The Institute for Pulsed Power and Microwave Technology (Institut für Hochleistungsimpuls- und Mikrowellentechnik - IHM) is doing research in the areas of pulsed power and high power microwave technologies. Both, research and development of high power sources as well as related applications are in the focus. Applications for pulsed power technologies are ranging from material processing to bioelectrics. Microwave technologies are focusing on RF sources for electron cyclotron resonance heating and on applications for material processing at microwave frequencies

Control óptico de la expresión génica en sistemas biológicos mediante nanopartículas de oro: Expresión génica fototérmica en Escherichia coli y silenciamiento génico en Chlamydomonas reinhardtii
Las nanopartículas de oro pueden ser encontrados de diferentes formas,
tamaños y que determinan sus características químicas y físicas. Las
propiedades físicas y químicas de las nanopartículas metálicas pueden ser
moduladas al cambiar su forma, tamaño y la química de su superficie. Por lo
tanto, esto ha permitido su uso en una gran variedad de aplicaciones en los
sectores industriales y académicos. Una de las características de las
nanopartículas metálicas es su habilidad para actuar como convertidos de
energía optotérmicos. Esta característica ha sido utilizada en muchas
aplicaciones donde las nanopartículas son acopladas con sistemas de respuesta
térmica para generar una respuesta óptica. En este estudio, nosotros
sintetizamos nanopartículas metálicas que son mayormente esféricas en su
forma con un promedio de diámetro de 20.07 nm. En este estudio, nosotrosutilizamos dos fuentes de luz: LED y láser. Diferentes enfoques estadísticos
fueron utilizados para medir la potencia y capacidad funcional de la luz láser y
LED así como identificar a la variable más necesaria para incrementar la
temperatura en una solución de nanopartículas de oro. En este trabajo se
realizaron simultáneamente técnicas teóricas y experimentales para evaluar los
diferentes factores que afectan la generación de calor en la superficie de
nanopartículas cuando son expuestas a una longitud de onda específica por la
luz láser y LED. Respecto al láser, los resultados mostraron que los factores que
más contribuyeron al cambio de temperatura exhibido en la solución de
nanopartículas resultaron ser el poder del láser, la concentración de las
nanopartículas de oro, la interacción tiempo × láser y el tiempo de iluminación.
Nosotros reportamos un modelo de regresión que permite predecir la generación
de calor y cambios de temperatura con errores estándares residuales en menos
de 4%. Los resultados son altamente relevantes para diseños futuros y en el
desarrollo de aplicaciones donde las aplicaciones de nanopartículas sean
incorporadas en los sistemas para inducir un cambio en la temperatura a partir
de la exposición de con luz. Respecto al LED, nosotros analizamos
estadísticamente la temperatura producida en la superficie de las nanopartículas
de oro cuando utilizando LED como fuente de luz. Los resultados mostrados que
los efectores principales y las interacciones de todos los factores fueron
significativos. Finalmente, basados en el modelo de regresión presentado, los
coeficientes de regresión y los resultados de ANOVA nos permiten presentan un
poderoso modelo de regresión que muestra las relaciones entre la temperatura
de cambio y sus variables. Nosotros simulamos el cambio de generación de
nuestras nanopartículas de oro cuando la solución con nanopartículas de oro era
iluminada con una fuente de luz LED. Nosotros demostramos que el máximo
incremento de temperatura en la solución de nanopartículas (resultados de
simulación) cotejaron excelentemente con nuestras observaciones (resultados
prácticos). Para evaluar nuestra aplicación fototérmica obtenida a partir de
nanopartículas de oro en un sistema biológico en células, evaluamos su
factibilidad en la producción de proteína con enfoque fototérmico por primera vez.
Para lograr este objetivo, utilizamos luz LED en vez de un dispositivo láser al
considerarse como un método nuevo, barato, inofensivo y conmutable para
sistemas biológicos vivos. Después de sintetizar las nanopartículas de oro y
obtener su perfil de temperatura, nosotros diseñamos un gen sintético, donde el
sitio de unión a ribosoma pudiera ser activo y trabajar eficientemente a 37°C.
