Exploring missing heritability in neurodevelopmental disorders:Learning from regulatory elements

    Get PDF
In this thesis, I aimed to solve part of the missing heritability in neurodevelopmental disorders, using computational approaches. Next to the investigations of a novel epilepsy syndrome and investigations aiming to elucidate the regulation of the gene involved, I investigated and prioritized genomic sequences that have implications in gene regulation during the developmental stages of human brain, with the goal to create an atlas of high confidence non-coding regulatory elements that future studies can assess for genetic variants in genetically unexplained individuals suffering from neurodevelopmental disorders that are of suspected genetic origin

Personality Dysfunction Manifest in Words : Understanding Personality Pathology Using Computational Language Analysis

    Get PDF
Personality disorders (PDs) are some of the most prevalent and high-risk mental health conditions, and yet remain poorly understood. Today, the development of new technologies means that there are advanced tools that can be used to improve our understanding and treatment of PD. One promising tool – indeed, the focus of this thesis – is computational language analysis. By looking at patterns in how people with personality pathology use words, it is possible to gain access into their constellation of thinking, feelings, and behaviours. To date, however, there has been little research at the intersection of verbal behaviour and personality pathology. Accordingly, the central goal of this thesis is to demonstrate how PD can be better understood through the analysis of natural language. This thesis presents three research articles, comprising four empirical studies, that each leverage computational language analysis to better understand personality pathology. Each paper focuses on a distinct core feature of PD, while incorporating language analysis methods: Paper 1 (Study 1) focuses on interpersonal dysfunction; Paper 2 (Studies 2 and 3) focuses on emotion dysregulation; and Paper 3 (Study 4) focuses on behavioural dysregulation (i.e., engagement in suicidality and deliberate self-harm). Findings from this research have generated better understanding of fundamental features of PD, including insight into characterising dimensions of social dysfunction (Paper 1), maladaptive emotion processes that may contribute to emotion dysregulation (Paper 2), and psychosocial dynamics relating to suicidality and deliberate self-harm (Paper 3) in PD. Such theoretical knowledge subsequently has important implications for clinical practice, particularly regarding the potential to inform psychological therapy. More broadly, this research highlights how language can provide implicit and unobtrusive insight into the personality and psychological processes that underlie personality pathology at a large-scale, using an individualised, naturalistic approach

Non-Market Food Practices Do Things Markets Cannot: Why Vermonters Produce and Distribute Food That\u27s Not For Sale

    Get PDF
Researchers tend to portray food self-provisioning in high-income societies as a coping mechanism for the poor or a hobby for the well-off. They describe food charity as a regrettable band-aid. Vegetable gardens and neighborly sharing are considered remnants of precapitalist tradition. These are non-market food practices: producing food that is not for sale and distributing food in ways other than selling it. Recent scholarship challenges those standard understandings by showing (i) that non-market food practices remain prevalent in high-income countries, (ii) that people in diverse social groups engage in these practices, and (iii) that they articulate diverse reasons for doing so. In this dissertation, I investigate the persistent pervasiveness of non-market food practices in Vermont. To go beyond explanations that rely on individual motivation, I examine the roles these practices play in society. First, I investigate the prevalence of non-market food practices. Several surveys with large, representative samples reveal that more than half of Vermont households grow, hunt, fish, or gather some of their own food. Respondents estimate that they acquire 14% of the food they consume through non-market means, on average. For reference, commercial local food makes up about the same portion of total consumption. Then, drawing on the words of 94 non-market food practitioners I interviewed, I demonstrate that these practices serve functions that markets cannot. Interviewees attested that non-market distribution is special because it feeds the hungry, strengthens relationships, builds resilience, puts edible-but-unsellable food to use, and aligns with a desired future in which food is not for sale. Hunters, fishers, foragers, scavengers, and homesteaders said that these activities contribute to their long-run food security as a skills-based safety net. Self-provisioning allows them to eat from the landscape despite disruptions to their ability to access market food such as job loss, supply chain problems, or a global pandemic. Additional evidence from vegetable growers suggests that non-market settings liberate production from financial discipline, making space for work that is meaningful, playful, educational, and therapeutic. Non-market food practices mend holes in the social fabric torn by the commodification of everyday life. Finally, I synthesize scholarly critiques of markets as institutions for organizing the production and distribution of food. Markets send food toward money rather than hunger. Producing for market compels farmers to prioritize financial viability over other values such as stewardship. Historically, people rarely if ever sell each other food until external authorities coerce them to do so through taxation, indebtedness, cutting off access to the means of subsistence, or extinguishing non-market institutions. Today, more humans than ever suffer from chronic undernourishment even as the scale of commercial agriculture pushes environmental pressures past critical thresholds of planetary sustainability. This research substantiates that alternatives to markets exist and have the potential to address their shortcomings

