TCM Database@Taiwan: The World's Largest Traditional Chinese Medicine Database for Drug Screening In Silico
Rapid advancing computational technologies have greatly speeded up the development of computer-aided drug design (CADD). Recently, pharmaceutical companies have increasingly shifted their attentions toward traditional Chinese medicine (TCM) for novel lead compounds. Despite the growing number of studies on TCM, there is no free 3D small molecular structure database of TCM available for virtual screening or molecular simulation. To address this shortcoming, we have constructed TCM Database@Taiwan (http://tcm.cmu.edu.tw/) based on information collected from Chinese medical texts and scientific publications. TCM Database@Taiwan is currently the world's largest non-commercial TCM database. This web-based database contains more than 20,000 pure compounds isolated from 453 TCM ingredients. Both cdx (2D) and Tripos mol2 (3D) formats of each pure compound in the database are available for download and virtual screening. The TCM database includes both simple and advanced web-based query options that can specify search clauses, such as molecular properties, substructures, TCM ingredients, and TCM classification, based on intended drug actions. The TCM database can be easily accessed by all researchers conducting CADD. Over the last eight years, numerous volunteers have devoted their time to analyze TCM ingredients from Chinese medical texts as well as to construct structure files for each isolated compound. Two Birds with One Stone? Possible Dual-Targeting H1N1 Inhibitors from Traditional Chinese Medicine
The H1N1 influenza pandemic of 2009 has claimed over 18,000 lives. During this pandemic, development of drug resistance further complicated efforts to control and treat the widespread illness. This research utilizes traditional Chinese medicine Database@Taiwan (TCM Database@Taiwan) to screen for compounds that simultaneously target H1 and N1 to overcome current difficulties with virus mutations. The top three candidates were de novo derivatives of xylopine and rosmaricine. Bioactivity of the de novo derivatives against N1 were validated by multiple machine learning prediction models. Ability of the de novo compounds to maintain CoMFA/CoMSIA contour and form key interactions implied bioactivity within H1 as well. Addition of a pyridinium fragment was critical to form stable interactions in H1 and N1 as supported by molecular dynamics (MD) simulation. Results from MD, hydrophobic interactions, and torsion angles are consistent and support the findings of docking. Multiple anchors and lack of binding to residues prone to mutation suggest that the TCM de novo derivatives may be resistant to drug resistance and are advantageous over conventional H1N1 treatments such as oseltamivir. How supplier selection criteria affects business performance? A study of UK automotive sector
According to KPMG international (2015), global sales of automobiles are forecasted to reach 73.9 million vehicles and expected to hit 100 million units in the next two years. This shows that automotive sector has a tremendous growth potential and UK automotive sector is no different. However, in recent years the growing environmental awareness has become a major concern for automotive sector as they are faced with pressure of reducing carbon emissions as well as the costs. Suppliers play a significant role in achieving environmental goals set by organisations. Under these circumstances it is worth exploring the criteria that are used in assessing suppliers including the green aspects and how that affects the business performance. Design/methodology/approach: This research adopts a mixed method research approach. In order to collect the quantitative data a survey questionnaire was constructed and sent to automotive businesses listed in the FAME database. In order to triangulate the findings of this study, survey was complemented with in-depth interviews. Around 100 automotive manufacturers were invited for the survey however only 38 usable responses were received. In total seven semi-structured interviews were also conducted with people from different backgrounds and work experiences in the automotive sector. Findings: Literature identified delivery, cost, quality and technology as the supplier assessment criteria commonly used in assessing suppliers in automotive industries. Yet the issue of culture and green supply chain practices (GSP) were also widely concerned in several studies. The data analysis showed that delivery, quality, cost, technology, culture are correlated with exception of green supply chain practices. GSP was only found to be correlated with technology and cultural criteria. Semi-structured interviews suggest delivery and quality as the most important criteria when assessing supplier because of their greater impact toward business performance and reputation. Findings from all respondents also showed that most automotive manufacturers have already adopted environmental competency in their criteria. However, interviewees mentioned that this criterion does not take a major role in assessment compared with other criteria. The results also indicate that all factors studied do affect the business performance of automotive organisations. Value: This study contributes to the limited literature focused on assessing supplier selection criteria and business performance linkage in the UK automotive organisations. In addition, most studies on supplier selection and business performance ignore the green practices as important criteria which this study aims to address. Research limitations/implications: The study is based on the findings from a limited survey responses and semi-structured interviews. Having larger sample population would certainly improve the validity of the findings. The perspective of SMEs and large businesses with regard to each supplier selection criterion may be different hence the future research in this domain would also provide some valuable contributions. Practical implications: The survey responses indicate green supply practices as one of the important criteria in supplier selection. This suggests that automotive manufacturers should realize the importance of green practices while selecting their suppliers. Migrants in their family contexts: Application of a methodology
The composition of immigrant families is a topic which has attracted considerable public and political attention in recent years. In the late 1980s concern was expressed over the size of some households of Pacific Island peoples in New Zealand. In the 1990s a more persistent concern has been with the incidence of what have been called âastronautâ families â families where one of the partners is persistently absent overseas. The âastronautâ family phenomenon has been most commonly associated with Chinese immigrants from Hong Kong and Taiwan.
