    The value of local food partnerships: Covid and beyond

    The Covid-19 pandemic, and – more recently – soaring food prices have focused attention on how local areas meet the challenges of a fractured food system. This report examines the impacts andachievements of Local Food Partnerships (LFPs) and how LFPs embed and amplify their work to deliver both local and national food priorities. LFPs have been uniquely placed to provide systems leadership and practical solutions through the strategic direction and support of the UK-wide Sustainable Food Places (SFP) programme, established a decade prior to the pandemic. LFPs have been able to pivot to respond with agility to an extended period of national crisis and have moved forward to offer a coherent framework for the transition of local food system. The four dimensions of ‘effectiveness’,‘efficiency’, ‘engagement’, and ‘equity’ highlight the value of LFPs to fill the leadership gap on local food issues

    Augmented classification for electrical coil winding defects

    A green revolution has accelerated over the recent decades with a look to replace existing transportation power solutions through the adoption of greener electrical alternatives. In parallel the digitisation of manufacturing has enabled progress in the tracking and traceability of processes and improvements in fault detection and classification. This paper explores electrical machine manufacture and the challenges faced in identifying failures modes during this life cycle through the demonstration of state-of-the-art machine vision methods for the classification of electrical coil winding defects. We demonstrate how recent generative adversarial networks can be used to augment training of these models to further improve their accuracy for this challenging task. Our approach utilises pre-processing and dimensionality reduction to boost performance of the model from a standard convolutional neural network (CNN) leading to a significant increase in accuracy

    Embodying entrepreneurship: everyday practices, processes and routines in a technology incubator

    The growing interest in the processes and practices of entrepreneurship has been dominated by a consideration of temporality. Through a thirty-six-month ethnography of a technology incubator, this thesis contributes to extant understanding by exploring the effect of space. The first paper explores how class structures from the surrounding city have appropriated entrepreneurship within the incubator. The second paper adopts a more explicitly spatial analysis to reveal how the use of space influences a common understanding of entrepreneurship. The final paper looks more closely at the entrepreneurs within the incubator and how they use visual symbols to develop their identity. Taken together, the three papers reject the notion of entrepreneurship as a primarily economic endeavour as articulated through commonly understood language and propose entrepreneuring as an enigmatic attractor that is accessed through the ambiguity of the non-verbal to develop the ‘new’. The thesis therefore contributes to the understanding of entrepreneurship and proposes a distinct role for the non-verbal in that understanding

