/SN) which has previously been associated with mindfulness in healthy subjects. We hypothesized lower mindfulness and reduced structural and functional connectivity (FC) of the thalamocortical system, particularly in the DMN/SN in AUD. We assumed that identified neurobiological alterations in AUD are associated with impairments of mindfulness. METHODS Forty-five abstinent patients with AUD during residential treatment and 20 healthy controls (HC) were recruited. Structural and resting-state functional MRI-scans were acquired. We analysed levels of mindfulness, thalamic volumes and network centrality degree of the MD-TN using multivariate statistics. Using seed-based whole brain analyses we investigated functional connectivity (FC) of the MD-TN. We performed exploratory correlational analyses of structural and functional DMN/SN measurements with levels of mindfulness. RESULTS In AUD we found significantly lower levels of mindfulness, lower bilateral thalamic and left MD-TN volumes, reduced FC between MD-TN and anterior cingulum/insula and lower network centrality degree of the left MD-TN as compared to HC. In AUD, lower mindfulness was associated with various reductions of structural and functional aspects of the MD-TN. CONCLUSION Our results suggest that structural and functional alterations of a network including the MD-TN and the DMN/SN underlies disturbed mindfulness in AUD

    Neuropsychological effect on long-term Ayahuasca use

    Ayahuasca is a hallucinogenic tea used in religious ritual ceremonies in eastern parts of South America. Ayahuasca is becoming more popular in western countries for therapeutic use for depression, drug addiction, and emotional distress (Domínguez-Clavé et al., 2016). Previous research has explored the effects ayahuasca may have on cognition, neurological functioning, and psychopathology. Although several studies have been done on cognition and psychopathology, few studies have examined the long-term use of neurological functioning. This study focused on neuropsychological effects chronic recurrent religious users of Ayahuasca may develop, using an existing data set. This study analyzed the default mode network (DMN) comprised of the posterior cingulate cortex, angular gyrus, and the medial prefrontal cortex. To assess the DMN functional magnetic resonance images (fMRI) scans were collected. This study included 12 Ayahuasca users from the U.S. branch of União do Vegetal (UDV) and 13 non-Ayahuasca-using controls from Albuquerque Protestant and Catholic churches. No differences were found between groups on connectivity in any region of interest nor in relation to mood assessments. However, the right medial prefrontal cortex and the angular gyrus revealed a positive associated with sleepiness. Possible effects can be assessed on default mode network (DMN) connectivity using a larger sample size. These findings display evidence that long-term use of Ayahuasca does not have essential lasting neuropsychological effects

    The Dynamics of Functional Brain Networks:Integrated Network States during Cognitive Task Performance

    Higher brain function relies upon the ability to flexibly integrate information across specialized communities of brain regions, however it is unclear how this mechanism manifests over time. In this study, we use time-resolved network analysis of functional magnetic resonance imaging data to demonstrate that the human brain traverses between two functional states that maximize either segregation into tight-knit communities or integration across otherwise disparate neural regions. The integrated state enables faster and more accurate performance on a cognitive task, and is associated with dilations in pupil diameter, suggesting that ascending neuromodulatory systems may govern the transition between these alternative modes of brain function. Our data confirm a direct link between cognitive performance and the dynamic reorganization of the network structure of the brain.Comment: 38 pages, 4 figure

    Confound modelling in UK Biobank brain imaging

    © 2020 Dealing with confounds is an essential step in large cohort studies to address problems such as unexplained variance and spurious correlations. UK Biobank is a powerful resource for studying associations between imaging and non-imaging measures such as lifestyle factors and health outcomes, in part because of the large subject numbers. However, the resulting high statistical power also raises the sensitivity to confound effects, which therefore have to be carefully considered. In this work we describe a set of possible confounds (including non-linear effects and interactions that researchers may wish to consider for their studies using such data). We include descriptions of how we can estimate the confounds, and study the extent to which each of these confounds affects the data, and the spurious correlations that may arise if they are not controlled. Finally, we discuss several issues that future studies should consider when dealing with confounds

    Heterogeneity in early language development in autism spectrum disorder (ASD) is clinically important and may reflect neurobiologically distinct subtypes. Here, we identified a large-scale association between multiple coordinated blood leukocyte gene coexpression modules and the multivariate functional neuroimaging (fMRI) response to speech. Gene coexpression modules associated with the multivariate fMRI response to speech were different for all pairwise comparisons between typically developing toddlers and toddlers with ASD and poor versus good early language outcome. Associated coexpression modules were enriched in genes that are broadly expressed in the brain and many other tissues. These coexpression modules were also enriched in ASD-associated, prenatal, human-specific, and language-relevant genes. This work highlights distinctive neurobiology in ASD subtypes with different early language outcomes that is present well before such outcomes are known. Associations between neuroimaging measures and gene expression levels in blood leukocytes may offer a unique in vivo window into identifying brain-relevant molecular mechanisms in ASD

    A behavioral and brain imaging dataset with focus on emotion regulation of women with fibromyalgia

    Fibromyalgia is a chronic condition characterized by widespread pain, as well as numerous symptoms related to central sensitization such as: fatigue, cognitive disturbances, constipation/diarrhea and sensory hypersensitivity. Furthermore, depression and anxiety are prevalent comorbidities, accompanied by emotion processing and regulation difficulties. Although fibromyalgia physiopathology is still not fully understood, neuroimaging research methods have shown brain structural and functional alterations as well as neuroinflammation abnormalities. We believe that open access to data may help fibromyalgia research advance more. Here, we present an open dataset of 33 fibromyalgia female patients and 33 paired healthy controls recruited from a Mexican population. Dataset includes demographic, clinical, behavioural and magnetic resonance imaging (MRI) data. The MRI data consists of: structural (T1- and T2- weighted) and functional (task-based and resting state) sequences. The task was an emotion processing and regulation task based on visual stimuli. The MRI data contained in the repository are unprocessed, presented in Brain Imaging Data Structure (BIDS) format and available on the OpenNeuro platform for future analysis

    Does the exogeneity of oil prices matter in the oil price-macro-economy relationship for Ghana?

    Using annual data from 1971 to 2014 we consider whether the relationship between crude oil prices and the macro-economy in the relatively small economy of Ghana is affected by the treatment of crude oil prices as exogenous or endogenous. We use vector autoregressions, vector error-correction models, scenario-based dynamic forecasting, and autoregressive distributive lag specifications. There is little evidence that international crude oil prices have a significant negative effect on Ghana’s output in either the short-run and long-run, regardless of whether crude oil prices are treated as exogenous or endogenous. This implies that increases in crude oil prices do not put a binding constraint on the monetary authorities to loosen monetary policy to offset its adverse effect on output. If inflation is a priority, policy makers could focus on inflation stabilization by tightening monetary policy when oil prices rise