Experiential-Based Research Depicting Humanistic Survival after Violent Victimization & Traumatization Exploring Human Resilience, Spirituality, Meditation & Expressive Writing

Unattended trauma-induced stressors resulting from incidents of violent victimization can escalate into mental health challenges, including trauma-related and substance-abuse disorders, which most often co-occur in a debilitating manner. Therefore, it is essential to conduct ongoing exploratory research on cognitive-based approaches (e.g., spirituality), psychological adaptations (e.g., human resilience), and trauma-informed approaches (e.g., meditation, expressive writing), which serve to reduce or offset the adverse impact of trauma. Twelve women who experienced victimization and traumatization but did not seek professional mental-health intervention were interviewed; six had been abused as children and eight as adults. Some did not contact or follow through with reports to the police either because they faced pressure to drop charges or because they were children when abused. Some also reported a lack of family intervention and support. Most reported withdrawing, secluding, and isolating themselves while benefitting from constructive coping activities that they could engage in alone (e.g., reflective journaling, prayer, and meditation), in addition to them reportedly engaging in structured support groups at church or in the community that could teach them about coping and leaving abusers. Spirituality also helped participants find new meaning regarding self-revaluating established relationships, in addition to helping them move positively forward. Some also mentioned that counseling could likely help them effectively cope futuristically, but they had not sought counseling because they thought they were not good at talking about feelings or they had negative perceptions of therapy. Key messages for survival were to be cautious and not remain in violent and abusive situations. Key messages for coping were to stay strong, address victimization and traumatization, and strive for love and happiness

Emotion regulation as a transdiagnostic construct : novel approaches to psychiatric diagnostics