    La distrofia miotónica tipo 1 es una enfermedad genética rara multisistémica que afecta a 1 de cada 3000-8000 personas. La causa molecular de la enfermedad proviene de repeticiones tóxicas “CTG” en el gen DMPK (DM Protein Kinase). Tras la transcripción, estas repeticiones forman una estructura de horquilla que se une con alta afinidad a la familia de proteínas MBNL (Muscleblind-like) que agota su función de regulación de la poliadenilación y el splicing alternativo postranscripcional en numerosos transcritos. La pérdida de función de MBNL provoca una cascada de efectos posteriores, que eventualmente conducen a síntomas clínicos que incluyen miotonía, debilidad y atrofia muscular, cataratas, disfunción cardíaca y trastorno cognitivo. La restauración de la función de la proteína MBNL es clave para aliviar los síntomas debilitantes de esta enfermedad. Se han utilizado oligonucleótidos antisentido (AON) para apuntar a las repeticiones de DMPK y liberar MBNL del secuestro, lo que da como resultado resultados terapéuticos prometedores en modelos celulares y animales de la enfermedad. Otro factor que interviene en la pérdida de función de las proteínas MBNL son los miRNAs que regulan su traducción. Aquí se muestra el uso de AON dirigidos a la actividad de miR-23b y miR-218, que se ha demostrado previamente que regulan directamente MBNL1 y MBNL2. Estos antimiRs recibieron modificaciones FANA para aumentar su entrega en las células y reducir la toxicidad. También se probaron los AON, denominados blockmiRs, que se unen de manera complementaria a los sitios de unión confirmados de miR-23b y miR-218 en los 3'-UTR de las transcripciones de MBNL1 y MBNL2. De esta manera, los miRNAs no pueden unirse y regular la traducción de MBNL, lo que aumenta la cantidad de proteína MBNL producida en una célula deficiente. Aquí se propone el uso de AON de nuevo diseño dirigidos a la actividad de miR-23b y miR-218 para regular MBNL1 y MBNL2 a través de (1) exploración del bloqueo de miRNA a través de FANA-antimiR AON in vitro, (2) exploración del bloqueo del sitio de unión de miRNA a través de la estrategia blockmiR in vitro e in vivo con el uso de modificaciones químicas de LNA, y (3) mejora de la química de la estrategia blockmiR mediante el uso de tecnología de péptidos de penetración celular in vitro e in vivo.Myotonic Dystrophy Type 1 is a multi-systemic rare genetic disease affecting 1 in 3000-8000 people. The molecular cause of the disease stems from toxic “CTG” repetitions in the DMPK (DM Protein Kinase) gene. Upon transcription, these repetitions form a hairpin structure that binds with high affinity to the MBNL (Muscleblind-like) family of proteins depleting their function of post-transcriptional alternative splicing and polyadenylation regulation on numerous transcripts. MBNL loss-of-function causes a cascade of downstream effects, which eventually lead to clinical symptoms including myotonia, muscle weakness and atrophy, cataracts, cardiac dysfunction, and cognitive disorder. The restoration of MBNL protein function is key to relieving the debilitating symptoms of this disease. Antisense oligonucleotides (AONs) have been used to target the DMPK repeats and release MBNL from sequestration resulting in promising therapeutic results in cellular and animal models of the disease. Another factor playing a role in the loss-of-function of MBNL proteins are the miRNAs that regulate their translation. Here is shown the use of AONs targeting miR-23b and miR-218 activity, which have been previously shown to directly regulate MBNL1 and MBNL2. These antimiRs were given FANA modifications to increase their delivery in cells and lower toxicity. Also tested are AONs, termed blockmiRs, that complementary bind to the confirmed binding sites of miR-23b and miR-218 in the 3’-UTRs of MBNL1 and MBNL2 transcripts. In this way, the miRNAs are unable to bind and regulate the translation of MBNL thereby augmenting the amount of MBNL protein made in an otherwise deficient cell. Proposed here is the use of newly designed AONs targeting miR-23b and miR-218 activity in order to regulate MBNL1 and MBNL2 through (1) exploration of miRNA blocking through FANA-antimiR AONs in vitro, (2) exploration of miRNA binding site blocking through blockmiR strategy in vitro and in vivo with the use of LNA chemical modifications, and (3) improvement of the chemistry of the blockmiR strategy through the use of cell penetrating peptide technology in vitro and in vivo

    An investigation of the geothermal potential of the Upper Devonian sandstones beneath eastern Glasgow

