    Age-dependent sensitization to the 7S-vicilin-like protein Cor a 11 from hazelnut (Corylus avellana) in a birch-endemic region

    Background: Hazelnut (Corylus avellana) allergy exhibits age and geographically distinct sensitization patterns that have not yet been fully resolved. Objective: To study sensitization to Cor a 11 in different age groups of hazelnut-allergic patients and infants with atopic dermatitis (AD) sensitized to hazelnut in a birch-endemic region. Methods: Sera from 80 hazelnut-allergic patients, 33 infants under 1 year of age with AD (24 sensitized and 9 not sensitized to hazelnut), 32 healthy control individuals, and 29 birch pollen–allergic but hazelnut-tolerant individuals were tested for immunoglobulin (Ig) E reactivity to Cor a 11 by ImmunoCAP. IgE reactivity to Cor a 1.01, Cor a 1.04, Cor a 8, and Cor a 9 was studied by ISAC microarray. Results: Forty patients (22 preschool children, 10 schoolchildren, and 8 adults) with systemic reactions on consumption of hazelnut were sensitized to Cor a 11 (respective rates of 36%, 40%, and 12.5%). Forty patients (6 preschool children, 10 schoolchildren, and 24 adults) reported oral allergy syndrome but only 2 of them (of preschool age) were sensitized to Cor a 11. Two (8%) of the AD infants sensitized to hazelnut showed IgE reactivity to Cor a 11. This reactivity was not observed in any of the AD infants without sensitization to hazelnut, in any of the birch-pollen allergic patients without hazelnut allergy, or in any of the healthy control individuals. Conclusion: Sensitization to Cor a 11 in a birch-endemic region is predominantly found in children with severe hazelnut allergy, a finding that is consistent with observations concerning sensitization to Cor a 9

    Analyses of phytosterol classes of olive and hazelnut oils collected from different countries by TLC, GC and GC-MS revealed considerable quantitative differences. The composition of 4-desmethyl- and 4-monomethylsterols was similar in both oils, but 4,4'-dimethylsterols composition differed. Lupeol and an unknown (lupane skeleton) compound were exclusively present in hazelnut oil 4,4´-dimethylsterols and could be used as markers to detect virgin olive oil adulteration with hazelnut oil at levels below 4%. Conventional TLC to separate phytosterol classes has a low recovery rate and is time-consuming. A new SPE method to separate phytosterol classes was developed with stepwise elution by increasing the polarity of the n-hexane:diethyl ether solvent mixture. Comparison of the results obtained for hazelnut and virgin olive oils with those of TLC revealed that the SPE method was faster and gave higher sterol recovery rates. Free and esterified forms of sterols provide detailed information on the identity and quality of vegetable oils, and therefore 4,4´-dimethylsterols were investigated in hazelnut oil and virgin olive oil. A sample of solvent-extracted hazelnut oil was refined to monitor the effects of processing on 4,4´-dimethylsterol levels and on specific marker compounds. Of the refining processes tested, only neutralisation and bleaching considerably reduced 4,4´-dimethylsterols. In fully-refined hazelnut oil, losses of marker compounds in free form were higher than losses in their esterified form. GC-MS analysis showed that adulteration of olive oil with fully-refined hazelnut oil could be detected at levels of 2% by tracing lupeol in total/esterified forms of 4,4´-dimethylsterols. Olive oil has many applications in the food industry, e.g. blended with oils such as palm stearin to produce margarine or shortening by chemical interesterification. Investigation on lipid and minor lipid components of an olive oil-palm stearin blend during chemical interesterification showed that sterols were esterified with fatty acids at a higher level at 120 °C (7%) than at 90 °C (4%). Despite heat treatment and several steps to produce an interesterified product, there were minor losses in phytosterol and tocopherol contents and no significant increases in phytosterol oxidation

    The adulteration of virgin olive oil with hazelnut oil is a common fraud in the food industry, which makes mandatory the development of accurate methods to guarantee the authenticity and traceability of virgin olive oil. In this work, we demonstrate the potential of a rapid luminescent method to characterize edible oils and to detect adulterations among them. A regression model based on five luminescent frequencies related to minor oil components was designed and validated, providing excellent performance for the detection of virgin olive oil adulteration

    Poplar from phytoremediation as a renewable energy source:gasification properties and pollution analysis

