    A blaVIM-2 Plasmid Disseminating in Extensively Drug-Resistant Clinical Pseudomonas aeruginosa and Serratia marcescens Isolates

    Infections caused by carbapenem-resistant Enterobacteriaceae isolates are an issue of major global concern (1). Genes coding for metallo-β-lactamases (MβLs) identified in clinical isolates are associated with mobile elements and subject to horizontal genetic transfer (HGT) events (2–6). VIM-2 is present on numerous plasmids, but only pNOR-2000 from Pseudomonas aeruginosa COL-1 from France (7, 8) and pLD209 from Pseudomonas putida LD209 from Argentina (9) have been completely sequenced. Here, we report the complete sequence and characterization of plasmid pDCPR1 harboring a blaVIM-2 gene cassette in a Tn402-type class 1 integron, which was isolated from two extensively drug-resistant strains: P. aeruginosa 802 (from a burn patient at the Hospital Municipal de Quemados, Argentina, 2005) and S. marcescens 68313 (Sanatorio Sagrado Corazón, Argentina, 2012).Fil: Vilacoba, Elisabet.     Genetic Transformation of Apomictic Grasses: Progress and Constraints

    The available methods for plant transformation and expansion beyond its limits remain especially critical for crop improvement. For grass species, this is even more critical, mainly due to drawbacks in in vitro regeneration. Despite the existence of many protocols in grasses to achieve genetic transformation through Agrobacterium or biolistic gene delivery, their efficiencies are genotype-dependent and still very low due to the recalcitrance of these species to in vitro regeneration. Many plant transformation facilities for cereals and other important crops may be found around the world in universities and enterprises, but this is not the case for apomictic species, many of which are C4 grasses. Moreover, apomixis (asexual reproduction by seeds) represents an additional constraint for breeding. However, the transformation of an apomictic clone is an attractive strategy, as the transgene is immediately fixed in a highly adapted genetic background, capable of large-scale clonal propagation. With the exception of some species like Brachiaria brizantha which is planted in approximately 100 M ha in Brazil, apomixis is almost non-present in economically important crops. However, as it is sometimes present in their wild relatives, the main goal is to transfer this trait to crops to fix heterosis. Until now this has been a difficult task, mainly because many aspects of apomixis are unknown. Over the last few years, many candidate genes have been identified and attempts have been made to characterize them functionally in Arabidopsis and rice. However, functional analysis in true apomictic species lags far behind, mainly due to the complexity of its genomes, of the trait itself, and the lack of efficient genetic transformation protocols. In this study, we review the current status of the in vitro culture and genetic transformation methods focusing on apomictic grasses, and the prospects for the application of new tools assayed in other related species, with two aims: to pave the way for discovering the molecular pathways involved in apomixis and to develop new capacities for breeding purposes because many of these grasses are important forage or biofuel resources.Fil: Bellido, Andres.     Lentivector Transduction Improves Outcomes Over Transplantation of Human HSCs Alone in NOD/SCID/Fabry Mice

    Fabry disease is a lysosomal storage disorder caused by a deficiency of a-galactosidase A (a-gal A) activity that results in progressive globotriaosylceramide (Gb(3)) deposition. We created a fully congenic nonobese diabetic (NOD)/severe combined immunodeficiency (SCID)/Fabry murine line to facilitate the in vivo assessment of human cell-directed therapies for Fabry disease. This pure line was generated after 11 generations of backcrosses and was found, as expected, to have a reduced immune compartment and background a-gal A activity. Next, we transplanted normal human CD34(+) cells transduced with a control (lentiviral vector-enhanced green fluorescent protein (LV-eGFP)) or a therapeutic bicistronic LV (LV-a-gal A/internal ribosome entry site (IRES)/hCD25). While both experimental groups showed similar engraftment levels, only the therapeutic group displayed a significant increase in plasma a-gal A activity. Gb(3) quantification at 12 weeks revealed metabolic correction in the spleen, lung, and liver for both groups. Importantly, only in the therapeutically-transduced cohort was a significant Gb(3) reduction found in the heart and kidney, key target organs for the amelioration of Fabry disease in humans.Fil: Pacienza, Natalia Alejandra.     Maternal taurine supplementation in rats partially prevents the adverse effects of early-life protein deprivation on b-cell function and insulin sensitivity

    Dietary protein restriction during pregnancy and lactation in rats impairs b-cell function and mass in neonates and leads to glucose intolerance in adult offspring. Maternal taurine (Tau) supplementation during pregnancy in rats restores b-cell function and mass in neonates, but its long-term effects are unclear. The prevention of postnatal catch-up growth has been suggested to improve glucose tolerance in adult offspring of low-protein (LP)-fed mothers. The objective of this study was to examine the relative contribution of b-cell dysfunction and insulin resistance to impaired glucose tolerance in 130-day-old rat offspring of LP-fed mothers and the effects of maternal Tau supplementation on b-cell function and insulin resistance in these offspring. Pregnant rats were fed i) control, ii) LP, and iii) LPCTau diets during gestation and lactation. Offspring were given a control diet following weaning. A fourth group consisting of offspring of LP-fed mothers, maintained on a LP diet following weaning, was also studied (LP-all life). Insulin sensitivity in the offspring of LP-fed mothers was reduced in females but not in males. In both genders, LP exposure decreased b-cell function. Tau supplementation improved insulin sensitivity in females and b-cell function in males. The LP-all life diet improved b-cell function in males. We conclude that i) maternal Tau supplementation has persistent effects on improving glucose metabolism (b-cell function and insulin sensitivity) in adult rat offspring of LP-fed mothers and ii) increasing the amount of protein in the diet of offspring adapted to a LP diet after weaning may impair glucose metabolism (b-cell function) in a gender-specific manner.Fil: Tang, Christine.     Recurrence and outcomes of Stevens-Johnson syndrome and toxic epidermal necrolysis in children

