    STRUCTURE, AGENCY AND CHANGE IN THE CAR REGIME. A REVIEW OF THE LITERATURE

    This paper is aimed at filling the gap between the already well structured literature on the 'car regime' and the debate on policies for sustainable transport. Two main results emerge from the literature on the past and current evolution of the car regime: ? the car regime was established thanks to the ability of purposeful private actors to use the technology of internal combustion to influence markets and institutions, and finally society as a whole; ? previous attempts to make urban and regional mobility more sustainable fail because multiple – and mutually reinforcing – path-dependence phenomena lock the society into the car regime. For the future, the dominant scenario appears to be the internal transformation of the existing car regime, which is currently driven by the automotive industry and based on hybrid technology; the emergence of an alternative electric car regime – driven by producers of batteries and managers of electric utilities – remains a secondary option. Further research is needed to understand how – starting from the existing alternatives to the car and the innovations in the car itself – a coalition of public and private actors may be promoted and sustained to create a new regime of sustainable mobility.Car-based mobility; Regime; Sustainable Transport; Transport Policy

    This thesis is based upon a prolonged research period, wherein a practical systems based tool (prototype), was researched, developed and tested, so as to gain outputs of integration improvements for service delivery in South Africa (SA) specifically, and in general for developmental economies. The research question can be summarised as: "to develop a systems-based intervention tool, able to provide practical integration improvements from concept to delivery". Existing systems methods and approaches were accessed, and based upon their utility for the local context, were used to varying degrees, in "building" the prototype, which was tested across a number of interventions, categorised under "world of the client"; and "world of the designer" (firm created for this purpose). Being aware of local and international implementation challenges by virtue of experience as consultant for a number of governments, whereby national planning and implementation techniques tend to be embed mechanistic models of thinking directly affecting how agents and agencies: understand the problem; plan to resolve the problem; and implement the designed solutions. The research sought to recover key systems insights in order to build a practical tool that could reduce negative outcomes, perpetrated by well-intended reforms, having limited integrative thinking, planning and delivery. The research required long-term observation, reflection, and extensive literature review.     Since the mid 1900’s, economist have come to recognize the role of innovative activity in firms productivity growth, particularly in the competitive market economies. However, the most conducive market environment for innovative activity has also become a subject of interest. Thus, a major constraint on industrial dynamism in African countries is said to be the dearth of indigenous entrepreneurs.     A framework to identify epigenome and transcription factor crosstalk

    While changes in chromatin are integral to transcriptional reprogramming during cellular differentiation, it is currently unclear how chromatin modifications are targeted to specific loci. To systematically identify transcription factors (TFs) that can direct chromatin changes during cell fate decisions, we model the genome-wide dynamics of chromatin marks in terms of computationally predicted TF binding sites. By applying this computational approach to a time course of Polycomb-mediated H3K27me3 marks during neuronal differentiation of murine stem cells, we identify several motifs that likely regulate dynamics of this chromatin mark. Among these, the motifs bound by REST and by the SNAIL family of TFs are predicted to transiently recruit H3K27me3 in neuronal progenitors. We validate these predictions experimentally and show that absence of REST indeed causes loss of H3K27me3 at target promoters in trans, specifically at the neuronal progenitor state. Moreover, using targeted transgenic insertion, we show that promoter fragments containing REST or SNAIL binding sites are sufficient to recruit H3K27me3 in cis, while deletion of these sites results in loss of H3K27me3. These findings illustrate that the occurrence of TF binding sites can determine chromatin dynamics. Local determination of Polycomb activity by Rest and Snail motifs exemplifies such TF based regulation of chromatin. Furthermore, our results show that key TFs can be identified ab initio through computational modeling of epigenome datasets using a modeling approach that we make readily accessible

    State and national initiatives have repositioned elementary teachers’ emphases. These mandates increase the frequency with which teachers utilize informational texts and students’ exposure to diverse perspectives of the same event or era. In short, history and social studies content will likely have a more prominent position within the incorporated literature in English/reading class. Teachers will intentionally supplement age-appropriate, engaging tradebooks with relevant, interrelated informational texts, like primary source material. To guide interested elementary teachers, we focused on tradebooks that centered on Native Americans, an oft-included topic in elementary curricula. We evaluated the tradebooks for their historical representation (and misrepresentation), located germane primary source material, and proffered discipline-specific activities

    Retrotransposon Tto1: functional analysis and engineering for insertional mutagenesis

    Retrotransposons are genomic parasites activated by stress conditions that can be seriously detrimental for their host. In this work I demonstrate that Tto1, a typical plant LTR retrotransposon with insertion preference into genes can be turned into a synthetic molecular tool for gene tagging in plants and can be used to predict models for its replication steps. Although retrotransposons have been already used in plant mutagenesis, such application always required establishing protocols for tissue cultures and regeneration in vitro. Here, I show that sequence engineering of Tto1 provides the possibility to obtain transposition in vivo, with a simple screening method based on PCR and with the advantage to skip all in vitro manipulations. An artificial -estradiol inducible promoter has been used to obtain transposition “on demand” in Arabidopsis plants, which generates stable unlinked insertions that follow mendelian segregation in the progeny. Comparing serial deletions of 3’ LTR of the engineered inducible Tto1 (iTto1), I have mapped its two natural terminators and identified the “minimal” R (redundant) region required to achieve the complete reverse transcription of the genomic mRNA into a new cDNA copy. Interestingly, the transcripts ending at the major “early” terminator cannot support reverse transcription, suggesting a mechanism of natural control on the expression. Transcripts with a more extended termination point contain 100 essential nucleotides that define the active nucleus of the R region. This sequence promotes the formation of a stable hairpin structure that “kisses” a complementary identical hairpin on the cDNA and determines the formation of the characteristic cDNA/mRNA heteroduplex. Since the LTR is a repeated sequence the definition of a minimal redundant region has also the important implication to reduce the only possible target for sequence-based gene silencing, which should lead to an increase of the mutagenic efficiency of iTto1. Additional investigations have been carried out in attempt to identify points of improvement of iTto1 performances. By sequence alignment I identified different versions of the integrase that might have influence on insertion efficiency. Furthermore I tested the pOp6/LhGR-N system that will provide higher expression levels in different host plants. The final goal of my work is to extend the application of iTto1 to crop mutagenesis, therefore a big part of my work has been spent to develop Tto1 constructs with activity in barley. Transgenic plants have been obtained, however the constructs still need further experimentation
