Towards Autonomous Selective Harvesting: A Review of Robot Perception, Robot Design, Motion Planning and Control
This paper provides an overview of the current state-of-the-art in selective
harvesting robots (SHRs) and their potential for addressing the challenges of
global food production. SHRs have the potential to increase productivity,
reduce labour costs, and minimise food waste by selectively harvesting only
ripe fruits and vegetables. The paper discusses the main components of SHRs,
including perception, grasping, cutting, motion planning, and control. It also
highlights the challenges in developing SHR technologies, particularly in the
areas of robot design, motion planning and control. The paper also discusses
the potential benefits of integrating AI and soft robots and data-driven
methods to enhance the performance and robustness of SHR systems. Finally, the
paper identifies several open research questions in the field and highlights
the need for further research and development efforts to advance SHR
technologies to meet the challenges of global food production. Overall, this
paper provides a starting point for researchers and practitioners interested in

Computertomographie-basierte Bestimmung von Aortenklappenkalk und seine Assoziation mit Komplikationen nach interventioneller Aortenklappenimplantation (TAVI)
Background: Severe aortic valve calcification (AVC) has generally been recognized as a key factor in the occurrence of adverse events after transcatheter aortic valve implantation (TAVI). To date, however, a consensus on a standardized calcium detection threshold for aortic valve calcium quantification in contrast-enhanced computed tomography angiography (CTA) is still lacking. The present thesis aimed at comparing two different approaches for quantifying AVC in CTA scans based on their predictive power for adverse events and survival after a TAVI procedure.
Methods: The extensive dataset of this study included 198 characteristics for each of the 965 prospectively included patients who had undergone TAVI between November 2012 and December 2019 at the German Heart Center Berlin (DHZB). AVC quantification in CTA scans was performed at a fixed Hounsfield Unit (HU) threshold of 850 HU (HU 850 approach) and at a patient-specific threshold, where the HU threshold was set by multiplying the mean luminal attenuation of the ascending aorta by 2 (+100 % HUAorta approach). The primary endpoint of this study consisted of a combination of post-TAVI outcomes (paravalvular leak ≥ mild, implant-related conduction disturbances, 30-day mortality, post-procedural stroke, annulus rupture, and device migration). The Akaike information criterion was used to select variables for the multivariable regression model. Multivariable analysis was carried out to determine the predictive power of the investigated approaches.
Results: Multivariable analyses showed that a fixed threshold of 850 HU (calcium volume cut-off 146 mm3) was unable to predict the composite clinical endpoint post-TAVI (OR=1.13, 95 % CI 0.87 to 1.48, p=0.35). In contrast, the +100 % HUAorta approach (calcium volume cut-off 1421 mm3) enabled independent prediction of the composite clinical endpoint post-TAVI (OR=2, 95 % CI 1.52 to 2.64, p=9.2x10-7). No significant difference in the Kaplan-Meier survival analysis was observed for either of the approaches.
Conclusions: The patient-specific calcium detection threshold +100 % HUAorta is more predictive of post-TAVI adverse events included in the combined clinical endpoint than the fixed HU 850 approach. For the +100 % HUAorta approach, a calcium volume cut-off of 1421 mm3 of the aortic valve had the highest predictive value.Hintergrund: Ein wichtiger Auslöser von Komplikationen nach einer Transkatheter-Aortenklappen-Implantation (TAVI) sind ausgeprägte Kalkablagerung an der Aortenklappe. Dennoch erfolgte bisher keine Einigung auf ein standardisiertes Messverfahren zur Quantifizierung der Kalklast der Aortenklappe in einer kontrastverstärkten dynamischen computertomographischen Angiographie (CTA). Die vorliegende Dissertation untersucht, inwieweit die Wahl des Analyseverfahrens zur Quantifizierung von Kalkablagerungen in der Aortenklappe die Prognose von Komplikationen und der Überlebensdauer nach einer TAVI beeinflusst.
