    Perturbation Detection Through Modeling of Gene Expression on a Latent Biological Pathway Network: A Bayesian hierarchical approach

    Cellular response to a perturbation is the result of a dynamic system of biological variables linked in a complex network. A major challenge in drug and disease studies is identifying the key factors of a biological network that are essential in determining the cell's fate. Here our goal is the identification of perturbed pathways from high-throughput gene expression data. We develop a three-level hierarchical model, where (i) the first level captures the relationship between gene expression and biological pathways using confirmatory factor analysis, (ii) the second level models the behavior within an underlying network of pathways induced by an unknown perturbation using a conditional autoregressive model, and (iii) the third level is a spike-and-slab prior on the perturbations. We then identify perturbations through posterior-based variable selection. We illustrate our approach using gene transcription drug perturbation profiles from the DREAM7 drug sensitivity predication challenge data set. Our proposed method identified regulatory pathways that are known to play a causative role and that were not readily resolved using gene set enrichment analysis or exploratory factor models. Simulation results are presented assessing the performance of this model relative to a network-free variant and its robustness to inaccuracies in biological databases

    Achieving Green and Healthy Homes and Communities in America

    In the Fall of 2010, the National Coalition to End Childhood Lead Poisioning contracted with the National Academy to develop and execute an online dialogue that would examine ways to increase the health, safety, and energy efficiency of low- to moderate-income homes. Since 1999, the National Coalition had worked to improve low- to moderate-income housing through the support and execution of home interventions that addressed multiple issues within a home at one time; an approach that often did not align with other traditional, single-issue housing assistance programs. By 2010, the National Coalition had taken on the leadership of the Green and Healthy Homes Initiative, a public-private partnership focused on integrating funding streams to improve low- to middle-income homes across the country.With plans to expand the GHHI's operations, the National Coalition partnered with the National Academy to conduct the National Dialogue on Green and Healthy Homes, a collaborative online dailogue in which participants were asked to identify challenges to, and innovative practices for, improving the health, safety and energy-efficiency of low- to moderate- income homes. The Dialogue was live from November 4-November 22, 2010, and collected 100 hundred ideas and 362 comments from 320 registered users. Over the course of its two and a half week duration, the Dialogue received more than 2,500 visits from over 1,100 people in 48 states and territories. Key FindingsBy reviewing the feedback received in the Dialogue, the Panel was able to make a number of recommendations on how the green and healthy homes community of practice could increase the health, safety and energy efficiency of homes across the country. These recommendations included: Conduct an evaluation of current housing standards to determine if they meet the Nation's health, safety, and energy efficiency needs; Develop a tiered performance standard for healthy, safe and energy efficient homes; Group government funding streams to better align programs with the comprehensive intervention approach; Develop a long-term funding strategy to support efforts after Recovery Act funding ends; and Educate government decisionmakers and the public on the importance of developing green and healthy homes and communities, and the work that supports that development

    Remarkable improvements have occurred recently in the maintenance management of physical assets and productive systems, so that less wastages of energy and resources occur. The requirement for optimal preventive maintenance using, for instance, justin-time (JIT) and total quality-management (TQM) techniques has given rise to whathas been called the total productive-maintenance (TPM) approach. This study explores the ways in which Nigerian manufacturing industries can implement TPM as a strategy and culture for improving its performance and suggests self-auditing and bench-marking as desirable prerequisites before TPM implementation

    Gender Mainstreaming Case Study - India: Chhattisgarh Irrigation Development Project

    Project Goals and Results • The Chhattisgarh Irrigation Development Project (CIDP) aimed to increase agricultural productivity, improve rural livelihoods, and reduce poverty by improving irrigation delivery, enhancing agricultural practices, and strengthening water resource management. • The CIDP supported the rehabilitation and upgrading of irrigation systems that serve more than 150,000 households; strengthened government capacity to assist water users’ associations (WUAs); and trained 1,250 out of 1,324 WUAs in Chhattisgarh based on the Participatory Irrigation Management (PIM) Act, a legislation that mandated the roles and responsibilities of WUAs, including the membership of women in irrigation management. It also trained 191 WUAs on how to improve agricultural techniques and how to be more effective in general. It offered opportunities for collective actions by communities, such as joint purchase of seeds, fertilizers, and pesticides, and joint marketing of produce. Overall, participants reported improved crop yields and higher household incomes. • Building on the new PIM Act, the CIDP strengthened women’s participation in WUAs. A widespread education campaign raised awareness on the importance of women’s participation in decision making and encouraged women to become active in these associations. In the 2007 WUA elections, women were elected in one-third of available positions. Processes and Management Tools • The PIM Act was passed through the CIDP. With this new law, the CIDP promoted PIM, especially women’s participation. The project predated the requirement for a Gender Action Plan and gender targets and indicators in the Design and Monitoring Framework (DMF) outputs. Gender-related strategies and targets were indicated in the Summary Poverty Reduction and Social Strategy. • All irrigation upgrading and rehabilitation activities included contractual obligations to offer “equal pay for equal work” to men and women. • Technical assistance supported activities to promote women’s participation in irrigation management and their election to WUA committees

