    The Influence of Neuroendocrine and Genetic Markers of Stress on Cognitive Processing and Intrusive Symptoms

    This body of research investigated the influence of neuroendocrine and genetic elements of arousal on cognitive processes in the development of intrusive memories and flash-forward intrusions as related to Post-Traumatic Stress Disorder. Specifically, this thesis investigated various mechanisms that may underlie intrusive symptoms as postulated by prevalent theories of PTSD. Study 1 examined the distinctive relationship between peritraumatic dissociation and subsequent re-experiencing symptoms. Network analyses revealed strong positive edges between peritraumatic dissociation and subsequent amnesia, as well as the re-experiencing symptoms of physical reactivity to reminders, flashbacks, intrusions, and dreams, and to a lesser extent emotional numbness and hypervigilance. The finding that peritraumatic dissociation is related to subsequent re-experiencing symptoms is consistent with cognitive models that emphasize the role of dissociative experiences during a traumatic event in the etiology of PTSD re-experiencing symptoms. Study 2 aimed to determine whether peri-traumatic stress, as measured via salivary cortisol and salivary alpha-amylase, as well as pre-existing genetic polymorphisms on the FKBP5 gene increased dissociation and data-driven processing, and subsequently impacted intrusive memories related to a trauma film. The findings revealed that greater noradrenergic arousal predicted less intrusive memory distress in individuals who scored higher on data-driven processing and trait dissociation, and in FKBP5 low-risk carriers. For individuals who reported less data-driven processing and trait dissociation, and in FKBP5 high-risk carriers, as noradrenergic arousal increased, intrusive memory distress increased. This study also showed no association between data-driven processing with memory fragmentation, and fragmentation with intrusive memories. Whilst these findings support some aspect of cognitive models of PTSD as they indicate a role for data-driven processing and dissociation in intrusive symptoms, they highlight a threshold at which these variables stop moderating the relationship between arousal and intrusive memories and suggest that memory fragmentation is not related to intrusive memories. Study 3 examined the role of cognitive control in flash-forward intrusions in the context of an enduring stressor, the COVID-19 pandemic. In line with expectations, results showed that as cognitive control worsened, FKBP5 high-risk carriers reported more flash-forward distress, and low-risk carriers reported less distress. These findings are considered in the context of hippocampal changes and are consistent with emerging theories of PTSD. Lastly, study 4 sought to investigate the role of two neurological processes, pattern separation and pattern completion in intrusive memories in individuals with PTSD compared to trauma exposed controls. Consistent with existing literature, the data indicate that individuals with PTSD reported more data-driven processing, more intrusive symptoms, and demonstrated better behavioural pattern completion than trauma-exposed controls. These findings are in line with current cognitive models of PTSD, as they again indicate a role for data-driven processing in PTSD. However, study 4 found no support for the postulate that deficient pattern separation is a feature of PTSD and found an opposite effect for the role of pattern completion. Whilst these findings are inconsistent with theory, they are in line with existing experimental studies. Overall, the findings from this thesis provide insight into cognitive and biological models of PTSD and shed light on the mechanisms underlying the nature and development of intrusive symptoms

    Comparative Multiple Case Study into the Teaching of Problem-Solving Competence in Lebanese Middle Schools

    This multiple case study investigates how problem-solving competence is integrated into teaching practices in private schools in Lebanon. Its purpose is to compare instructional approaches to problem-solving across three different programs: the American (Common Core State Standards and New Generation Science Standards), French (Socle Commun de Connaissances, de Compétences et de Culture), and Lebanese with a focus on middle school (grades 7, 8, and 9). The project was conducted in nine schools equally distributed among three categories based on the programs they offered: category 1 schools offered the Lebanese program, category 2 the French and Lebanese programs, and category 3 the American and Lebanese programs. Each school was treated as a separate case. Structured observation data were collected using observation logs that focused on lesson objectives and specific cognitive problem-solving processes. The two logs were created based on a document review of the requirements for the three programs. Structured observations were followed by semi-structured interviews that were conducted to explore teachers' beliefs and understandings of problem-solving competence. The comparative analysis of within-category structured observations revealed an instruction ranging from teacher-led practices, particularly in category 1 schools, to more student-centered approaches in categories 2 and 3. The cross-category analysis showed a reliance on cognitive processes primarily promoting exploration, understanding, and demonstrating understanding, with less emphasis on planning and executing, monitoring and reflecting, thus uncovering a weakness in addressing these processes. The findings of the post-observation semi-structured interviews disclosed a range of definitions of problem-solving competence prevalent amongst teachers with clear divergences across the three school categories. This research is unique in that it compares problem-solving teaching approaches across three different programs and explores underlying teachers' beliefs and understandings of problem-solving competence in the Lebanese context. It is hoped that this project will inform curriculum developers about future directions and much-anticipated reforms of the Lebanese program and practitioners about areas that need to be addressed to further improve the teaching of problem-solving competence

