    Benchtop Centrifuge for Materials Science

    The Benchtop Centrifuge was designed to serve for research purposes within theMechanical Engineering Department at Santa Clara University. The prototype has been completely assembled and is functioning to the desired specifications of applying up to 1000 g’s of force for over 4 hours. The current uses are anticipated for separation of particles within materials for material processing and testing. The overall systemdesign has been adapted froma legacy project within the University. Various tests were conducted in order to ensure safety and usability of the system. Through Abaqus analysis and drop-test experiments, it was found enclosure itself can withstand an impact from a bucket at max-speed. The a SolidWorks analysis, the natural frequency of the enclosure was found to be 104.46 Hz, which translates to a rotational speed of 6267.6 RPM; this is well above what the system will be operating at. The team hopes that future students and faculty will be able to expand their current research through the use of this system

    The aim of this manual is to provide a comprehensive practical tool for the generation and analysis of genetic data for subsequent application in aquatic resources management in relation to genetic stock identification in inland fisheries and aquaculture. The material only covers general background on genetics in relation to aquaculture and fisheries resource management, the techniques and relevant methods of data analysis that are commonly used to address questions relating to genetic resource characterisation and population genetic analyses. No attempt is made to include applications of genetic improvement techniques e.g. selective breeding or producing genetically modified organisms (GMOs). The manual includes two ‘stand-alone’ parts, of which this is the second volume: Part 1 – Conceptual basis of population genetic approaches: will provide a basic foundation on genetics in general, and concepts of population genetics. Issues on the choices of molecular markers and project design are also discussed. Part 2 – Laboratory protocols, data management and analysis: will provide step-by-step protocols of the most commonly used molecular genetic techniques utilised in population genetics and systematic studies. In addition, a brief discussion and explanation of how these data are managed and analysed is also included. This manual is expected to enable NACA member country personnel to be trained to undertake molecular genetic studies in their own institutions, and as such is aimed at middle and higher level technical grades. The manual can also provide useful teaching material for specialised advanced level university courses in the region and postgraduate students. The manual has gone through two development/improvement stages. The initial material was tested at a regional workshop and at the second stage feedback from participants was used to improve the contents

    Marking Drosophila suzukii (Diptera : Drosophilidae) with fluorescent dusts

    The marking of Drosophila suzukii can be an important instrument for studying the ecology and behaviour of this economically important fruit pest, aiding the development of new Integrated Pest Management (IPM) tools or strategies. There is, however, a need for a cost-effective methodology that provides an easily detectable and stable mark. Whereas fluorescent pigment powders are often used in entomological research, the pigments (series, dyes), application techniques, or doses need to be evaluated for each studied species in terms of their efficacy and possible adverse effects on the performance of the insect. The effectiveness of different application techniques and dyes (RadGlo® TP-series) and their effect on the survival of adult D. suzukii were investigated in the laboratory. Furthermore, the influence of the marking on the behaviour of the flies was examined in laboratory trap assays (olfaction) and a field recapture study (general orientation). The persistence and detectability of the marks was evaluated both on living flies (for different application techniques) and dead flies under trapping/storage conditions. The use of fluorescent powders to mark D. suzukii flies yielded a clearly detectable and highly persistent mark, without any adverse effects on the survival and behaviour of the flies

    Acute Myeloid Leukemia

    Acute myeloid leukemia (AML) is the most common type of leukemia. The Cancer Genome Atlas Research Network has demonstrated the increasing genomic complexity of acute myeloid leukemia (AML). In addition, the network has facilitated our understanding of the molecular events leading to this deadly form of malignancy for which the prognosis has not improved over past decades. AML is a highly heterogeneous disease, and cytogenetics and molecular analysis of the various chromosome aberrations including deletions, duplications, aneuploidy, balanced reciprocal translocations and fusion of transcription factor genes and tyrosine kinases has led to better understanding and identification of subgroups of AML with different prognoses. Furthermore, molecular classification based on mRNA expression profiling has facilitated identification of novel subclasses and defined high-, poor-risk AML based on specific molecular signatures. However, despite increased understanding of AML genetics, the outcome for AML patients whose number is likely to rise as the population ages, has not changed significantly. Until it does, further investigation of the genomic complexity of the disease and advances in drug development are needed. In this review, leading AML clinicians and research investigators provide an up-to-date understanding of the molecular biology of the disease addressing advances in diagnosis, classification, prognostication and therapeutic strategies that may have significant promise and impact on overall patient survival