    OBJECTIVES: To report clinical course, etiology, management, and long-term outcomes of children suffering from Stevens-Johnson syndrome (SJS) or toxic epidermal necrolysis (TEN). METHODS: We conducted a study of all pediatric patients with SJS or TEN admitted between 2000 and 2007 to the Hospital for Sick Children and Children's Hospital Boston, and particular attention was paid to clinical manifestations, etiology, mortality, and long-term outcomes. RESULTS: We identified 55 cases of SJS (n = 47), TEN (n = 5), or SJS/TEN overlap syndrome (n - 3). Drugs were identified as the most likely etiologic agent in 29 children (53%); antiepileptic drugs were the most common agents (n = 16), followed by sulfonamide antibiotics (n = 7) and chemotherapy drugs (n = 2). Acute Mycoplasma pneumoniae infection was confirmed in 12 children (22%), and herpes simplex virus was confirmed in 5 children (9%). Treatment regimens differed significantly between participating sites and included systemic antimicrobial agents (67%), systemic corticosteroids (40%), and antiviral drugs (31%). Intravenous immunoglobulin was administered to 21 children (38%), of whom 8 received concomitant systemic corticosteroids. Ten children (18%) had recurrence of SJS up to 7 years after the index episode, and 3 experienced multiple recurrences. Twenty-six children (47%) suffered long-term sequelae that mostly involved the skin and eyes. CONCLUSIONS: Mortality rate in children was lower than that reported in adults, but half of affected children suffered long-term complications. The recurrence rate of SJS was high (1 in 5), which suggests vulnerability and potential genetic predisposition.     ENGLISH: The Inter-American Tropical Tuna Commission (IATTC) operates under the authority and direction of a convention originally entered into by Costa Rica and the United States. The convention, which came into force in 1950, is open to adherence by other governments whose nationals fish for tropical tunas and tuna-like species in the eastern Pacific Ocean (EPO). Under this provision Panama adhered in 1953, Ecuador in 1961, Mexico in 1964, Canada in 1968, Japan in 1970, France and Nicaragua in 1973, Vanuatu in 1990, Venezuela in 1992, El Salvador in 1997, Guatemala in 2000, Peru in 2002, Spain in 2003, the Republic of Korea in 2005, and Colombia in 2007. Canada withdrew from the IATTC in 1984. The IATTC's responsibilities are met with two programs, the Tuna-Billfish Program and the Tuna- Dolphin Program. SPANISH: La Comisión Interamericana del Atún Tropical (CIAT) funciona bajo la autoridad y dirección de una convención suscrita originalmente por Costa Rica y los Estados Unidos de América. La Convención, vigente desde 1950, está abierta a la afiliación de cualquier país cuyos ciudadanos pesquen atunes tropicales y especies afines en el Océano Pacífico oriental (OPO). Bajo esta estipulación, la República de Panamá se afilió en 1953, Ecuador en 1961, México en 1964, Canadá en 1968, Japón en 1970, Francia y Nicaragua en 1973, Vanuatu en 1990, Venezuela en 1992, El Salvador en 1997, Guatemala en 2000, Perú en 2002, España en 2003, la República de Corea en 2005, y Colombia en 2007. Con respecto a la posible independencia de Quebec, el artículo habla a favor de un Canadá unido sin miedo de ofender a los québécois. Y se desprenden dos conclusiones: una emocional y otra racional. La emocional nos recuerda las palabras del profesor Russell: “Amo a Canadá”, “Canadá es un gran país”. En la racional se nos recuerda las trágicas consecuencias de la desintegración de imperios y naciones. Y se afirma que los québécois no van a vivir mejor si se separan del resto de Canadá. Lo que se necesita verdaderamente es un federalismo más interactivo y más económicamente independiente de los Estados Unidos. This article examines the history of the linguistic conflict between Francophones and Anglophones within the double cultural reality of Canada and shows the English imperialism towards the Quebecois throughout their history. With regard to the possible independence of Quebec, the article discusses Canadian unity without fear of offending the Quebecois. Two conclusions stand out: one emotional and another rational. The emotional one reminds us of Professor Russell’s words: “I love Canada”, “Canada is a great place”. The rational conclusion reminds us of the tragic consequences of the disintegration of empires and nations. It is asserted that the Quebecois will not live better if they separate from the rest of Canada. What the country really needs is a federalism, more interactive and more economically independent from the United States