Methodik: Der Untersuchung liegt ein umfangreicher Datensatz von 965 Patienten mit 198 Merkmalen pro Patienten zugrunde, welche sich zwischen 2012 und 2019 am Deutschen Herzzentrum Berlin einer TAVI unterzogen haben. Die Quantifizierung der Kalkablagerung an der Aortenklappe mittels CTA wurde einerseits mit einem starren Grenzwert von 850 Hounsfield Einheiten (HU) (HU 850 Verfahren) und andererseits anhand eines individuellen Grenzwertes bemessen. Letzterer ergibt sich aus der HU-Dämpfung in dem Lumen der Aorta ascendens multipliziert mit 2 (+100 % HUAorta Verfahren). Der primäre klinische Endpunkt dieser Dissertation besteht aus einem aus sechs Variablen zusammengesetzten klinischen Endpunkt, welcher ungewünschte Ereignisse nach einer TAVI abbildet (paravalvuläre Leckage ≥mild, Herzrhythmusstörungen nach einer TAVI, Tod innerhalb von 30 Tagen, post-TAVI Schlaganfall, Ruptur des Annulus und Prothesendislokation). Mögliche Störfaktoren, die auf das Eintreten der Komplikationen nach TAVI Einfluss haben, wurden durch den Einsatz des Akaike Informationskriterium ermittelt. Um die Vorhersagekraft von Komplikationen nach einer TAVI durch beide Verfahren zu ermitteln, wurde eine multivariate Regressionsanalyse durchgeführt.
Ergebnisse: Die multivariaten logistischen Regressionen zeigen, dass die Messung der Kalkablagerungen anhand der HU 850 Messung (Kalklast Grenzwert von 146 mm3) die Komplikationen und die Überlebensdauer nicht vorhersagen konnten (OR=1.13, 95 % CI 0.87 bis 1.48, p=0.35). Die nach dem +100 % HUAorta Verfahren (Kalklast Grenzwert von 1421 mm3) individualisierte Kalkmessung erwies sich hingegen als sehr aussagekräftig, da hiermit Komplikationen nach einer TAVI signifikant vorhergesagt werden konnten (OR=2, 95 % CI 1.52 bis 2.64, p=9.2x10-7). In Hinblick auf die postoperative Kaplan-Meier Überlebenszeitanalyse kann auch mit dem +100 % HUAorta Verfahren keine Vorhersage getroffen werden.
Fazit: Aus der Dissertation ergibt sich die Empfehlung, die Messung von Kalkablagerungen nach dem +100 % HUAorta Verfahren vorzunehmen, da Komplikationen wesentlich besser und zuverlässiger als nach der gängigen HU 850 Messmethode vorhergesagt werden können. Für das +100 % HUAorta Verfahren lag der optimale Kalklast Grenzwert bei 1421 mm3
Gasificação direta de biomassa para produção de gás combustível
The excessive consumption of fossil fuels to satisfy the world necessities of
energy and commodities led to the emission of large amounts of greenhouse
gases in the last decades, contributing significantly to the greatest
environmental threat of the 21st century: Climate Change. The answer to this
man-made disaster is not simple and can only be made if distinct stakeholders
and governments are brought to cooperate and work together. This is
mandatory if we want to change our economy to one more sustainable and
based in renewable materials, and whose energy is provided by the eternal
nature energies (e.g., wind, solar). In this regard, biomass can have a main role
as an adjustable and renewable feedstock that allows the replacement of fossil
fuels in various applications, and the conversion by gasification allows the
necessary flexibility for that purpose. In fact, fossil fuels are just biomass that
underwent extreme pressures and heat for millions of years. Furthermore,
biomass is a resource that, if not used or managed, increases wildfire risks.
Consequently, we also have the obligation of valorizing and using this
resource.
