    Learning from Complex Neuroimaging Datasets

    Advancements in Magnetic Resonance Imaging (MRI) allowed for the early diagnosis of neurodevelopmental disorders and neurodegenerative diseases. Neuroanatomical abnormalities in the cerebral cortex are often investigated by examining group-level differences of brain morphometric measures extracted from highly-sampled cortical surfaces. However, group-level differences do not allow for individual-level outcome prediction critical for the application to clinical practice. Despite the success of MRI-based deep learning frameworks, critical issues have been identified: (1) extracting accurate and reliable local features from the cortical surface, (2) determining a parsimonious subset of cortical features for correct disease diagnosis, (3) learning directly from a non-Euclidean high-dimensional feature space, (4) improving the robustness of multi-task multi-modal models, and (5) identifying anomalies in imbalanced and heterogeneous settings. This dissertation describes novel methodological contributions to tackle the challenges above. First, I introduce a Laplacian-based method for quantifying local Extra-Axial Cerebrospinal Fluid (EA-CSF) from structural MRI. Next, I describe a deep learning approach for combining local EA-CSF with other morphometric cortical measures for early disease detection. Then, I propose a data-driven approach for extending convolutional learning to non-Euclidean manifolds such as cortical surfaces. I also present a unified framework for robust multi-task learning from imaging and non-imaging information. Finally, I propose a semi-supervised generative approach for the detection of samples from untrained classes in imbalanced and heterogeneous developmental datasets. The proposed methodological contributions are evaluated by applying them to the early detection of Autism Spectrum Disorder (ASD) in the first year of the infant’s life.     An economic development model analyzes the adoption of alternative strategy capable of leveraging the economy, based essentially on RES. The combination of wind turbine, PV installation with new technology battery energy storage, DSM network and RES forecasting algorithms maximizes RES integration in isolated islands. An innovative model of power system (PS) imbalances is presented, which aims to capture various features of the stochastic behavior of imbalances and to reduce in average reserve requirements and PS risk. Deep learning techniques for medium-term wind speed and solar irradiance forecasting are presented, using for first time a specific cloud index. Scalability-replicability of the FLEXITRANSTORE technology innovations integrates hardware-software solutions in all areas of the transmission system and the wholesale markets, promoting increased RES. A deep learning and GIS approach are combined for the optimal positioning of wave energy converters. An innovative methodology to hybridize battery-based energy storage using supercapacitors for smoother power profile, a new control scheme and battery degradation mechanism and their economic viability are presented. An innovative module-level photovoltaic (PV) architecture in parallel configuration is introduced maximizing power extraction under partial shading. A new method for detecting demagnetization faults in axial flux permanent magnet synchronous wind generators is presented.     Mechanisms of chaperonin-assisted protein folding

    The bacterial chaperonin GroEL-GroES promotes protein folding through ATP-regulated cycles of substrate protein binding, encapsulation, and release. Here, we have used recent advances in cryoEM to determine cryoEM structures of GroEL, GroEL-ADP·BeFx, and GroEL-GroESADP·AlFx, all bound to the model substrate Rhodospirillum rubrum RuBisCO in a non-native state. The use of the Chameleon vitrification instrument reduced preferred orientation and air-water interface denaturation effects in cryoEM. Previous cryoEM reconstructions of GroEL-substrate complexes have been limited in resolution, and there are no previously reported structures of GroEL-ATP (or ATP analogue) bound to a non-native substrate. Our structures show how the conformation and interactions of a non-native substrate protein change as GroEL proceeds through its reaction cycle. We observe specific charged and hydrophobic GroEL residues forming strong initial contacts with non-native RuBisCO. Binding of an ATP analogue results in an enhanced interaction of substrate with the disordered C-terminal tails of GroEL. The cryoEM structure of GroEL-ADP·BeFx-RuBisCO displays striking asymmetry in the substrate-occupied ring. We observe individual GroEL subunits fulfilling different functions. Four of the GroEL subunits sequester non-native substrate inside the cavity while the remaining three adopt an extended conformation allowing GroEL to efficiently recruit GroES. Our structure explains how GroEL can recruit GroES without releasing bound substrate. The cryoEM structure of GroELGroES-RuBisCO shows a native-like RuBisCO interacting weakly with the walls of GroEL and the inner face of GroES. Additionally, the trans ring of our nucleotide bound structures show a novel conformation that may be related to the negative cooperativity of non-native RuBisCObinding

