    Making machines of animals: the international livestock exposition, 1900-1920

    This dissertation examines the establishment and influence of the International Livestock Exposition, an annual show that began in Chicago in 1900 and that served as the central hub of the national livestock improvement movement. Industrial meatpacking firms and land-grant university professors worked together to transform the genetic composition and physiology of American meat-producing animals. Packers hosted the Exposition at the Union Stockyards to address market irregularities in quality and supply. University researchers intended to solve a larger set of problems that included rural population decline, the need for more food output to feed a growing population, and diminishing soil fertility. These unlikely partners created the International to eliminate inferior, or “scrub,” livestock. The International played a pivotal role in remaking livestock genotypes and phenotypes. Its organizers and participants favored “improved” animals descended from purebred, British livestock with recorded ancestries—a preference rooted in the reformers’ pseudo-scientific belief in eugenics. Purebred animals had standard bodies with a narrow set of physiological outcomes, which amounted to biotic technology. But genetic homogeneity was only a building block for improvement. The International also employed contests, demonstrations, and advocacy to reconfigure American livestock by making them smaller, more compact, and early-maturing. This study also analyzes the larger shift in American agriculture toward the Corn Belt model of grain feeding. Treating animals as dynamic historical agents, it suggests that machinery, tractors, seeds, and implements did not alone accomplish the industrialization of agriculture. Meat-producing cattle, sheep, and pigs were a requisite component in an emerging industrial sequence. These grain-fed modern livestock and their farmer caretakers fit into a developing web of mutually dependent agricultural specialists. The International united this movement into a singular body at the end of each year in Chicago, and in the process, shaped American agricultural practices and encouraged farm specialization until the show closed in 1975.     Genomic and proteomic approaches in pig meat quality research field

    Pig meat and carcass quality is a complex concept determined by environmental and genetic factors concurring to the phenotypic variation in qualitative characteristics of meat (fat content, tenderness, juiciness, flavor,etc). This thesis shows the results of different investigations to study and to analyze pig meat and carcass quality focusing mainly on genomic; moreover proteomic approach has been also used. The aim was to analyze data from association studies between genes considered as candidate and meat and carcass quality in different pig breeds. The approach was used to detect new SNP in genes functionally associated to the studied traits and to confirm as candidate other genes already known. Five polymorphisms (one new SNP in Calponin 1 gene and four additional polymorphism already known in other genes) were considered on chromosome 2 (SSC2). Calponin 1 (CNN1) was associated to the studied traits and furthermore the results reported confirmed the data already known for Lactate dehydrogenase A (LDHA), Low density lipoprotein receptor (LDLR), Myogenic differentiation 1 (MYOD1) e Ubiquitin-like 5 (UBL5), in Italian Large White pigs. Using an in silico search it was possible to detect on SSC2 a new SNP of Deoxyhypusine synthase (DHPS) gene partially overlapping with WD repeat domain 83 (WDR83) gene and significant for the meat pH variation in Italian Large White (ILW) pigs. Perilipin 1 (PLIN1) mapping on chromosome 7 and Perilipin 2 (PLIN2) mapping on chromosome 1 were studied and the results obtained in Duroc breed have shown significant associations with carcass traits. Moreover a study of protein composition of porcine LD muscle, indicated an effect of temperature treatment of carcass, on proteins of the sarcoplasmic fraction and in particular on PGM1 phosphorylation. Future studies on pig meat quality should be based on the integration of different experimental approaches (genomics, proteomics, transcriptomics, etc)

    Aromatic and proteomic analyses corroborate the distinction between Mediterranean landraces and modern varieties of durum wheat

    In this paper volatile organic compounds (VOCs) from durum wheat cultivars and landraces were analyzed using PTR-TOF-MS. The aim was to characterize the VOC’s profile of the wholemeal flour and of the kernel to find out if any VOCs were specific to varieties and sample matrices. The VOC data is accompanied by SDS-PAGE analyses of the storage proteins (gliadins and glutenins). Statistical analyses was carried out both on the signals obtained by MS and on the protein profiles. The difference between the VOC profile of two cultivars or two preparations of the same sample - matrices, in this case kernel vs wholemeal flour - can be very subtle; the high resolution of PTR-TOF-MS - down to levels as low as pptv - made it possible to recognize these differences. The effects of grinding on the VOC profiles were analyzed using SIMPER and Tanglegram statistical methods. Our results show that it is possible describe samples using VOC profiles and protein data