In this work, it was obtained new scientific knowledge to support the
development of direct (air) gasification of biomass in bubbling fluidized bed
reactors to obtain a fuel gas with suitable properties to replace natural gas in
industrial gas burners. This is the first step for the integration and development
of gasification-based biorefineries, which will produce a diverse number of
value-added products from biomass and compete with current petrochemical
refineries in the future. In this regard, solutions for the improvement of the raw
producer gas quality and process efficiency parameters were defined and
analyzed. First, addition of superheated steam as primary measure allowed the
increase of H2 concentration and H2/CO molar ratio in the producer gas without
compromising the stability of the process. However, the measure mainly
showed potential for the direct (air) gasification of high-density biomass (e.g.,
pellets), due to the necessity of having char accumulation in the reactor bottom
bed for char-steam reforming reactions. Secondly, addition of refused derived
fuel to the biomass feedstock led to enhanced gasification products, revealing
itself as a highly promising strategy in terms of economic viability and
environmental benefits of future gasification-based biorefineries, due to the
high availability and low costs of wastes. Nevertheless, integrated techno economic and life cycle analyses must be performed to fully characterize the
process. Thirdly, application of low-cost catalyst as primary measure revealed
potential by allowing the improvement of the producer gas quality (e.g., H2 and
CO concentration, lower heating value) and process efficiency parameters with
distinct solid materials; particularly, the application of concrete, synthetic
fayalite and wood pellets chars, showed promising results. Finally, the
economic viability of the integration of direct (air) biomass gasification
processes in the pulp and paper industry was also shown, despite still lacking
interest to potential investors. In this context, the role of government policies
and appropriate economic instruments are of major relevance to increase the
implementation of these projects.O consumo excessivo de combustíveis fósseis para garantir as necessidades e

gases com efeito de estufa nas últimas décadas, contribuindo
significativamente para a maior ameaça ambiental do século XXI: Alterações
Climáticas. A solução para este desastre de origem humana é de caráter
complexo e só pode ser atingida através da cooperação de todos os governos
e partes interessadas. Para isto, é obrigatória a criação de uma bioeconomia
como base de um futuro mais sustentável, cujas necessidades energéticas e
materiais sejam garantidas pelas eternas energias da natureza (e.g., vento,
sol). Neste sentido, a biomassa pode ter um papel principal como uma matéria prima ajustável e renovável que permite a substituição de combustíveis fósseis
num variado número de aplicações, e a sua conversão através da gasificação
pode ser a chave para este propósito. Afinal, na prática, os combustíveis
fósseis são apenas biomassa sujeita a elevada temperatura e pressão durante
milhões de anos. Além do mais, a gestão eficaz da biomassa é fundamental
para a redução dos riscos de incêndio florestal e, como tal, temos o dever de
utilizar e valorizar este recurso.
Neste trabalho, foi obtido novo conhecimento científico para suporte do
desenvolvimento das tecnologias de gasificação direta (ar) de biomassa em
leitos fluidizados borbulhantes para produção de gás combustível, com o
objetivo da substituição de gás natural em queimadores industriais. Este é o
primeiro passo para o desenvolvimento de biorrefinarias de gasificação, uma
potencial futura indústria que irá providenciar um variado número de produtos
de valor acrescentado através da biomassa e competir com a atual indústria
petroquímica. Neste sentido, foram analisadas várias medidas para a melhoria
da qualidade do gás produto bruto e dos parâmetros de eficiência do processo.