The survey was defined with the aim to involve the highest possible number of relevant CSR topics and give the issue a more wholesome perspective. It provides a basis for further comprehension and deeper analyses of specific CSR areas. The conditions determining the success of CSR in Romania have been defined in the paper on the basis of the previously cumulative knowledge as well as the results of various researches.     Augmentation of Brain Function: Facts, Fiction and Controversy. Volume III: From Clinical Applications to Ethical Issues and Futuristic Ideas

    The final volume in this tripartite series on Brain Augmentation is entitled “From Clinical Applications to Ethical Issues and Futuristic Ideas”. Many of the articles within this volume deal with translational efforts taking the results of experiments on laboratory animals and applying them to humans. In many cases, these interventions are intended to help people with disabilities in such a way so as to either restore or extend brain function. Traditionally, therapies in brain augmentation have included electrical and pharmacological techniques. In contrast, some of the techniques discussed in this volume add specificity by targeting select neural populations. This approach opens the door to where and how to promote the best interventions. Along the way, results have empowered the medical profession by expanding their understanding of brain function. Articles in this volume relate novel clinical solutions for a host of neurological and psychiatric conditions such as stroke, Parkinson’s disease, Huntington’s disease, epilepsy, dementia, Alzheimer’s disease, autism spectrum disorders (ASD), traumatic brain injury, and disorders of consciousness. In disease, symptoms and signs denote a departure from normal function. Brain augmentation has now been used to target both the core symptoms that provide specificity in the diagnosis of a disease, as well as other constitutional symptoms that may greatly handicap the individual. The volume provides a report on the use of repetitive transcranial magnetic stimulation (rTMS) in ASD with reported improvements of core deficits (i.e., executive functions). TMS in this regard departs from the present-day trend towards symptomatic treatment that leaves unaltered the root cause of the condition. In diseases, such as schizophrenia, brain augmentation approaches hold promise to avoid lengthy pharmacological interventions that are usually riddled with side effects or those with limiting returns as in the case of Parkinson’s disease. Brain stimulation can also be used to treat auditory verbal hallucination, visuospatial (hemispatial) neglect, and pain in patients suffering from multiple sclerosis. The brain acts as a telecommunication transceiver wherein different bandwidth of frequencies (brainwave oscillations) transmit information. Their baseline levels correlate with certain behavioral states. The proper integration of brain oscillations provides for the phenomenon of binding and central coherence. Brain augmentation may foster the normalization of brain oscillations in nervous system disorders. These techniques hold the promise of being applied remotely (under the supervision of medical personnel), thus overcoming the obstacle of travel in order to obtain healthcare. At present, traditional thinking would argue the possibility of synergism among different modalities of brain augmentation as a way of increasing their overall effectiveness and improving therapeutic selectivity. Thinking outside of the box would also provide for the implementation of brain-to-brain interfaces where techniques, proper to artificial intelligence, could allow us to surpass the limits of natural selection or enable communications between several individual brains sharing memories, or even a global brain capable of self-organization. Not all brains are created equal. Brain stimulation studies suggest large individual variability in response that may affect overall recovery/treatment, or modify desired effects of a given intervention. The subject’s age, gender, hormonal levels may affect an individual’s cortical excitability. In addition, this volume discusses the role of social interactions in the operations of augmenting technologies. Finally, augmenting methods could be applied to modulate consciousness, even though its neural mechanisms are poorly understood. Finally, this volume should be taken as a debate on social, moral and ethical issues on neurotechnologies. Brain enhancement may transform the individual into someone or something else. These techniques bypass the usual routes of accommodation to environmental exigencies that exalted our personal fortitude: learning, exercising, and diet. This will allow humans to preselect desired characteristics and realize consequent rewards without having to overcome adversity through more laborious means. The concern is that humans may be playing God, and the possibility of an expanding gap in social equity where brain enhancements may be selectively available to the wealthier individuals. These issues are discussed by a number of articles in this volume. Also discussed are the relationship between the diminishment and enhancement following the application of brain-augmenting technologies, the problem of “mind control” with BMI technologies, free will the duty to use cognitive enhancers in high-responsibility professions, determining the population of people in need of brain enhancement, informed public policy, cognitive biases, and the hype caused by the development of brain- augmenting approaches
