    Methods for Analysing Endothelial Cell Shape and Behaviour in Relation to the Focal Nature of Atherosclerosis

    The aim of this thesis is to develop automated methods for the analysis of the spatial patterns, and the functional behaviour of endothelial cells, viewed under microscopy, with applications to the understanding of atherosclerosis. Initially, a radial search approach to segmentation was attempted in order to trace the cell and nuclei boundaries using a maximum likelihood algorithm; it was found inadequate to detect the weak cell boundaries present in the available data. A parametric cell shape model was then introduced to fit an equivalent ellipse to the cell boundary by matching phase-invariant orientation fields of the image and a candidate cell shape. This approach succeeded on good quality images, but failed on images with weak cell boundaries. Finally, a support vector machines based method, relying on a rich set of visual features, and a small but high quality training dataset, was found to work well on large numbers of cells even in the presence of strong intensity variations and imaging noise. Using the segmentation results, several standard shear-stress dependent parameters of cell morphology were studied, and evidence for similar behaviour in some cell shape parameters was obtained in in-vivo cells and their nuclei. Nuclear and cell orientations around immature and mature aortas were broadly similar, suggesting that the pattern of flow direction near the wall stayed approximately constant with age. The relation was less strong for the cell and nuclear length-to-width ratios. Two novel shape analysis approaches were attempted to find other properties of cell shape which could be used to annotate or characterise patterns, since a wide variability in cell and nuclear shapes was observed which did not appear to fit the standard parameterisations. Although no firm conclusions can yet be drawn, the work lays the foundation for future studies of cell morphology. To draw inferences about patterns in the functional response of cells to flow, which may play a role in the progression of disease, single-cell analysis was performed using calcium sensitive florescence probes. Calcium transient rates were found to change with flow, but more importantly, local patterns of synchronisation in multi-cellular groups were discernable and appear to change with flow. The patterns suggest a new functional mechanism in flow-mediation of cell-cell calcium signalling

    Hybrid image representation methods for automatic image annotation: a survey

    In most automatic image annotation systems, images are represented with low level features using either global methods or local methods. In global methods, the entire image is used as a unit. Local methods divide images into blocks where fixed-size sub-image blocks are adopted as sub-units; or into regions by using segmented regions as sub-units in images. In contrast to typical automatic image annotation methods that use either global or local features exclusively, several recent methods have considered incorporating the two kinds of information, and believe that the combination of the two levels of features is beneficial in annotating images. In this paper, we provide a survey on automatic image annotation techniques according to one aspect: feature extraction, and, in order to complement existing surveys in literature, we focus on the emerging image annotation methods: hybrid methods that combine both global and local features for image representation

    Face detection and clustering for video indexing applications

    This paper describes a method for automatically detecting human faces in generic video sequences. We employ an iterative algorithm in order to give a confidence measure for the presence or absence of faces within video shots. Skin colour filtering is carried out on a selected number of frames per video shot, followed by the application of shape and size heuristics. Finally, the remaining candidate regions are normalized and projected into an eigenspace, the reconstruction error being the measure of confidence for presence/absence of face. Following this, the confidence score for the entire video shot is calculated. In order to cluster extracted faces into a set of face classes, we employ an incremental procedure using a PCA-based dissimilarity measure in con-junction with spatio-temporal correlation.     A robust adaptive wavelet-based method for classification of meningioma histology images

    Intra-class variability in the texture of samples is an important problem in the domain of histological image classification. This issue is inherent to the field due to the high complexity of histology image data. A technique that provides good results in one trial may fail in another when the test and training data are changed and therefore, the technique needs to be adapted for intra-class texture variation. In this paper, we present a novel wavelet based multiresolution analysis approach to meningioma subtype classification in response to the challenge of data variation.We analyze the stability of Adaptive Discriminant Wavelet Packet Transform (ADWPT) and present a solution to the issue of variation in the ADWPT decomposition when texture in data changes. A feature selection approach is proposed that provides high classification accuracy

    Semantic spaces revisited: investigating the performance of auto-annotation and semantic retrieval using semantic spaces

    Semantic spaces encode similarity relationships between objects as a function of position in a mathematical space. This paper discusses three different formulations for building semantic spaces which allow the automatic-annotation and semantic retrieval of images. The models discussed in this paper require that the image content be described in the form of a series of visual-terms, rather than as a continuous feature-vector. The paper also discusses how these term-based models compare to the latest state-of-the-art continuous feature models for auto-annotation and retrieval