Em primeiro, a adição de vapor sobreaquecido como medida primária permitiu
o aumento da concentração de H2 e da razão molar H2/CO no gás produto sem
comprometer a estabilidade do processo. No entanto, esta medida somente
revelou potencial para a gasificação direta (ar) de biomassa de alta densidade
(e.g., pellets) devido à necessidade da acumulação de carbonizados no leito
do reator para a ocorrência de reações de reforma com vapor. Em segundo, a
mistura de combustíveis derivados de resíduos e biomassa residual florestal
permitiu a melhoria dos produtos de gasificação, constituindo desta forma uma
estratégia bastante promissora a nível económico e ambiental, devido à
elevada abundância e baixo custo dos resíduos urbanos. Contudo, devem ser
efetuadas análises técnico-económicas e de ciclo de vida para a completa
caraterização do processo. Em terceiro, a aplicação de catalisadores de baixo
custo como medida primária demonstrou elevado potencial para a melhoria do
gás produto (e.g., concentração de H2 e CO, poder calorífico inferior) e para o
incremento dos parâmetros de eficiência do processo; em particular, a
aplicação de betão, faialite sintética e carbonizados de pellets de madeira,
demonstrou resultados promissores. Finalmente, foi demonstrada a viabilidade
económica da integração do processo de gasificação direta (ar) de biomassa
na indústria da pasta e papel, apesar dos parâmetros determinados não serem
atrativos para potenciais investidores. Neste contexto, a intervenção dos
governos e o desenvolvimento de instrumentos de apoio económico é de

Cost-effective non-destructive testing of biomedical components fabricated using additive manufacturing
Biocompatible titanium-alloys can be used to fabricate patient-specific medical components using additive manufacturing (AM). These novel components have the potential to improve clinical outcomes in various medical scenarios. However, AM introduces stability and repeatability concerns, which are potential roadblocks for its widespread use in the medical sector. Micro-CT imaging for non-destructive testing (NDT) is an effective solution for post-manufacturing quality control of these components. Unfortunately, current micro-CT NDT scanners require expensive infrastructure and hardware, which translates into prohibitively expensive routine NDT. Furthermore, the limited dynamic-range of these scanners can cause severe image artifacts that may compromise the diagnostic value of the non-destructive test. Finally, the cone-beam geometry of these scanners makes them susceptible to the adverse effects of scattered radiation, which is another source of artifacts in micro-CT imaging.
In this work, we describe the design, fabrication, and implementation of a dedicated, cost-effective micro-CT scanner for NDT of AM-fabricated biomedical components. Our scanner reduces the limitations of costly image-based NDT by optimizing the scanner\u27s geometry and the image acquisition hardware (i.e., X-ray source and detector). Additionally, we describe two novel techniques to reduce image artifacts caused by photon-starvation and scatter radiation in cone-beam micro-CT imaging.
Our cost-effective scanner was designed to match the image requirements of medium-size titanium-alloy medical components. We optimized the image acquisition hardware by using an 80 kVp low-cost portable X-ray unit and developing a low-cost lens-coupled X-ray detector. Image artifacts caused by photon-starvation were reduced by implementing dual-exposure high-dynamic-range radiography. For scatter mitigation, we describe the design, manufacturing, and testing of a large-area, highly-focused, two-dimensional, anti-scatter grid.
