    Light-regulated Gene Expression in Bacteria : Fundamentals, Advances, and Perspectives

    Numerous photoreceptors and genetic circuits emerged over the past two decades and now enable the light-dependent i.e., optogenetic, regulation of gene expression in bacteria. Prompted by light cues in the near-ultraviolet to near-infrared region of the electromagnetic spectrum, gene expression can be up- or downregulated stringently, reversibly, non-invasively, and with precision in space and time. Here, we survey the underlying principles, available options, and prominent examples of optogenetically regulated gene expression in bacteria. While transcription initiation and elongation remain most important for optogenetic intervention, other processes e.g., translation and downstream events, were also rendered light-dependent. The optogenetic control of bacterial expression predominantly employs but three fundamental strategies: light-sensitive two-component systems, oligomerization reactions, and second-messenger signaling. Certain optogenetic circuits moved beyond the proof-of-principle and stood the test of practice. They enable unprecedented applications in three major areas. First, light-dependent expression underpins novel concepts and strategies for enhanced yields in microbial production processes. Second, light-responsive bacteria can be optogenetically stimulated while residing within the bodies of animals, thus prompting the secretion of compounds that grant health benefits to the animal host. Third, optogenetics allows the generation of precisely structured, novel biomaterials. These applications jointly testify to the maturity of the optogenetic approach and serve as blueprints bound to inspire and template innovative use cases of light-regulated gene expression in bacteria. Researchers pursuing these lines can choose from an ever-growing, versatile, and efficient toolkit of optogenetic circuits

    Innovation and failure in mechatronics design education

    Innovative engineering design always has associated with it the risk of failure, and it is the role of the design engineer to mitigate the possibilities of failure in the final system. Education should however provide a safe space for students to both innovate and to learn about and from failures. However, pressures on course designers and students can result in their adopting a conservative, and risk averse, approach to problem solving. The paper therefore considers the nature of both innovation and failure, and looks at how these might be effectively combined within mechatronics design education

    Systematic innovation and the underlying principles behind TRIZ and TOC

    Innovative developments in the design of product and manufacturing systems are often marked by simplicity, at least in retrospect, that has previously been shrouded by restrictive mental models or limited knowledge transfer. These innovative developments are often associated with the breaking of long established trade-off compromises, as in the paradigm shift associated with JIT & TQM, or the resolution of design contradictions, as in the case of the dual cyclone vacuum cleaner. The rate of change in technology and the commercial environment suggests the opportunity for innovative developments is accelerating, but what systematic support is there to guide this innovation process. This paper brings together two parallel, but independent theories on inventive problem solving; one in mechanical engineering, namely the Russian Theory of Inventive Problem Solving (TRIZ) and the other originating in manufacturing management as the Theory of Constraints (TOC). The term systematic innovation is used to describe the use of common underlying principles within these two approaches. The paper focuses on the significance of trade-off contradictions to innovation in these two fields and explores their relationship with manufacturing strategy development

    Beyond enterprise resource planning projects: innovative strategies for competitive advantage

    ABSTRACT A rapidly changing business environment and legacy IT problems has resulted in many organisations implementing standard package solutions. This 'common systems' approach establishes a common IT and business process infrastructure within organisations and its increasing dominance raises several important strategic issues. These are to what extent do common systems impose common business processes and management systems on competing firms, and what is the source of competitive advantage if the majority of firms employ almost identical information systems and business processes? A theoretical framework based on research into legacy systems and earlier IT strategy literature is used to analyse three case studies in the manufacturing, chemical and IT industries. It is shown that the organisations are treating common systems as the core of their organisations' abilities to manage business transactions. To achieve competitive advantage they are clothing these common systems with information systems designed to capture information about competitors, customers and suppliers, and to provide a basis for sharing knowledge within the organisation and ultimately with economic partners. The importance of these approaches to other organisations and industries is analysed and an attempt is made at outlining the strategic options open to firms beyond the implementation of common business systems

    How innovation systems emerge to solve ecological problems: Biofuels in the United States and Brazil

    This paper discusses the re-emergence of biofuel innovation systems in the United States and Brazil. We argue that innovation systems emerge and evolve to solve a problem, and that the way the problem is framed and articulated has a significant impact on the direction and momentum of this evolution. Additionally, innovation sequences occur with a recurrent pattern of changing problems and innovative solutions. We consider the role of the State as a core actor in the mobilisation of innovation systems and discuss how specific institutional arrangements, political contexts and technological competencies influence how problems are framed. We find that role of the State varies across time as well as across different geographical regions. Finally, we suggest that as ecological problems intensify we might expect to see an increase in State intervention in innovation systems

    The impact of Digital Platforms on Business Models: an empirical investigation on innovative start-ups

    Digital platforms have the ability to connect people, organizations and resources with the aim of facilitating the core interactions between businesses and consumers as well as assuring a greater efficiency for the business management. New business concepts, such as innovative start-ups, are therefore created based on innovation, scalability and the relationships within the community around them. The purpose of this work is to deeply understand the evolution of business models brought by innovative and dynamic companies operating through online platforms. In order to achieve the objectives set, an exploratory multiple-case study was designed based on in-depth structured interviews. The aim was to conduct a mixed analysis, in order to rely both on qualitative and quantitative data. The structured interview protocol was therefore designed to collect and then analyse data concerning the company profile and managers’ perspectives on the phenomenon of interest. The interview protocol was submitted in advance and then face-to-face interviews were carried out with the following professional figures: Chief Executive Officer (CEO), General Manager, Chief Technology Officer (CTO), Marketing Manager and Developers. Collected data were analysed and processed through the Canvas Business Model in order to clearly outline similarities and differences among the sample. Results can be considered under two viewpoints. On the one hand, this work provides a detailed overview of the companies interviewed, according to the dimensions of: reference market dynamics, type and number of customers, scalability. On the other one, they allow to identify some success patterns regarding key activities, key resources, channel mix strategy, costs management, value proposition, customer segmentation, key partners and the way to obtain revenues. Results from the multiple-case study with 15 Italian start-ups provide interesting insights by comparing the innovative business models developed and highlighting key differences and similarities. verall, the start-ups analyzed, operating in several sectors, showed great growth prospects and the possibility to create value for their customers through innovative products and services offered through digital platforms
