    Identifying Agile Requirements Engineering Patterns in Industry

    Agile Software Development (ASD) is gaining in popularity in today´s business world. Industry is adopting agile methodologies both to accelerate value delivery and to enhance the ability to deal with changing requirements. However, ASD has a great impact on how Requirements Engineering (RE) is carried out in agile environments. The integration of Human-Centered Design (HCD) plays an important role due to the focus on user and stakeholder involvement. To this end, we aim to introduce agile RE patterns as main objective of this paper. On the one hand, we will describe our pattern mining process based on empirical research in literature and industry. On the other hand, we will discuss our results and provide two examples of agile RE patterns. In sum, the pattern mining process identifies 41 agile RE patterns.     Teaching telecommunication standards: bridging the gap between theory and practice

Standards are needed to enable cooperation while promoting competition. Within the framework of a standard, the companies involved in the standardization process contribute and agree on appropriate technical specifications to ensure diversity and compatibility, and facilitate worldwide commercial deployment and evolution. Those parts of the system that can create competitive advantages are intentionally left open in the specifications. Such specifications are extensive, complex, and minimalistic. This makes telecommunication standards education a difficult endeavor, but it is much demanded by industry and governments to spur economic growth. This article describes a methodology for teaching wireless communications standards. We define our methodology around six learning stages that assimilate the standardization process and identify key learning objectives for each. Enabled by software-defined radio technology, we describe a practical learning environment that facilitates developing many of the needed technical and soft skills without the inherent difficulty and cost associated with radio frequency components and regulation. Using only open source software and commercial of-the-shelf computers, this environment is portable and can easily be recreated at other educational institutions and adapted to their educational needs and constraints.     The Impact of Human Error in the Use of Agricultural Tractors: A Case Study Research in Vineyard Cultivation in Italy

    Recently, standards and regulations concerning occupational safety have become more and more rigorous. Nevertheless, the number of accidents and victims has not decreased significantly, as reported by official statistics. In Italy, the agricultural sector is certainly one of the most affected by this situation, especially taking into account the occurrence of serious injuries and fatalities related to the use of tractors. The main reasons for such a situation can be ascribed to the peculiarities of agricultural operations. Therefore, when analyzing the root causes of agricultural accidents, a user-centered approach is needed in order to make the development of health and safety interventions easier and more effective. Based on this, the present paper proposes a practical case study research focused on integrating the factor of human error into the risk assessment procedures of agricultural activities in vineyard cultivation. Such an approach allowed us to consider the impact of human errorwhile performing work activities (e.g., the use of a tractor)on hazards and related hazardous events in a thorough manner. The proposed approach represents a novelty in the sector of the safety assessment of agricultural activities, providing a first valuable basis for further analysis and implementation by researchers and practitioners

    Targeted mutagenesis in a human-parasitic nematode.

    Parasitic nematodes infect over 1 billion people worldwide and cause some of the most common neglected tropical diseases. Despite their prevalence, our understanding of the biology of parasitic nematodes has been limited by the lack of tools for genetic intervention. In particular, it has not yet been possible to generate targeted gene disruptions and mutant phenotypes in any parasitic nematode. Here, we report the development of a method for introducing CRISPR-Cas9-mediated gene disruptions in the human-parasitic threadworm Strongyloides stercoralis. We disrupted the S. stercoralis twitchin gene unc-22, resulting in nematodes with severe motility defects. Ss-unc-22 mutations were resolved by homology-directed repair when a repair template was provided. Omission of a repair template resulted in deletions at the target locus. Ss-unc-22 mutations were heritable; we passed Ss-unc-22 mutants through a host and successfully recovered mutant progeny. Using a similar approach, we also disrupted the unc-22 gene of the rat-parasitic nematode Strongyloides ratti. Our results demonstrate the applicability of CRISPR-Cas9 to parasitic nematodes, and thereby enable future studies of gene function in these medically relevant but previously genetically intractable parasites

    A Longitudinal Study of Identifying and Paying Down Architectural Debt

    Architectural debt is a form of technical debt that derives from the gap between the architectural design of the system as it "should be" compared to "as it is". We measured architecture debt in two ways: 1) in terms of system-wide coupling measures, and 2) in terms of the number and severity of architectural flaws. In recent work it was shown that the amount of architectural debt has a huge impact on software maintainability and evolution. Consequently, detecting and reducing the debt is expected to make software more amenable to change. This paper reports on a longitudinal study of a healthcare communications product created by Brightsquid Secure Communications Corp. This start-up company is facing the typical trade-off problem of desiring responsiveness to change requests, but wanting to avoid the ever-increasing effort that the accumulation of quick-and-dirty changes eventually incurs. In the first stage of the study, we analyzed the status of the "before" system, which indicated the impacts of change requests. This initial study motivated a more in-depth analysis of architectural debt. The results of this analysis were used to motivate a comprehensive refactoring of the software system. The third phase of the study was a follow-on architectural debt analysis which quantified the improvements made. Using this quantitative evidence, augmented by qualitative evidence gathered from in-depth interviews with Brightsquid's architects, we present lessons learned about the costs and benefits of paying down architecture debt in practice.Comment: Submitted to ICSE-SEIP 201

    ICS Materials. Towards a re-Interpretation of material qualities through interactive, connected, and smart materials.