Our results demonstrate that cost-effective NDT using low-cost equipment is feasible for medium-sized, titanium-alloy, AM-fabricated medical components. Our proposed high-dynamic-range strategy improved by 37% the penetration capabilities of an 80 kVp micro-CT imaging system for a total x-ray path length of 19.8 mm. Finally, our novel anti-scatter grid provided a 65% improvement in CT number accuracy and a 48% improvement in low-contrast visualization. DEEP REINFORCEMENT LEARNING AND MODEL PREDICTIVE CONTROL APPROACHES FOR THE SCHEDULED OPERATION OF DOMESTIC REFRIGERATORS
Excess capacity of the UK’s national grid is widely quoted to be reducing to around 4% over the coming years as a consequence of increased economic growth (and hence power usage) and reductions in power generation plants. There is concern that short term variations in power demand could lead to serious wide-scale disruption on a national scale. This is therefore spawning greater attention on augmenting traditional generation plants with renewable and localized energy storage technologies, and consideration of improved demand side responses (DSR), where power consumers are incentivized to switch off assets when the grid is under pressure. It is estimated, for instance, that refrigeration/HVAC systems alone could account for ~14% of the total UK energy usage, with refrigeration and water heating/cooling systems, in particular, being able to act as real-time ‘buffer’ technologies that can be demand-managed to accommodate transient demands by being switched-off for short periods without damaging their outputs. Large populations of thermostatically controlled loads (TCLs) hold significant potential for performing ancillary services in power systems since they are well-established and widely distributed around the power network. In the domestic sector, refrigerators and freezers collectively constitute a very large electrical load since they are continuously connected and are present in almost most households. The rapid proliferation of the ‘Internet of Things’ (IoT) now affords the opportunity to monitor and visualise smart buildings appliances performance and specifically, schedule the operation of the widely distributed domestic refrigerator and freezers to collectively improve energy efficiency and reduce peak power consumption on the electrical grid. To accomplish this, this research proposes the real-time estimation of the thermal mass of individual refrigerators in a network using on-line parameter identification, and the co-ordinated (ON-OFF) scheduling of the refrigerator compressors to maintain their respective temperatures within specified hysteresis bands—commensurate with accommodating food safety standards. Custom Model Predictive Control (MPC) schemes and a Machine Learning algorithm (Reinforcement Learning) are researched to realize an appropriate scheduling methodology which is implemented through COTS IoT hardware. Benefits afforded by the proposed schemes are investigated through experimental trials which show that the co-ordinated operation of domestic refrigerators can 1) reduce the peak power consumption as seen from the perspective of the electrical power grid (i.e. peak power shaving), 2) can adaptively control the temperature hysteresis band of individual refrigerators to increase operational efficiency, and 3) contribute to a widely distributed aggregated load shed for Demand Side Response purposes in order to aid grid stability. Comparative studies of measurements from experimental trials show that the co-ordinated scheduling of refrigerators allows energy savings of between 19% and 29% compared to their traditional isolated (non-co-operative) operation. Moreover, by adaptively changing the hysteresis bands of individual fridges in response to changes in thermal behaviour, a further 20% of savings in energy are possible at local refrigerator level, thereby providing benefits to both network supplier and individual consumer
Novel Cardiac Mapping Approaches and Multimodal Techniques to Unravel Multidomain Dynamics of Complex Arrhythmias Towards a Framework for Translational Mechanistic-Based Therapeutic Strategies
[ES] Las arritmias cardíacas son un problema importante para los sistemas de salud en el mundo desarrollado debido a su alta incidencia y prevalencia a medida que la población envejece. La fibrilación auricular (FA) y la fibrilación ventricular (FV) se encuentran entre las arritmias más complejas observadas en la práctica clínica. Las consecuencias clínicas de tales alteraciones arrítmicas incluyen el desarrollo de eventos cardioembólicos complejos en la FA, y repercusiones dramáticas debido a procesos fibrilatorios sostenidos que amenazan la vida infringiendo daño neurológico tras paro cardíaco por FV, y que pueden provocar la muerte súbita cardíaca (MSC). Sin embargo, a pesar de los avances tecnológicos de las últimas décadas, sus mecanismos intrínsecos se comprenden de forma incompleta y, hasta la fecha, las estrategias terapéuticas carecen de una base mecanicista suficiente y poseen bajas tasas de éxito.
Entre los mecanismos implicados en la inducción y perpetuación de arritmias cardíacas, como la FA, se cree que las dinámicas de las fuentes focales y reentrantes de alta frecuencia, en sus diferentes modalidades, son las fuentes primarias que mantienen la arritmia. Sin embargo, se sabe poco sobre los atractores, así como, de la dinámica espacio-temporal de tales fuentes fibrilatorias primarias, específicamente, las fuentes focales o rotacionales dominantes que mantienen la arritmia. Por ello, se ha desarrollado una plataforma computacional, para comprender los factores (activos, pasivos y estructurales) determinantes, y moduladores de dicha dinámica. Esto ha permitido establecer un marco para comprender la compleja dinámica de los rotores con énfasis en sus propiedades deterministas para desarrollar herramientas basadas en los mecanismos para ayuda diagnóstica y terapéutica.