    Colour technologies for content production and distribution of broadcast content

    The requirement of colour reproduction has long been a priority driving the development of new colour imaging systems that maximise human perceptual plausibility. This thesis explores machine learning algorithms for colour processing to assist both content production and distribution. First, this research studies colourisation technologies with practical use cases in restoration and processing of archived content. The research targets practical deployable solutions, developing a cost-effective pipeline which integrates the activity of the producer into the processing workflow. In particular, a fully automatic image colourisation paradigm using Conditional GANs is proposed to improve content generalisation and colourfulness of existing baselines. Moreover, a more conservative solution is considered by providing references to guide the system towards more accurate colour predictions. A fast-end-to-end architecture is proposed to improve existing exemplar-based image colourisation methods while decreasing the complexity and runtime. Finally, the proposed image-based methods are integrated into a video colourisation pipeline. A general framework is proposed to reduce the generation of temporal flickering or propagation of errors when such methods are applied frame-to-frame. The proposed model is jointly trained to stabilise the input video and to cluster their frames with the aim of learning scene-specific modes. Second, this research explored colour processing technologies for content distribution with the aim to effectively deliver the processed content to the broad audience. In particular, video compression is tackled by introducing a novel methodology for chroma intra prediction based on attention models. Although the proposed architecture helped to gain control over the reference samples and better understand the prediction process, the complexity of the underlying neural network significantly increased the encoding and decoding time. Therefore, aiming at efficient deployment within the latest video coding standards, this work also focused on the simplification of the proposed architecture to obtain a more compact and explainable model

    Real-Time Hybrid Visual Servoing of a Redundant Manipulator via Deep Reinforcement Learning

    Fixtureless assembly may be necessary in some manufacturing tasks and environ-ments due to various constraints but poses challenges for automation due to non-deterministic characteristics not favoured by traditional approaches to industrial au-tomation. Visual servoing methods of robotic control could be effective for sensitive manipulation tasks where the desired end-effector pose can be ascertained via visual cues. Visual data is complex and computationally expensive to process but deep reinforcement learning has shown promise for robotic control in vision-based manipu-lation tasks. However, these methods are rarely used in industry due to the resources and expertise required to develop application-specific systems and prohibitive train-ing costs. Training reinforcement learning models in simulated environments offers a number of benefits for the development of robust robotic control algorithms by reducing training time and costs, and providing repeatable benchmarks for which algorithms can be tested, developed and eventually deployed on real robotic control environments. In this work, we present a new simulated reinforcement learning envi-ronment for developing accurate robotic manipulation control systems in fixtureless environments. Our environment incorporates a contemporary collaborative industrial robot, the KUKA LBR iiwa, with the goal of positioning its end effector in a generic fixtureless environment based on a visual cue. Observational inputs are comprised of the robotic joint positions and velocities, as well as two cameras, whose positioning reflect hybrid visual servoing with one camera attached to the robotic end-effector, and another observing the workspace respectively. We propose a state-of-the-art deep reinforcement learning approach to solving the task environment and make prelimi-nary assessments of the efficacy of this approach to hybrid visual servoing methods for the defined problem environment. We also conduct a series of experiments ex-ploring the hyperparameter space in the proposed reinforcement learning method. Although we could not prove the efficacy of a deep reinforcement approach to solving the task environment with our initial results, we remain confident that such an ap-proach could be feasible to solving this industrial manufacturing challenge and that our contributions in this work in terms of the novel software provide a good basis for the exploration of reinforcement learning approaches to hybrid visual servoing in accurate manufacturing contexts