    The urban development of the city of Glasgow is a consequence of its economic development, in part fuelled by local coalfields which exploited rocks in the same sedimentary basin within which geothermal resources in flooded abandoned mine workings, and deeper hot sedimentary aquifers (HSA), are present. This creates an opportunity to provide geothermal heating to areas of dense urban population with high heat demand. The depth of the target HSA geothermal resource, in Upper Devonian aged sandstones of the Stratheden Group, beneath eastern Glasgow was determined by gravity surveying and structural geological modelling. The estimated depth of the geothermal resource ranged from c.1500-2000 m, in the eastward deepening sedimentary basin. To reliably estimate the temperature of the geothermal resource, rigorous corrections to account for the effects of palaeoclimate and topography on heat flow were applied to boreholes in the Greater Glasgow area. The mean regional corrected heat flow was calculated as 75.7 mW m-2, an increase of 13.8 mW m-2 from the uncorrected value of 61.9 mW m-2, emphasising the extent to which heat flow was previously underestimated. Extrapolation of the geothermal gradient, calculated from the mean regional corrected heat flow, results in aquifer temperatures of c. 64-79 °C at depths of c.1500-2000 m beneath eastern Glasgow. The geothermal resource may, therefore, be capable of supporting a wide variety of direct heat use applications if sufficient matrix permeability or fracture networks are present. However, diagenetic effects such as quartz and carbonate cementation were found to restrict the porosity in Upper Devonian sandstones in a borehole and outcrop analogue study. These effects may likewise reduce porosity and intergranular permeability in the target aquifer, although this crucial aspect cannot be fully understood without deep exploratory drilling. To quantify the magnitude of the deep geothermal resource, the indicative thermal power outputs of geothermal doublet wells located in Glasgow’s East End were calculated for the first time, with outputs ranging from 1.3-2.1 MW dependent upon the aquifer depth. This, however, is predicated upon an aquifer permeability of c. 40 mD, which if reduced to 10 mD or less due to the effects of diagenesis, significantly reduces the thermal power outputs to 230-390 kW. The lack of assured project-success, given uncertainties related to the aquifer properties at depth, coupled with high capital costs of drilling, pose barriers to the development of deep geothermal energy in Glasgow. Further investigation of the economic viability of geothermal exploration, and alternative technological solutions is therefore required to mitigate the technical and economic risks. However, if sufficient matrix permeability or fracture networks are present at depth in the Upper Devonian sandstone sequence, then the potential contribution that geothermal energy could make to meeting local heat demand, reducing greenhouse gas emissions, and addressing the ‘energy trilemma’ in Glasgow is significant

    DeviceD: Audience–dancer interaction via social media posts and wearable for haptic feedback

    The performative installation DeviceD utilizes a network of systems toward facilitating interaction between dancer, digital media, and audience. Central to the work is a wearable haptic feedback system able to wirelessly deliver vibrotactile stimuli, with the latter initiated by the audience through posting on Twitter social media platform; the system in use searches for specific mentions, hashtags, and keywords, with positive results causing the system to trigger patterns of haptic biofeedback across the wearable’s four actuator motors. The system acts as the intermediator between the audience’s online actions and the dancer receiving physical stimuli; the dancer interprets these biofeedback signals according to Laban’s Effort movement qualities, with the interpretation informing different states of habitual and conscious choreographic performance. In this article, the authors reflect on their collaborative process while developing DeviceD alongside a multidisciplinary team of technologists, detailing their experience of refining the technology and methodology behind the work while presenting it in three different settings. A literature review is used to situate the work among contemporary research on interaction over internet and haptics in performance practice; haptic feedback devices have been widely used within artistic work for the past 25 years, with more recent practice and research outputs suggesting an increased interest for haptics in the field of dance research. The authors detail both technological and performative elements making up the work, and provide a transparent evaluation of the system, as means of providing a foundation for further research on wearable haptic devices

    Interactive Sonic Environments: Sonic artwork via gameplay experience

    The purpose of this study is to investigate the use of video-game technology in the design and implementation of interactive sonic centric artworks, the purpose of which is to create and contribute to the discourse and understanding of its effectiveness in electro-acoustic composition highlighting the creative process. Key research questions include: How can the language of electro-acoustic music be placed in a new framework derived from videogame aesthetics and technology? What new creative processes need to be considered when using this medium? Moreover, what aspects of 'play' should be considered when designing the systems? The findings of this study assert that composers and sonic art practitioners need little or no coding knowledge to create exciting applications and the myriad of options available to the composer when using video-game technology is limited only by imagination. Through a cyclic process of planning, building, testing and playing these applications the project revealed advantages and unique sonic opportunities in comparison to other sonic art installations. A portfolio of selected original compositions, both fixed and open are presented by the author to complement this study. The commentary serves to place the work in context with other practitioners in the field and to provide compositional approaches that have been taken