Comprender los procesos fibrilatorios es clave para desarrollar marcadores y herramientas fisiológica- y clínicamente relevantes para la ayuda de diagnóstico temprano. Específicamente, las propiedades espectrales y de tiempo-frecuencia de los procesos fibrilatorios han demostrado resaltar el comportamiento determinista principal de los mecanismos intrínsecos subyacentes a las arritmias y el impacto de tales eventos arrítmicos. Esto es especialmente relevante para determinar el pronóstico temprano de los supervivientes comatosos después de un paro cardíaco debido a fibrilación ventricular (FV).
Las técnicas de mapeo electrofisiológico, el mapeo eléctrico y óptico cardíaco, han demostrado ser recursos muy valiosos para dar forma a nuevas hipótesis y desarrollar nuevos enfoques mecanicistas y estrategias terapéuticas mejoradas. Esta tecnología permite además el trabajo multidisciplinar entre clínicos y bioingenieros, para el desarrollo y validación de dispositivos y metodologías para identificar biomarcadores multi-dominio que permitan rastrear con precisión la dinámica de las arritmias identificando fuentes dominantes y atractores con alta precisión para ser dianas de estrategias terapeúticas innovadoras. Es por ello que uno de los objetivos fundamentales ha sido la implantación y validación de nuevos sistemas de mapeo en distintas configuraciones que sirvan de plataforma de desarrollo de nuevas estrategias terapeúticas. Aunque el mapeo panorámico es el método principal y más completo para rastrear simultáneamente biomarcadores electrofisiológicos, su adopción por la comunidad científica es limitada principalmente debido al coste elevado de la tecnología. Aprovechando los avances tecnológicos recientes, nos hemos enfocado en desarrollar, y validar, sistemas de mapeo óptico de alta resolución para registro panorámico cardíaco, utilizando modelos clínicamente relevantes para la investigación básica y la bioingeniería.[CA] Les arítmies cardíaques són un problema important per als sistemes de salut del món desenvolupat a causa de la seva alta incidència i prevalença a mesura que la població envelleix. La fibril·lació auricular (FA) i la fibril·lació ventricular (FV), es troben entre les arítmies més complexes observades a la pràctica clínica. Les conseqüències clíniques d'aquests trastorns arítmics inclouen el desenvolupament d'esdeveniments cardioembòlics complexos en FA i repercussions dramàtiques a causa de processos fibril·latoris sostinguts que posen en perill la vida amb danys neurològics posteriors a la FV, que condueixen a una aturada cardíaca i a la mort cardíaca sobtada (SCD). Tanmateix, malgrat els avanços tecnològics de les darreres dècades, els seus mecanismes intrínsecs s'entenen de forma incompleta i, fins a la data, les estratègies terapèutiques no tenen una base mecanicista suficient i tenen baixes taxes d'èxit.
La majoria dels avenços en el desenvolupament de biomarcadors òptims i noves estratègies terapèutiques en aquest camp provenen de tècniques valuoses en la investigació de mecanismes d'arítmia. Entre els mecanismes implicats en la inducció i perpetuació de les arítmies cardíaques, es creu que les fonts primàries subjacents a l'arítmia són les fonts focals reingressants d'alta freqüència dinàmica i AF, en les seves diferents modalitats. Tot i això, se sap poc sobre els atractors i la dinàmica espaciotemporal d'aquestes fonts primàries fibril·ladores, específicament les fonts rotacionals o focals dominants que mantenen l'arítmia. Per tant, s'ha desenvolupat una plataforma computacional per entendre determinants actius, passius, estructurals i moduladors d'aquestes dinàmiques. Això va permetre establir un marc per entendre la complexa dinàmica multidomini dels rotors amb ènfasi en les seves propietats deterministes per desenvolupar enfocaments mecanicistes per a l'ajuda i la teràpia diagnòstiques.