    Get PDF
Diagnostic co-occurrence among mental health problems is common. It has been suggested that more research which incorporates analysis on the symptom level and cuts across categorical diagnostic boundaries is needed. This type of transdiagnostic research could have the potential to further increase our understanding of psychopathology and how we can improve treatment outcomes. Emotion regulation is one transdiagnostic process which is thought to be of relevance to several types of mental health problems. These problems include eating disorders and substance use disorders, two areas that both show a significant overlap and are associated with emotion regulation difficulties. The current thesis thus aimed to contribute to the further understanding of (a) symptom-level change and (b) measurement of emotion regulation and its role across different levels of mental health problems (with a focus on eating- and substance use disorders). The aim of Study I was to investigate symptom change over time, by assessing if treatment-seeking adults with eating disorders (N = 3159) demonstrated “symptom shifting” (simultaneously decreasing in some symptoms whilst increasing in others). Individuals demonstrating symptom shifting were then compared to nonshifting individuals on various indicators of mental illness. In the sample, 13% were found to demonstrate symptom shifting among eating disorder-related behaviors. The proportion of patients with symptom shifting increased to 19% when including shifts to deliberate self-harm or substance use (in addition to eating disorder-related behaviors). Individuals with symptom shifting reported significantly higher levels of other psychiatric symptoms (such as depression and anxiety), clinical impairment, and emotion regulation difficulties compared to nonshifting individuals. It was concluded that individuals with symptom shifting could be a group in need of tailored monitoring in clinical practice, as well as treatment approaches targeting several types of risky behaviors. The aim of Study II was to further investigate symptom change over time, by investigating if symptom shifting occurred between symptoms of eating- and substance use disorders in a general population sample (N = 3315). Individuals with symptom shifting were also compared to other individuals with different temporal symptom profiles. A small group of individuals (2% of the whole sample) demonstrated symptom shifting (which represented 10% of individuals who suffered persistent problems in any or both areas). Individuals with symptom shifting reported higher levels of psychiatric symptoms (such as depression, anxiety, suicidality) and demonstrated higher polygenic risk scores for psychiatric disorders. Levels were comparable to individuals who suffered persistent co-occurring problems with eating- and substance use disorder symptoms over time. The results contribute to the understanding of the overlap between eating- and substance use disorders, and again highlight the need for health care services and treatments that address several problem areas simultaneously. The aim of Study III was to evaluate psychometric properties and contrast two of the most used measures of emotion regulation, for which a community sample of adults residing in the US was used (N = 843). The Emotion Regulation Questionnaire (ERQ) and the Difficulties in Emotion Regulation Scale (DERS; including its short form DERS-16) were evaluated using correlations and confirmatory factor analyses. The measures were found to converge in expected ways. Satisfactory fits were found for the ERQ and DERS- 16, but not for the original DERS. The DERS showed greater concurrent validity with psychiatric symptoms (depression, anxiety, stress, substance use) than the ERQ. It was concluded that the validity of both measures to assess emotion regulation was supported, and that the DERS-16 with its shorter format is a clinically useful tool to assess emotion regulation in practice. The aim of Study IV was to investigate the relationship between emotion regulation difficulties across different levels of substance use, and the role of co-occurring psychiatric symptoms. Data from two main sources were used, including the community sample in Study III (n = 843) and two substance use disorder populations from residential treatment clinics in the US (n = 415). Emotion regulation was significantly associated with both substance use frequency and severity across a variety of substances. Of the different facets of emotion regulation (DERS subscales), difficulties controlling impulsive behaviors was found to be specifically associated with greater substance use severity. No significant moderating effect of psychiatric symptoms (depression, anxiety, stress) was found. Results suggested that emotion regulation difficulties are of increasing relevance across the spectrum of substance use, from no use, to misuse, to poly-substance dependence. The studies could contribute to method development in the area of symptom-level change and furthers our understanding of emotion regulation as a relevant transdiagnostic construct. They also could contribute by highlighting important clinically relevant outcomes (such as symptom shifting), which could have potential implications in psychiatric research and clinical practice/health care. Studying common functions of risky behaviors, such as eating disorder-related behaviors, substance use, and deliberate self-harm, from an emotion regulation perspective is suggested to be of importance to consider in future research

    Conversations on Empathy

    Get PDF
In the aftermath of a global pandemic, amidst new and ongoing wars, genocide, inequality, and staggering ecological collapse, some in the public and political arena have argued that we are in desperate need of greater empathy — be this with our neighbours, refugees, war victims, the vulnerable or disappearing animal and plant species. This interdisciplinary volume asks the crucial questions: How does a better understanding of empathy contribute, if at all, to our understanding of others? How is it implicated in the ways we perceive, understand and constitute others as subjects? Conversations on Empathy examines how empathy might be enacted and experienced either as a way to highlight forms of otherness or, instead, to overcome what might otherwise appear to be irreducible differences. It explores the ways in which empathy enables us to understand, imagine and create sameness and otherness in our everyday intersubjective encounters focusing on a varied range of "radical others" – others who are perceived as being dramatically different from oneself. With a focus on the importance of empathy to understand difference, the book contends that the role of empathy is critical, now more than ever, for thinking about local and global challenges of interconnectedness, care and justice

A Critical Review Of Post-Secondary Education Writing During A 21st Century Education Revolution

    Get PDF
Educational materials are effective instruments which provide information and report new discoveries uncovered by researchers in specific areas of academia. Higher education, like other education institutions, rely on instructional materials to inform its practice of educating adult learners. In post-secondary education, developmental English programs are tasked with meeting the needs of dynamic populations, thus there is a continuous need for research in this area to support its changing landscape. However, the majority of scholarly thought in this area centers on K-12 reading and writing. This paucity presents a phenomenon to the post-secondary community. This research study uses a qualitative content analysis to examine peer-reviewed journals from 2003-2017, developmental online websites, and a government issued document directed toward reforming post-secondary developmental education programs. These highly relevant sources aid educators in discovering informational support to apply best practices for student success. Developmental education serves the purpose of addressing literacy gaps for students transitioning to college-level work. The findings here illuminate the dearth of material offered to developmental educators. This study suggests the field of literacy research is fragmented and highlights an apparent blind spot in scholarly literature with regard to English writing instruction. This poses a quandary for post-secondary literacy researchers in the 21st century and establishes the necessity for the literacy research community to commit future scholarship toward equipping college educators teaching writing instruction to underprepared adult learners