    On Transforming Reinforcement Learning by Transformer: The Development Trajectory

    Transformer, originally devised for natural language processing, has also attested significant success in computer vision. Thanks to its super expressive power, researchers are investigating ways to deploy transformers to reinforcement learning (RL) and the transformer-based models have manifested their potential in representative RL benchmarks. In this paper, we collect and dissect recent advances on transforming RL by transformer (transformer-based RL or TRL), in order to explore its development trajectory and future trend. We group existing developments in two categories: architecture enhancement and trajectory optimization, and examine the main applications of TRL in robotic manipulation, text-based games, navigation and autonomous driving. For architecture enhancement, these methods consider how to apply the powerful transformer structure to RL problems under the traditional RL framework, which model agents and environments much more precisely than deep RL methods, but they are still limited by the inherent defects of traditional RL algorithms, such as bootstrapping and "deadly triad". For trajectory optimization, these methods treat RL problems as sequence modeling and train a joint state-action model over entire trajectories under the behavior cloning framework, which are able to extract policies from static datasets and fully use the long-sequence modeling capability of the transformer. Given these advancements, extensions and challenges in TRL are reviewed and proposals about future direction are discussed. We hope that this survey can provide a detailed introduction to TRL and motivate future research in this rapidly developing field.Comment: 26 page

    Klarnetin staccato tekniğini güçlendirme aşamaları eser çalışmalarıyla uygulanmıştır. Staccato geçişlerini hızlandıracak ritim ve nüans çalışmalarına yer verilmiştir. Çalışmanın en önemli amacı sadece staccato çalışması değil parmak-dilin eş zamanlı uyumunun hassasiyeti üzerinde de durulmasıdır. Staccato çalışmalarını daha verimli hale getirmek için eser çalışmasının içinde etüt çalışmasına da yer verilmiştir. Çalışmaların üzerinde titizlikle durulması staccato çalışmasının ilham verici etkisi ile müzikal kimliğe yeni bir boyut kazandırmıştır. Sekiz özgün eser çalışmasının her aşaması anlatılmıştır. Her aşamanın bir sonraki performans ve tekniği güçlendirmesi esas alınmıştır. Bu çalışmada staccato tekniğinin hangi alanlarda kullanıldığı, nasıl sonuçlar elde edildiği bilgisine yer verilmiştir. Notaların parmak ve dil uyumu ile nasıl şekilleneceği ve nasıl bir çalışma disiplini içinde gerçekleşeceği planlanmıştır. Kamış-nota-diyafram-parmak-dil-nüans ve disiplin kavramlarının staccato tekniğinde ayrılmaz bir bütün olduğu saptanmıştır. Araştırmada literatür taraması yapılarak staccato ile ilgili çalışmalar taranmıştır. Tarama sonucunda klarnet tekniğin de kullanılan staccato eser çalışmasının az olduğu tespit edilmiştir. Metot taramasında da etüt çalışmasının daha çok olduğu saptanmıştır. Böylelikle klarnetin staccato tekniğini hızlandırma ve güçlendirme çalışmaları sunulmuştur. Staccato etüt çalışmaları yapılırken, araya eser çalışmasının girmesi beyni rahatlattığı ve istekliliği daha arttırdığı gözlemlenmiştir. Staccato çalışmasını yaparken doğru bir kamış seçimi üzerinde de durulmuştur. Staccato tekniğini doğru çalışmak için doğru bir kamışın dil hızını arttırdığı saptanmıştır. Doğru bir kamış seçimi kamıştan rahat ses çıkmasına bağlıdır. Kamış, dil atma gücünü vermiyorsa daha doğru bir kamış seçiminin yapılması gerekliliği vurgulanmıştır. Staccato çalışmalarında baştan sona bir eseri yorumlamak zor olabilir. Bu açıdan çalışma, verilen müzikal nüanslara uymanın, dil atış performansını rahatlattığını ortaya koymuştur. Gelecek nesillere edinilen bilgi ve birikimlerin aktarılması ve geliştirici olması teşvik edilmiştir. Çıkacak eserlerin nasıl çözüleceği, staccato tekniğinin nasıl üstesinden gelinebileceği anlatılmıştır. Staccato tekniğinin daha kısa sürede çözüme kavuşturulması amaç edinilmiştir. Parmakların yerlerini öğrettiğimiz kadar belleğimize de çalışmaların kaydedilmesi önemlidir. Gösterilen azmin ve sabrın sonucu olarak ortaya çıkan yapıt başarıyı daha da yukarı seviyelere çıkaracaktır