    The domain of materials for design is changing under the influence of an increased technological advancement, miniaturization and democratization. Materials are becoming connected, augmented, computational, interactive, active, responsive, and dynamic. These are ICS Materials, an acronym that stands for Interactive, Connected and Smart. While labs around the world are experimenting with these new materials, there is the need to reflect on their potentials and impact on design. This paper is a first step in this direction: to interpret and describe the qualities of ICS materials, considering their experiential pattern, their expressive sensorial dimension, and their aesthetic of interaction. Through case studies, we analyse and classify these emerging ICS Materials and identified common characteristics, and challenges, e.g. the ability to change over time or their programmability by the designers and users. On that basis, we argue there is the need to reframe and redesign existing models to describe ICS materials, making their qualities emerge

    Methods for Scarless, Selection-Free Generation of Human Cells and Allele-Specific Functional Analysis of Disease-Associated SNPs and Variants of Uncertain Significance.

    With the continued emergence of risk loci from Genome-Wide Association studies and variants of uncertain significance identified from patient sequencing, better methods are required to translate these human genetic findings into improvements in public health. Here we combine CRISPR/Cas9 gene editing with an innovative high-throughput genotyping pipeline utilizing KASP (Kompetitive Allele-Specific PCR) genotyping technology to create scarless isogenic cell models of cancer variants in ~1 month. We successfully modeled two novel variants previously identified by our lab in the PALB2 gene in HEK239 cells, resulting in isogenic cells representing all three genotypes for both variants. We also modeled a known functional risk SNP of colorectal cancer, rs6983267, in HCT-116 cells. Cells with extremely low levels of gene editing could still be identified and isolated using this approach. We also introduce a novel molecular assay, ChIPnQASO (Chromatin Immunoprecipitation and Quantitative Allele-Specific Occupation), which uses the same technology to reveal allele-specific function of these variants at the DNA-protein interaction level. We demonstrated preferential binding of the transcription factor TCF7L2 to the rs6983267 risk allele over the non-risk. Our pipeline provides a platform for functional variant discovery and validation that is accessible and broadly applicable for the progression of efforts towards precision medicine

    Evaluation of Landsat-8 and Sentinel-2A Aerosol Optical Depth Retrievals Across Chinese Cities and Implications for Medium Spatial Resolution Urban Aerosol Monitoring

    In urban environments, aerosol distributions may change rapidly due to building and transport infrastructure and human population density variations. The recent availability of medium resolution Landsat-8 and Sentinel-2 satellite data provide the opportunity for aerosol optical depth (AOD) estimation at higher spatial resolution than provided by other satellites. AOD retrieved from 30 m Landsat-8 and 10 m Sentinel-2A data using the Land Surface Reflectance Code (LaSRC) were compared with coincident ground-based Aerosol Robotic Network (AERONET) Version 3 AOD data for 20 Chinese cities in 2016. Stringent selection criteria were used to select contemporaneous data; only satellite and AERONET data acquired within 10 min were considered. The average satellite retrieved AOD over a 1470 m1470 m window centered on each AERONET site was derived to capture fine scale urban AOD variations. AERONET Level 1.5 (cloud-screened) and Level 2.0 (cloud-screened and also quality assured) data were considered. For the 20 urban AERONET sites in 2016 there were 106 (Level 1.5) and 67 (Level 2.0) Landsat-8 AERONET AOD contemporaneous data pairs, and 118 (Level 1.5) and 89 (Level 2.0) Sentinel-2A AOD data pairs. The greatest AOD values (>1.5) occurred in Beijing, suggesting that the Chinese capital was one of the most polluted cities in China in 2016. The LaSRC Landsat-8 and Sentinel-2A AOD retrievals agreed well with the AERONET AOD data (linear regression slopes > 0.96; coefficient of determination r(exp 2) > 0.90; root mean square deviation < 0.175) and demonstrate that the LaSRC is an effective and applicable medium resolution AOD retrieval algorithm over urban environments. The Sentinel-2A AOD retrievals had better accuracy than the Landsat-8 AOD retrievals, which is consistent with previously published research.The implications of the research and the potential for urban aerosol monitoring by combining the freely available Landsat-8 and Sentinel-2 satellite data are discussed