False Advertising and Young Girls’ Self Esteem

It is becoming impossible to recognize the level of photoshop on advertisements. However, little is known how continuous exposure to such images can negatively impact the short- and long-term psyche of young girls aged 6 to 14 years old. The study’s theoretical framework was Benet’s polarities of democracy, emphasizing freedom and authority and human rights and communal obligations. The research question focused on the negative impacts false advertising has on young girls and what public policies could assist in mitigating these short- and long-term impacts. A general qualitative design, with semistructured interviews of seven adult participants with relevant insight on the negative impacts to young girls, generated themes through multicycle coding. Themes included (a) impressionable young girls, (b) deceptive advertisers, and (c) protecting women. Congress can use these findings to inform policies supportive to protecting young girls’ self-esteem from false advertising. Such policies could assist in reducing current behaviors of young girls from such advertisements including but not limited to eating disorders, body dysmorphia, and depression. Implications for positive social change include informing Congress of potential systemic issues that either hinder or democratize young girls’ rights within Congress and further supports protecting the mental health development of young girls. Therefore, providing a tax or monetary benefit to participating businesses proving they did not use photoshop could provide a greater motivation in producing natural models as well as set an example for what models actually look like for young girls

Constructing a profile for proactive career self-management in public higher education institutions in Ghana

Text in English with abstracts and keywords in English, isiXhosa, and isiZuluThe principal focus of this research study was to investigate the relationship among psychosocial career pre-occupation, social connectedness, and organisational commitment, and to determine whether an overall proactive career management profile can be constructed to inform career self-management practices in public higher education institutions (HEIs) in Ghana. Again, the research study intended to provide a background for testing hypotheses and theories as well as moderating the effect of socio-demographic characteristics (age, gender, marital status and job level) on the relationship between psychosocial career pre-occupations, social connectedness and organisational commitment. A cross-sectional quantitative approach was conducted on a purposively selected sample of senior staff comprising academic and administrative staff (n = 288), from a single public higher educational institution in Ghana. The measuring instruments include a career pre-occupations inventory workplace friendship scale and an organisational commitment scale. Descriptive statistics (mean, standard deviation, skewness and kurtosis), bi-variate correlation analysis (Pearson product moment correlation coefficient), and inferential and multivariate statistics (SEM analysis, moderated regression analysis, ANOVAs and independent sample t-test) were used for the study. Descriptive, bi-variate correlation and inferential statistics revealed that individual psychosocial career pre-occupations, social connectedness and organisational commitment can be used as elements within a proactive career self-management framework within the Ghanaian higher education institutions. The results of the moderated analysis showed that respondents’ gender and job level moderated employees’ level of psychosocial career pre-occupations in predicting organisational commitment. Furthermore, the test for significant mean differences revealed that gender, marital status and job level differed marginally in their psychosocial career pre-occupations, social connectedness and commitment to the organisation. Theoretically and empirically, the results advanced the career construction theory by empirically validating the core elements of career self-management. Practically, a recommendation was made to inform human resource (HR) managers and HR practitioners in public higher education institutions in Ghana and the consequences indicated in the study offer the opportunity to monitor and provide strategies and interventions for employees in their quest for career choices.Ugqaliselo oluphambili kolu phononongo lophando yayikukuphanda ubudlelwane phakathi komsebenzi wangaphambi kwekhondo ngokwesimo sengqondo nangokwezentlalo (psychosocial career pre-occupation), ukuziphatha, nokunxulumana kwezinto zokuziphatha nezentlalo (social connectedness), kunye nokuzibophelela kulungelelaniso /kucwangciso lombutho (organisational commitment), nokuqonda ukuba ingaba iprofayili yeendlela zokulawula ikhondo elisebenzayo elipheleleyo inokwakhelwa ukwazisa izenzo zekhondo lomsebenzi lokuzilawula kumaziko emfundo ephakamileyo karhulumente/oluntu (HEIs) eGhana.
