    Infection-generated electric field in gut epithelium drives bidirectional migration of macrophages.

    Many bacterial pathogens hijack macrophages to egress from the port of entry to the lymphatic drainage and/or bloodstream, causing dissemination of life-threatening infections. However, the underlying mechanisms are not well understood. Here, we report that Salmonella infection generates directional electric fields (EFs) in the follicle-associated epithelium of mouse cecum. In vitro application of an EF, mimicking the infection-generated electric field (IGEF), induces directional migration of primary mouse macrophages to the anode, which is reversed to the cathode upon Salmonella infection. This infection-dependent directional switch is independent of the Salmonella pathogenicity island 1 (SPI-1) type III secretion system. The switch is accompanied by a reduction of sialic acids on glycosylated surface components during phagocytosis of bacteria, which is absent in macrophages challenged by microspheres. Moreover, enzymatic cleavage of terminally exposed sialic acids reduces macrophage surface negativity and severely impairs directional migration of macrophages in response to an EF. Based on these findings, we propose that macrophages are attracted to the site of infection by a combination of chemotaxis and galvanotaxis; after phagocytosis of bacteria, surface electrical properties of the macrophage change, and galvanotaxis directs the cells away from the site of infection

    The biogenesis pathway of tRNA-derived piRNAs in Bombyx germ cells.

    Transfer RNAs (tRNAs) function in translational machinery and further serves as a source of short non-coding RNAs (ncRNAs). tRNA-derived ncRNAs show differential expression profiles and play roles in many biological processes beyond translation. Molecular mechanisms that shape and regulate their expression profiles are largely unknown. Here, we report the mechanism of biogenesis for tRNA-derived Piwi-interacting RNAs (td-piRNAs) expressed in Bombyx BmN4 cells. In the cells, two cytoplasmic tRNA species, tRNAAspGUC and tRNAHisGUG, served as major sources for td-piRNAs, which were derived from the 5\u27-part of the respective tRNAs. cP-RNA-seq identified the two tRNAs as major substrates for the 5\u27-tRNA halves as well, suggesting a previously uncharacterized link between 5\u27-tRNA halves and td-piRNAs. An increase in levels of the 5\u27-tRNA halves, induced by BmNSun2 knockdown, enhanced the td-piRNA expression levels without quantitative change in mature tRNAs, indicating that 5\u27-tRNA halves, not mature tRNAs, are the direct precursors for td-piRNAs. For the generation of tRNAHisGUG-derived piRNAs, BmThg1l-mediated nucleotide addition to -1 position of tRNAHisGUG was required, revealing an important function of BmThg1l in piRNA biogenesis. Our study advances the understanding of biogenesis mechanisms and the genesis of specific expression profiles for tRNA-derived ncRNAs

    The student review of the science curriculum

    This paper presents some of the main findings of a student-led review of the science curriculum in England. Over 350 students, aged 16-19, designed possible questions for a web-based questionnaire at regional meetings held across England. In the six weeks to 8th February 2002, a total of 1,493 questionnaires were submitted. Analysis of these responses shows considerable student dissatisfaction with the science curriculum. A list of 10 student recommendations is provided

    Characterization of Exopolysaccharides Produced by Bacillus cereus and Brachybacterium sp. Isolated from Asian Sea Bass (Lates calcarifer)

    Aims: EPS extracted from marine bacteria, which associated with Asian sea bass has potential antimicrobial activities.Methodology and Results: Two marine Bacteria were isolated from Asian sea bass (Lates calcarifer) obtained from aquaculture farm, located at Johor bahru Malaysia. 16S rRNA analysis for bacteria identity revealed that bacteria ors1 had 99 % identity to Bacillus cereus and ors2 had 96 % identity with Brachybacterium sp. All bacteria shared many similarities and variation in terms of biochemical reactions and microscopic observation. Exopolysaccharides (EPSs) were extracted and purified from bacteria as they produced mucous colonies. Average analysis of EPS components showed 50 % carbohydrates, 26 % protein and 24 % fatty acids. The FTIR analysis confirmed the functional groups of the EPS. Screening for antimicrobial activities assays using Kirby-Bauer methods against both grams positive and negative had shown presence of inhibition zones.Conclusion, significance and impact of study: This study recommends that bacteria isolated from Asian sea bass are having antimicrobial activities and could be used as a potential source for the development of marine drugs

    Is game theory meant to describe actual choices by people and institutions or not? It is remarkable how much game theory has been done while largely ignoring this question. The seminal book by von Neumann and Morgenstern, The Theory of Games and Economic Behavior, was clearly about how rational players would play against others they knew were rational. In more recent work, game theorists are not always explicit about what they aim to describe or advise. At one extreme, highly mathematical analyses have proposed rationality requirements that people and firms are probably not smart enough to satisfy in everyday decisions. At the other extreme, adaptive and evolutionary approaches use very simple models-mostly developed to describe nonhuman animals-in which players may not realize they are playing a game at all. When game theory does aim to describe behavior, it often proceeds with a disturbingly low ratio of careful observation to theorizing

    5\u27-Terminal nucleotide variations in human cytoplasmic tRNAHisGUG and its 5\u27-halves.

    Transfer RNAs (tRNAs) are fundamental adapter components of translational machinery. tRNAs can further serve as a source of tRNA-derived noncoding RNAs that play important roles in various biological processes beyond translation. Among all species of tRNAs, tRNA(HisGUG) has been known to uniquely contain an additional guanosine residue at the -1 position (G-1) of its 5\u27-end. To analyze this -1 nucleotide in detail, we developed a TaqMan qRT-PCR method that can distinctively quantify human mature cytoplasmic tRNA(HisGUG) containing G-1, U-1, A-1, or C-1 or lacking the -1 nucleotide (starting from G1). Application of this method to the mature tRNA fraction of BT-474 breast cancer cells revealed the presence of tRNA(HisGUG) containing U-1 as well as the one containing G-1 Moreover, tRNA lacking the -1 nucleotide was also detected, thus indicating the heterogeneous expression of 5\u27-tRNA(HisGUG) variants. A sequence library of sex hormone-induced 5\u27-tRNA halves (5\u27-SHOT-RNAs), identified via cP-RNA-seq of a BT-474 small RNA fraction, also demonstrated the expression of 5\u27-tRNA(HisGUG) halves containing G-1, U-1, or G1 as 5\u27-terminal nucleotides. Although the detected 5\u27-nucleotide species were identical, the relative abundances differed widely between mature tRNA and 5\u27-half from the same BT-474 cells. The majority of mature tRNAs contained the -1 nucleotide, whereas the majority of 5\u27-halves lacked this nucleotide, which was biochemically confirmed using a primer extension assay. These results reveal the novel identities of tRNA(HisGUG) molecules and provide insights into tRNA(HisGUG) maturation and the regulation of tRNA half production