    Biomass gasification is a very efficient process to produce clean energy in the form of a fuel gas (syngas). Hazelnut shells and poplar have good energy production potential and they are abundant in nature. Hazelnut shells have the characteristics of a very good fuel and poplar is among the fastest growing trees; furthermore, poplar demonstrated the capability to absorb organic contaminants (i.e. heavy metals) from the soil in which they are cultivated. However, poplar is not usually used for biomass gasification and its potential is not fully assessed. Here, 3 types of biomass, hazelnut shells (HS), simple poplar (P) and poplar coming from a phytoremediation procedure (PHYP), were chosen as representative samples to be characterized and tested in a steam gasification process carried out on a bench scale fluidized bed gasifier. A comparison is reported on gasification results, such as gas composition, tar production and gas yield for the biomass feedstocks mentioned above. It was concluded that hazelnut shells and poplar (P and PHYP) could be easily gasified in a fluidized bed gasifier, thus producing a good quality gas with low polluting by-products. The PHYP sample showed lower tar content and higher gas yield. It is guessed that Ca and Mg, found in higher quantities in the PHYP sample, could have had a catalytic effect in tar reforming thus producing lower quantity of heavy hydrocarbons

    Development of a new hazelnut sandwich ELISA based on detection of Cor a 9, a major hazelnut allergen

    The emerging health problem of food-induced allergic reactions, including life-threatening anaphylaxis, presents an important challenge to the food. In the framework of the current ALLERRISK project, analytical approaches for allergen detection are evaluated and developed. Cor a 9 is a major hazelnut protein with nutrient reservoir function, and that also known as a major food allergen. Presence of Cor a 9 indicates contamination of a product with hazelnut and, consequently, potential risk of allergenicity. Crude extract of hazelnut was obtained from milled mixture of hazelnuts from 10 commercially available brands. Then the extract was concentrated and applied to gel filtration and Con A affinity chromatography columns for Cor a 9 isolation and purification. The final product was characterized by analytical gel permeation chromatography, SDS PAGE and MALDI mass spectrometry. The purified Cor a 9 was used as an antigen for the production of polyclonal antibodies in chickens according to the protocol developed in our laboratory. A part of the most active antibodies was further purified by gel permeation chromatography and characterized by SDS PAGE before further use. The prepared antibodies were applied for development of a sandwich ELISA using polyclonal antibody-enzyme adducts as a tracer. Optimization of reagent concentrations led us to achieve detection limit ~10 ng/ml. Influence of ionic strength, pH and buffer composition on analytical parameters of the developed ELISA were thoroughly studied. The developed ELISA can be successfully applied for quantitative detection of Cor a 9 in large pH (pH = 5 – 10) and ionic strength (0.05 – 1.5 M salts solution) ranges. Cross-reactivity with series of nuts, ovalbumin, whey, and wheat proteins was also investigated (Figure 2) and showed that the developed assay was very specific. Moreover, presence of several other proteins (peanut, cashew, ovalbumin, whey and wheat) at high concentrations (1 mg/ml) in the presence of Cor a 9 did not influence the calibration curve of Cor a 9. Detection of hazelnut protein in cookies with known amount of hazelnut inside initial dough showed good recovery (35-40%) and absence of significant signal in blank samples (i.e. without spiking of hazelnut)

    Trace elements in Turkish biomass fuels: Ashes of wheat straw, olive bagasse and hazelnut shell

    Ash contents of wheat straw, olive bagasse and hazelnut shells were 7.9%, 3.9%, 1.2%, respectively, which seemed to be within the average values of ash of biomass. The microstructure of ashes included smooth, polygonal, granular and molten drop structures. A large percentage of particles present in ashes are commonly 1–20 lm in size. SEM/EDS analyses performed on the major ash forming elements in different ashes indicated that Si, Ca, K and Mg and P were generally the most abundant species. Trace element levels in ash samples of various biomass types such as hazelnut shell, wheat straw, olive bagasse were analysed using ICP spectroscopy. The elements determined were some of those considered being of great environmental concern such as, Cr, Mn, Fe, Co, Ni, Cu, Zn, Pb. In all of the ashes studied Fe had the highest concentration among other trace elements, Mn was the second element that exhibited higher concentrations. The order of concentration of elements in the ashes from the highest to the lowest values was as follows: Fe > Mn > Zn > Cu > Ni > Cr > Pb > Co

    A Mesolithic settlement site at Howick, Northumberland: a preliminary report

    Excavations at a coastal site at Howick during 2000 and 2002 have revealed evidence for a substantial Mesolithic settlement and a Bronze Age cist cemetery. Twenty one radiocarbon determinations of the earlier eighth millennium BP (Cal.) indicate that the Mesolithic site is one of the earliest known in northern Britain. An 8m core of sediment was recovered from stream deposits adjacent to the archaeological site which provides information on local environmental conditions. Howick offers a unique opportunity to understand aspects of hunter-gatherer colonisation and settlement during a period of rapid palaeogeographical change around the margins of the North Sea basin, at a time when it was being progressively inundated by the final stages of the postglacial marine transgression. The cist cemetery will add to the picture of Bronze Age occupation of the coastal strip and again reveals a correlation between the location of Bronze Age and Mesolithic sites which has been observed elsewhere in the region