Basado en el modelo de regresión lineal y en análisis de respuesta de superficie
de curva, nosotros encontramos el cómo proveer la temperatura necesaria. De
esta manera, nosotros mostramos el uso de nanopartículas metálicas y LED
como fuente de luz pueden trabajar eficientemente en una estructura tipo stem –
loop que contiene un sitio a unión a ribosoma y consecuentemente una alta
producción de mCherry es logrado. Además, para mostrar su factibilidad en la
desbridamiento de dsDNA unido a nanopartículas metálicas a partir de LED como
fuente de luz, nosotros elaboramos conmutadores (nanopartículas de oro
acoplados con dsDNA) y finalmente fueron caracterizados. Entonces, nosotros
mostramos la factibilidad del desbridamiento del dsDNA unido a nanopartículas
de oro (prueba in vitro) utilizando LED como fuente de luz bajo diferentes
longitudes de onda. La prueba demostró ser exitosa y se mostró la probabilidad
de que el calor generado fototérmica pueda ser utilizado para el silenciamiento
de genes por antisentido en células de microalgas vivas
Nonosecond Pulsed Electric Field Induced Changes in Dielectric Properties of Biological Cells
Nanosecond pulsed electric field induced biological effects have been a focus of research interests since the new millennium. Promising biomedical applications, e.g. tumor treatment and wound healing, are emerging based on this principle. Although the exact mechanisms behind the nanosecond pulse-cell interactions are not completely understood yet, it is generally believed that charging along the cell membranes (including intracellular membranes) and formation of membrane pores trigger subsequent biological responses, and the number and quality of pores are responsible for the cell fate. The immediate charging response of a biological cell to a nanosecond pulsed electric field exposure relies on the dielectric properties of its cellular components. Conversely, intense nanosecond pulses will change these properties due to conformational and functional changes. Hence, an understanding of biodielectric phenomena is necessary to explain the underlying interaction mechanisms between nanosecond pulses and biological materials.
To this end, we have investigated the changes in dielectric characteristics of biological cells and tissues after exposure to multiple nanosecond pulses. Significant differences have been observed in dielectric properties and membrane integrity of Jurkat cells for exposures to nanosecond and microsecond pulsed electric fields despite delivery of the same energy, suggesting different pore formation and development mechanisms. The effect of nanosecond pulsed electric fields on the dielectric properties of Jurkat cells is long-lasting which is consistent with predictions of much longer pore resealing times for shorter pulses. Strong correlation between short-term plasma membrane conductivity and long-term cell survival has also been observed for different nanosecond-exposure conditions. Together with the studies on tissues, we demonstrate that dielectric spectroscopy is capable of assessing conformational and possibly functional changes of cells after exposure to nanosecond pulsed electric fields on biologically relevant time scales, and in turn, evaluate and compare the efficacy of chosen pulse parameters
Femtosecond laser microfabricated devices for biophotonic applications
Femtosecond Laser DirectWriting has emerged as a key enabling technology
for realising miniaturised biophotonic applications offering clear advantages
over competing soft-lithography, ion-exchange and sol-gel based fabrication
techniques. Waveguide writing and selective etching with three-dimensional
design flexibility allows the development of innovative and unprecedented
optofluidic architectures using this technology. The work embodied in this
thesis focuses on utilising the advantages offered by direct laser writing in
fabricating integrated miniaturised devices tailored for biological analysis.
The first application presented customised the selective etching phenomenon
in fused silica by tailoring the femtosecond pulse properties during the writing
process. A device with an embedded network of microchannels with a
significant difference in aspect-ratio was fabricated, which was subsequently
applied in achieving the high-throughput label-free sorting of mammalian
cells based on cytoskeletal deformability. Analysis on the device output cell
population revealed minimal effect of the device on cell viability.
The second application incorporated an embedded microchannel in fused
silica with a monolithically integrated near-infrared optical waveguide. This
optofluidic device implemented the thermally sensitive emission spectrum
of semiconductor nanocrystals in undertaking remote thermometry of the
localised microchannel environment illuminated by the waveguide. Aspects
relating to changing the wavelength of illumination from the waveguide were
analysed. The effect of incorporating carbon nanotubes as efficient heaters
within the microchannel was investigated. Spatio-thermal imaging of the
microchannel illuminated by the waveguide revealed the thermal effects to
extend over distances appreciably longer than the waveguide cross-section.