    Anticholinergic use in the UK: longitudinal trends and associations with cognitive outcomes

    Observational studies have shown an association between the use of anticholinergic drugs and various negative health outcomes. However, when studying cognitive outcomes, there is great heterogeneity in previous results. The objectives of the present thesis are threefold. First, to explore the longitudinal patterns of anticholinergic prescribing in the UK. Second, to examine the association between anticholinergic burden and dementia. Third, to probe the relationship between anticholinergic burden, general cognitive ability, and brain structural MRI in relatively healthy participants. Chapter 1 provides an overview of the role of acetylcholine as a neurotransmitter in the human body. It begins with a description of its molecular characteristics and continues with a summary of anatomical and cellular features of cholinergic pathways in the brain. The chapter concludes with a description of the relevance of cholinergic processing in cognition and Alzheimer’s disease. Chapter 2 gives a summary of anticholinergic drugs. It describes the history of anticholinergic compounds and their present use in medicine. It then appraises the tools used to gauge the anticholinergic potency of drugs. I conclude the Chapter by evaluating the available evidence on the effects of anticholinergic drugs on various important health outcomes. Chapter 3 focuses on UK Biobank, the sample used in all analyses presented in this thesis. The chapter briefly describes the conception of the study, the timeline of assessments, and the available variables. I focus in my descriptions on the variables that were used in the present thesis, especially cognitive tests, brain imaging, and linked health data. Chapters 4 to 6 present the empirical work conducted as part of this thesis. Chapter 4 presents an analysis of anticholinergic prescribing trends in UK primary care from the year 1990 to 2015. I first calculate an anticholinergic burden (AChB) according to 13 different anticholinergic scales and an average to derive a “Meta-scale”. I then describe the prevalence of anticholinergic prescribing and its longitudinal trend for all scales. I use different plots of age-, period- and cohort effects on the AChB according to the Meta-scale to evaluate the contributions of these effects to the linear longitudinal trend. The study finds AChB to have increased 9-fold over 25 years and that this effect was attributable to both age- and cohort/period-related changes. In other words, ageing of the sample is not sufficient to explain the increase in anticholinergic prescribing; cohort- or period-effects must have contributed to the observed changes. Chapter 5 explores the relationship between anticholinergic prescribing and dementia. Previous studies on this topic had provided varied results. One of the goals of the present study was to probe potential factors for this heterogeneity. We find that greater AChB according to most of the studied anticholinergic scales (standardised HRs range: 1.027-1.125), as well as the slope of anticholinergic change (HR=1.094; 95% CI: 1.068-1.119), are associated with dementia. However, we find that not all drug classes are associated with dementia. Antidepressants (HR=1.11, 95% CI=1.07-1.17), antiepileptics (HR=1.07, 95% CI=1.04-1.11), and the antidiuretic furosemide (HR=1.06, 95% CI=1.02-1.10) exhibit the strongest effects. Interestingly, when exploring the effects of groups of anticholinergic drugs with different anticholinergic potencies, only the moderate potency group shows significant associations with dementia (HR=1.10, 95% CI=1.05-1.15). Chapter 6 examines the association between AChB, general cognitive ability, and brain structural MRI. It aims both to explore the potential sources of heterogeneity in previous work, as well as to expand on it by studying relatively healthy community-dwelling adults. We study brain structural MRI in a much bigger sample (at least 5x bigger) and use many more outcomes than previous studies. We find weak, but significant associations between AChB and general cognitive ability, and with 7/9 individual cognitive tests (standardised betas (β) range: -0.039, -0.003). Again, AChB in only some drug classes is associated with lower general cognitive ability, especially β-lactam antibiotics (β=-0.035, pFDR. Finally, chapter 7 summarizes the findings presented in chapters 4 to 6. The chapter also provides a critique of the sample and of my approach when conducting the analyses presented in the present thesis. The chapter concludes by discussing suggestions for future work on this topic