La comprensió dels processos fibril·latoris és clau per desenvolupar puntuacions i eines rellevants fisiològicament i clínicament per ajudar al diagnòstic precoç. Concretament, les propietats espectrals i de temps-freqüència dels processos fibril·latoris han demostrat destacar un comportament determinista important dels mecanismes intrínsecs subjacents a les arítmies i l'impacte d'aquests esdeveniments arítmics. Mitjançant coneixements previs, processament de senyals, tècniques d'aprenentatge automàtic i anàlisi de dades, es va desenvolupar una puntuació de risc mecanicista a la aturada cardíaca per FV.
Les tècniques de cartografia òptica cardíaca i electrofisiològica han demostrat ser recursos inestimables per donar forma a noves hipòtesis i desenvolupar nous enfocaments mecanicistes i estratègies terapèutiques. Aquesta tecnologia ha permès durant molts anys provar noves estratègies terapèutiques farmacològiques o ablatives i desenvolupar mètodes multidominis per fer un seguiment precís de la dinàmica d'arrímies que identifica fonts i atractors dominants. Tot i que el mapatge panoràmic és el mètode principal per al seguiment simultani de paràmetres electrofisiològics, la seva adopció per part de la comunitat multidisciplinària d'investigació cardiovascular està limitada principalment pel cost de la tecnologia. Aprofitant els avenços tecnològics recents, ens centrem en el desenvolupament i la validació de sistemes de mapes òptics de baix cost per a imatges panoràmiques mitjançant models clínicament rellevants per a la investigació bàsica i la bioenginyeria.[EN] Cardiac arrhythmias are a major problem for health systems in the developed world due to their high incidence and prevalence as the population ages. Atrial fibrillation (AF) and ventricular fibrillation (VF), are amongst the most complex arrhythmias seen in the clinical practice. Clinical consequences of such arrhythmic disturbances include developing complex cardio-embolic events in AF, and dramatic repercussions due to sustained life-threatening fibrillatory processes with subsequent neurological damage under VF, leading to cardiac arrest and sudden cardiac death (SCD). However, despite the technological advances in the last decades, their intrinsic mechanisms are incompletely understood, and, to date, therapeutic strategies lack of sufficient mechanistic basis and have low success rates.
Most of the progress for developing optimal biomarkers and novel therapeutic strategies in this field has come from valuable techniques in the research of arrhythmia mechanisms. Amongst the mechanisms involved in the induction and perpetuation of cardiac arrhythmias such AF, dynamic high-frequency re-entrant and focal sources, in its different modalities, are thought to be the primary sources underlying the arrhythmia. However, little is known about the attractors and spatiotemporal dynamics of such fibrillatory primary sources, specifically dominant rotational or focal sources maintaining the arrhythmia. Therefore, a computational platform for understanding active, passive and structural determinants, and modulators of such dynamics was developed. This allowed stablishing a framework for understanding the complex multidomain dynamics of rotors with enphasis in their deterministic properties to develop mechanistic approaches for diagnostic aid and therapy.
Understanding fibrillatory processes is key to develop physiologically and clinically relevant scores and tools for early diagnostic aid. Specifically, spectral and time-frequency properties of fibrillatory processes have shown to highlight major deterministic behaviour of intrinsic mechanisms underlying the arrhythmias and the impact of such arrhythmic events. Using prior knowledge, signal processing, machine learning techniques and data analytics, we aimed at developing a reliable mechanistic risk-score for comatose survivors of cardiac arrest due to VF.