This report uses a novel methodology to examine the incidence of âastronautâ families in New Zealand at the time of the 1991 and 1996 censuses. The methodology is described in some detail in the first section, and it is hoped that the careful attention to the procedures used to examine migrants, who have been identified in the census, in their family contexts will stimulate further research in this area.
The second part of the report presents the findings of an initial exploration of 1991 and 1996 census data using the methodology outlined in the first section. It is clear from results of this inquiry that the âastronautâ family phenomenon is well established amongst some components of New Zealandâs Asian community. However, it is not as widespread as media comment in 1995 and 1996, before the 1996 election, suggested.
It is important to develop ways of assessing characteristics of immigrant family structures in order to counter unsubstantiated assertions which promote negative stereotypes of immigrant communities. This research, which builds on a project supported by the Marsden Fund in 1997, suggest one fruitful avenue for making more extensive use of census data on immigrants in New Zealand to provide more objective assessment of âmigrants in social contextâ
Resistance and resilience of socialâecological systems to recurrent typhoon disturbance on a subtropical island: Taiwan
Tropical cyclones (TCs) have major effects on ecological and social systems. However, studies integrating the effects of TCs on both social and ecological systems are rare, especially in the northwest Pacific, where the frequency of TCs (locally named typhoons) is the highest in the world. We synthesized studies of effects of recurrent typhoons on social and ecological systems in Taiwan over the last several decades. Many responses to TCs are comparable between social and ecological systems. High forest ecosystem resistance, evident from tree mortality below 2% even following multiple strong typhoons, is comparable with resistance of social systems, including the only 4% destruction of river embankments following a typhoon that brought nearly 3000 mm rainfall in three days. High resilience as reflected by quick returns of leaf area index, mostly in one year, and streamwater chemistry, one to several weeks to preâtyphoon levels of ecosystems, are comparable to quick repair of the power grid within one to several days and returns of vegetable price within several weeks to preâtyphoon levels of the social systems. Landslides associated with intense typhoons have buried mountain villages and transported large quantities of woody debris to the coast, affecting the coastal plains and reefs, illustrating a ridgeâtoâreef link between ecological and societal systems. Metrics of both social and ecological function showed large fluctuations in response to typhoons but quickly returned to preâdisturbance levels, except when multiple intense typhoons occurred within a single season. Our synthesis illustrates that the socialâecological systems in Taiwan are highly dynamic and responsive to frequent typhoon disturbance, with extraordinarily high resistance and resilience. For ecosystems, the efficient responsiveness results from the selective force of TCs on ecosystem structure and processes. For social systems, it is the result of the effects of TCs on planning and decision making by individuals (e.g., farmers), management sectors, and ultimately the government. In regions with frequent TCs, the socialâecological systems are inevitably highly dynamic and rapid responses are fundamental to system resistance and resilience which in turn is key to maintaining structure and function of the socialâecological systems
Fifteen new risk loci for coronary artery disease highlight arterial-wall-specific mechanisms
Coronary artery disease (CAD) is a leading cause of morbidity and mortality worldwide. Although 58 genomic regions have been associated with CAD thus far, most of the heritability is unexplained, indicating that additional susceptibility loci await identification. An efficient discovery strategy may be larger-scale evaluation of promising associations suggested by genome-wide association studies (GWAS). Hence, we genotyped 56,309 participants using a targeted gene array derived from earlier GWAS results and performed meta-analysis of results with 194,427 participants previously genotyped, totaling 88,192 CAD cases and 162,544 controls. We identified 25 new SNP-CAD associations (P < 5 Ă 10(-8), in fixed-effects meta-analysis) from 15 genomic regions, including SNPs in or near genes involved in cellular adhesion, leukocyte migration and atherosclerosis (PECAM1, rs1867624), coagulation and inflammation (PROCR, rs867186 (p.Ser219Gly)) and vascular smooth muscle cell differentiation (LMOD1, rs2820315). Correlation of these regions with cell-type-specific gene expression and plasma protein levels sheds light on potential disease mechanisms
Peripheral trauma and risk of dystonia: What are the evidences and potential co-risk factors from a population insurance database?
BACKGROUND: Dystonia is a neurological syndrome typically resulting in abnormal postures. OBJECTIVES: We tested the role of physical injury as potential risk factor for development of dystonia using The National Health Insurance Research Database of Taiwan. METHODS: We identified 65704 people who were coded in the database as having had peripheral traumatic injuries (ICD-9-CM 807-848 and 860-959) in the year 2000. Patients with traumatic brain or spine injuries were excluded from analysis. We matched them using purposive sampling with 65704 people in the database who had not suffered peripheral trauma. We looked then at the incidence of dystonia occurring at least 1 year from the date of the peripheral trauma until 2011. Psychiatric symptoms (depression and anxiety) and sleeps difficulties have been investigated as potential covariates. RESULTS: We found 189 patients with dystonia (0.28%) in the trauma group, and 52 patients with dystonia (0.08%) in the non-trauma group. Trauma was independently associated with dystonia (adjusted HR = 3.12, 95% CI = 2.30-4.24). The incidence density of dystonia in the trauma group was 2.27 per 10000 person-years, while it was 0.71 per 10000 person-years in the non-trauma group Beyond the peripheral trauma, other variables associated to the incidence of dystonia included female sex, aged 40 years and above, depression and sleep disorders. CONCLUSION: These data from a large population dataset support traumatic injury as a risk factor for the development of dystonia