On the material side of direct laser writing, ultra-high selective etching is
demonstrated in the well-known laser crystal Nd:YAG. This work presents
Nd:YAG as a material with the potential to develop next-generation optofluidic
devices
Analysis of spine plasticity in CA1 hippocampal pyramidal neurons employing live cell nanoscopic imaging
In der Großhirnrinde von Säugetieren befindet sich die Mehrheit erregender Synapsen auf Dornfortsätzen, kleinen dendritischen Ausbuchtungen, die in Größe und Form stark variieren. Die Auslösung aktivitätsabhängiger synaptischer Langzeitplastizität geht mit strukturellen Veränderungen dendritischer Dornen einher. Da das beugungsbegrenzte Auflösungsvermögen konventioneller Lichtmikroskope nicht ausreicht um die Morphologie der Dornen verlässlich zu untersuchen, stellte die Elektronenmikroskopie bisher das wichtigste bildgebende Verfahren zur Erforschung von struktureller Plastizität dar, blieb dabei jedoch auf die Betrachtung fixierter Gewebeproben beschränkt. Die Anwendung hochauflösender Laser-Raster-Mikroskopie mit Stimulierter-Emissions-Auslöschung hat es mir möglich gemacht, die Dynamik dendritischer Dornenmorphologie in lebenden Zellen zu studieren. Die N-Methyl-D-Aspartat-Rezeptor-abhängige Langzeitpotenzierung von Pyramidenzellen der Cornu-Ammonis Region 1 des Hippocampus bildete dabei den Mechanismus, welcher plastische Veränderungen hervorrief. Nach Potenzierung exzitatorischer Synapsen durch die lokale Ultraviolett-Photolyse von caged-Glutamat wurde ein starker, vorübergehender Anstieg des Anteils dendritischer Dornen mit sichelförmigen Köpfen und ein leichter, anhaltender Zuwachs an pilzförmigen Dornfortsätzen über einen Zeitraum von 50 Minuten beobachtet. Meine Untersuchungen ergänzen frühere Studien zur Wechselbeziehung zwischen synaptischer Potenzierung und struktureller Plastizität dendritischer Dornen und korrespondieren mit dem aktuellen Kenntnisstand der zu Grunde liegenden molekularen Mechanismen.The majority of excitatory synapses in the cortex of mammalian brains is situated on dendritic spines, small protrusions, heterogeneous in size and shape. The induction of activity-dependent long-term synaptic plasticity has been associated with changes in the ultrastructure of spines, particularly in size, head shape and neck width. Since the dimensions of dendritic spines are at the border of the diffraction-limited resolving power of conventional light microscopes, until recently, electron microscopy on fixed tissue constituted the primary method for investigations on spine morphology. I have employed live cell stimulated emission depletion imaging to analyse spine motility and structural transitions in response to n-methyl-d-aspartate receptor dependent long-term potentiation over time at super-resolution in Cornu Ammonis area 1 pyramidal neurons of the hippocampus. Local induction of long-term potentiation via ultraviolet photolysis of caged glutamate facilitated a strong transient increase in the proportion of spines with curved heads and a subtle persistent growth in the amount of mushroom spines over a time course of 50 minutes. My findings reinforce previous investigations on the relation of synaptic potentiation and spine motility, and are in good agreement with the current knowledge of the molecular mechanisms underlying long-term plasticity
Theranostic Gelatin Nanoparticles for Antigen Delivery and Combined Strategies for Transcutaneous Application
Transcutaneous application of vaccines is a promising strategy to enhance the effectiveness of vaccination using a reachable route of administration. Additionally, replacing the conventional needles with skin mechanical penetration techniques as microneedles or skin laser microporation will offer great advantages. This transcutaneous delivery techniques are pain-free and will help to avoid the hazards of needles. For the delivery of antigens, nanocarriers are so promising to enhance and modulate their immune response. The nanocarriers offer merits such as antigen protection from degradation, and controlling the release rate of the antigen. Additionally, due to the particulate nature of the nanocarriers, they can potentially display the antigen in a way that better mimics pathogens. For this aim, ovalbumin as a model antigen has been delivered using functionalized theranostic gelatin nanoparticles to bone marrow-derived dendritic cells (BMDCs). The nanoparticles were rendered fluorescent by using a novel imaging agent (gold and silver alloy nanoclusters) that emits near-infra red light. This was beneficial to study the nanoparticles uptake by BMDCs and also to image the nanoparticles within the skin tissue. Finally, the developed theranostic nanocarriers induced the maturation of the BMDCs and enhanced the proliferation of both helper T cells (CD4+) and cytotoxic T cells (CD8+). This indicates the potential efficacy of the delivery system for vaccination either against allergy or viruses and tumors