    Civic Republicanism Provides Theoretical Support for Making Individuals More Environmentally Responsible

    The genesis for this essay is the recognition that individual behavior is contributing in a significant way to the remaining environmental problems we have. For a variety of reasons, ranging from the difficulty of trying to identify and then regulate all of these individual sources to the political backlash that might result if such regulation was tried, efforts to control that behavior have either failed or not been tried. The phenomenon of individuals as irresponsible environmental actors seems counter-intuitive given the durability of the environmental protection norm and polls that consistently show that people contribute to environmental causes, are willing to pay more to protect environmental resources, and consider protecting the environment among their highest priorities. This conflict between thought and deed and its serious effect, if not resolved, is the puzzle that has sent me on this quest. This essay is the author\u27s third attempt at unraveling the problem of irresponsible individual environmental behavior and at suggesting possible ways to reform how people behave toward the environment. The first article proposed expanding the abstract environmental protection norm to include individual environmental responsibility as the approach most likely to overcome barriers to behavioral change. The article recommended enlisting environmental groups as the most effective norm entrepreneurs to achieve widespread change in personal environmental conduct. In that piece, she concluded that the best way to change norms and thus change behavior was through education, but additional measures might be necessary. The second article expanded on the earlier discussion of norms and their influence on behavior, and why changing norms, though difficult, is more effective than other means of inciting behavioral change. However, given the difficulty inherent in creating or changing norms, the second article also identified and evaluated other norm and behavior-changing tactics, such as shaming, public education, and market-based incentives, which might supplement norms as a means of changing behavior. The article concluded that no one approach alone is sufficient to secure both norm and behavior change, but a combination of any or all of them when properly tailored to the source and nature of the harm and when accompanied by public education can lead to both norm and behavioral changes. Thus, both articles concluded that public education plays a critical role in any effort to alter public behavior through changing norms. This essay examines how republican theory supports that conclusion and provides the theoretical framework within which norm change can occur. All three pieces start with the premise that the current crisis over global climate change has created the circumstances in which norm change can occur--circumstances that collectively have created what the author calls a second environmental republican moment. This second republican moment, like the first one in the 1970s, might result in widespread public support for a variety of environmentally protective legislative and regulatory initiatives and offers a rare, albeit brief, opportunity in which to educate the public about its contribution to environmental harm. This essay develops the republican aspect of that thought further, demonstrating how the overlapping strands of republican thought and norm development support the creation of a new norm of personal environmental responsibility. The essay also shows how, during republican moments, the public is more amenable to being educated about civic matters, including their responsibilities as environmental citizens. It is particularly during republican moments that people acquire information that may influence their expressed preferences, lending a sense of urgency to the present moment we find ourselves in. This essay begins by discussing the concept of an environmental republican moment, and why the public\u27s response to the crisis of global climate change appears to be such a moment. The essay then identifies the key features of republican theory and shows how those features replicate many of the elements necessary for norm and behavioral change. The essay concludes by showing how republicanism--with its emphasis on public education, civic involvement, and achieving the common good through civic virtue--provides a useful construct for thinking about how to make people behave in more environmentally responsible ways

    Improving WASH Service Delivery in Protracted Crises: The Case of the Democratic Republic of Congo

    Delivering Water, Sanitation and Hygiene (WASH) services during humanitarian emergencies and immediate recovery phases is essential for saving lives and responding to basic needs, yet choices about how WASH services are delivered can undermine future development and peace. Longer-term interventions can also overlook how they equip communities, households and government to prepare and respond to future emergencies. This is increasingly evident in protracted or recurrent crises, in which overlapping and cyclical phases of emergency, relief, recovery and development interventions coexist. In these contexts, practitioners and academics alike have acknowledged the problem of reconciling the fundamentally different institutional cultures, assumptions, values, structures and ways of working that characterise the humanitarian and the development communities.In this report, we analyse humanitarian and development approaches in a specific sector, in a particular country: WASH interventions in the Democratic Republic of Congo (DRC). We consider how and why siloes have arisen. We argue that the problem is not so much about filling a 'gap' between humanitarian and development siloes, but about aligning the principles and practices of both communities in specific contexts so that the overall response can meet changing needs and constraints. We identify a number of ways through which improved complementarity might be achieved, differentiating between national and sub-national levels