Cardiac optical mapping and electrophysiological mapping techniques have shown to be unvaluable resources to shape new hypotheses and develop novel mechanistic approaches and therapeutic strategies. This technology has allowed for many years testing new pharmacological or ablative therapeutic strategies, and developing multidomain methods to accurately track arrhymia dynamics identigying dominant sources and attractors. Even though, panoramic mapping is the primary method for simultaneously tracking electrophysiological parameters, its adoption by the multidisciplinary cardiovascular research community is limited mainly due to the cost of the technology. Taking advantage of recent technological advances, we focus on developing and validating low-cost optical mapping systems for panoramic imaging using clinically relevant models for basic research and bioengineering.Calvo Saiz, CJ. Biorefinaria marinha: valorização de algas para produtos de valor acrescentado
The consumption paradigm is changing. Consumers are more
attentive to what they consume and to the sustainability questions
addressed to those products. This change was driven by the
healthy label given to natural products but also by the urgent need
to find more sustainable sources of raw materials and to
implement processes as a way to mitigate climate changes,
without compromising the quality and affecting the price of the
final product. Pigments are compounds that can be obtained from
various sources of biomass, being recognized by their numerous
biological activities. Due to their spectral and biological properties,
pigments can find application in many fields, from food to
cosmetics, nutraceutics, and medicine. However, conventional
methodologies to obtain pigments demand very often the use of
costly equipment and/or toxic organic solvents. In this work,
alternative methodologies to extract and purify pigments from
algae of were developed. Simple and efficient methodologies
mostly based on aqueous solvents were used to recover one or
more pigments from algae within a blue biorefinery framework. In
all chapters, a variety of solvents were screened, and operational
conditions were studied and optimized in order to reach high
yields of extractions and/or purities. Aqueous solutions of
tensioactive ionic liquids have shown great potential for the
recovery of hydrophobic pigments such as chlorophylls and
carotenoids. In the other hand, induced precipitation of proteins is
shown to be a good alternative for the selective precipitation (and
further purification) of phycobiliproteins. Economic and
environmental impacts were assessed and encouraging results
were obtained from both perspectives, leading us to believe in the
potential for the industrial implementation of some of the results
here obtained.O paradigma do consumo está a mudar. Os consumidores estão
mais atentos ao que consomem e a questões relacionadas com
a sustentabilidade desses produtos. Esta mudança foi motivada
pelo cuidado de saúde associado ao consumo de produtos
naturais, mas também pela necessidade de implementar
recursos e processos mais sustentáveis como forma de combater
as mudanças climáticas, sem comprometer a qualidade e o preço
do produto final. Os pigmentos são uma das classes de
compostos que podem ser obtidos a partir de biomassa, sendo
reconhecidos pelas suas inúmeras atividades biológicas. Pelas
suas propriedades espectrais e biológicas, os pigmentos podem
ser aplicados em diversos setores de atividade que vão desde a
alimentação humana, à cosmética, nutracêutica e medicina.
Contudo, as metodologias convencionais para obtenção de
pigmentos exigem frequentemente o uso de equipamento
dispendioso e/ou o uso de solventes orgânicos considerados
tóxicos. Nesta tese, processos de extração e purificação
alternativos para recuperar os pigmentos de algas foram
desenvolvidos e propostos. Metodologias simples e eficientes,
maioritariamente baseadas em solventes aquosos, foram usadas
para obter um ou mais pigmentos de uma mesma alga. Em todos
os capítulos apresentados, um conjunto de solventes foi testado
assim como foram estudadas as condições operacionais de
forma a atingir os melhores rendimentos e/ou purezas. Soluções
aquosas de líquidos iónicos tensioativos mostraram um enorme
potencial, sob várias perspetivas, na obtenção de pigmentos
hidrofóbicos tais como clorofilas e carotenoides. Por outro lado,
a precipitação induzida de proteínas provou ser uma boa
alternativa na precipitação seletiva (e consequente purificação)
de ficobiliproteinas. Os impactos económicos e ambientais foram
estudados e resultados encorajadores foram obtidos de ambos
as perspetivas, levando-nos a acreditar no sucesso da
