    Implantable Neural Probes for Brain-Machine Interfaces - Current Developments and Future Prospects

    A Brain-Machine interface (BMI) allows for direct communication between the brain and machines. Neural probes for recording neural signals are among the essential components of a BMI system. In this report, we review research regarding implantable neural probes and their applications to BMIs. We first discuss conventional neural probes such as the tetrode, Utah array, Michigan probe, and electroencephalography (ECoG), following which we cover advancements in next-generation neural probes. These next-generation probes are associated with improvements in electrical properties, mechanical durability, biocompatibility, and offer a high degree of freedom in practical settings. Specifically, we focus on three key topics: (1) novel implantable neural probes that decrease the level of invasiveness without sacrificing performance, (2) multi-modal neural probes that measure both electrical and optical signals, (3) and neural probes developed using advanced materials. Because safety and precision are critical for practical applications of BMI systems, future studies should aim to enhance these properties when developing next-generation neural probes

    Comparison of Sub-Scalp EEG and Endovascular Stent-Electrode Array for Visually Evoked Potential Brain-Computer Interface

    Brain-computer interfaces (BCI) have the potential to improve the quality of life for persons with paralysis. Sub-scalp EEG provides an alternative BCI signal acquisition method that compromises between the limitations of traditional EEG systems and the risks associated with intracranial electrodes, and has shown promise in long-term seizure monitoring. However, sub-scalp EEG has not yet been assessed for suitability in BCI applications. This study presents a preliminary comparison of visually-evoked potentials (VEPs) recorded using subscalp and endovascular stent electrodes in a sheep.     Strategies for neural control of prosthetic limbs: From electrode interfacing to 3D printing

    Limb amputation is a major cause of disability in our community, for which motorised prosthetic devices offer a return to function and independence. With the commercialisation and increasing availability of advanced motorised prosthetic technologies, there is a consumer need and clinical drive for intuitive user control. In this context, rapid additive fabrication/prototyping capacities and biofabrication protocols embrace a highly-personalised medicine doctrine that marries specific patient biology and anatomy to high-end prosthetic design, manufacture and functionality. Commercially-available prosthetic models utilise surface electrodes that are limited by their disconnect between mind and device. As such, alternative strategies of mind-prosthetic interfacing have been explored to purposefully drive the prosthetic limb. This review investigates mind to machine interfacing strategies, with a focus on the biological challenges of long-term harnessing of the user\u27s cerebral commands to drive actuation/movement in electronic prostheses. It covers the limitations of skin, peripheral nerve and brain interfacing electrodes, and in particular the challenges of minimising the foreign-body response, as well as a new strategy of grafting muscle onto residual peripheral nerves. In conjunction, this review also investigates the applicability of additive tissue engineering at the nerve-electrode boundary, which has led to pioneering work in neural regeneration and bioelectrode development for applications at the neuroprosthetic interface

    Central nervous system microstimulation: Towards selective micro-neuromodulation

    Electrical stimulation technologies capable of modulating neural activity are well established for neuroscientific research and neurotherapeutics. Recent micro-neuromodulation experimental results continue to explain neural processing complexity and suggest the potential for assistive technologies capable of restoring or repairing of basic function. Nonetheless, performance is dependent upon the specificity of the stimulation. Increasingly specific stimulation is hypothesized to be achieved by progressively smaller interfaces. Miniaturization is a current focus of neural implants due to improvements in mitigation of the body's foreign body response. It is likely that these exciting technologies will offer the promise to provide large-scale micro-neuromodulation in the future. Here, we highlight recent successes of assistive technologies through bidirectional neuroprostheses currently being used to repair or restore basic brain functionality. Furthermore, we introduce recent neuromodulation technologies that might improve the effectiveness of these neuroprosthetic interfaces by increasing their chronic stability and microstimulation specificity. We suggest a vision where the natural progression of innovative technologies and scientific knowledge enables the ability to selectively micro-neuromodulate every neuron in the brain

    Abstract: This paper looks at advancements made in the area of thought controlled mechanical prosthesis that are being developed for amputees in order for them to regain mobility. It focuses on the brain-machine interface which is hardware and software that is used to control mechanical prosthesis or bionic limbs by sending and receiving signals between the prosthetic and the users mind. There is signaling feedback from the prosthesis to the user that indicates how much pressure is being applied to an object that is being grasped for instance. This paper explores the notion of the value of pain as a warning in the form of artificial feedback to help prevent damage and death to people and posits that pain should be included in the feedback loop so that when, for example, an artificial hand is in imminent danger of being burned the wearer is alerted

    Motor neuroprosthesis implanted with neurointerventional surgery improves capacity for activities of daily living tasks in severe paralysis: first in-human experience

    Background: Implantable brainā€“computer interfaces (BCIs), functioning as motor neuroprostheses, have the potential to restore voluntary motor impulses to control digital devices and improve functional independence in patients with severe paralysis due to brain, spinal cord, peripheral nerve or muscle dysfunction. However, reports to date have had limited clinical translation. Methods Two participants with amyotrophic lateral sclerosis (ALS) underwent implant in a single-arm, open-label, prospective, early feasibility study. Using a minimally invasive neurointervention procedure, a novel endovascular Stentrode BCI was implanted in the superior sagittal sinus adjacent to primary motor cortex. The participants undertook machinelearning-assisted training to use wirelessly transmitted electrocorticography signal associated with attempted movements to control multiple mouse-click actions, including zoom and left-click. Used in combination with an eye-tracker for cursor navigation, participants achieved Windows 10 operating system control to conduct instrumental activities of daily living (IADL) tasks. Results: Unsupervised home use commenced from day 86 onwards for participant 1, and day 71 for participant 2. Participant 1 achieved a typing task average click selection accuracy of 92.63% (100.00%, 87.50%ā€“ 100.00%) (trial mean (median, Q1ā€“Q3)) at a rate of 13.81 (13.44, 10.96ā€“16.09) correct characters per minute (CCPM) with predictive text disabled. Participant 2 achieved an average click selection accuracy of 93.18% (100.00%, 88.19%ā€“100.00%) at 20.10 (17.73, 12.27ā€“26.50) CCPM. Completion of IADL tasks including text messaging, online shopping and managing finances independently was demonstrated in both participants.     Personalized computational models of deep brain stimulation

Advisor: Matthew Johnson. 1 computer file (PDF); xii, 138 pages.Deep brain stimulation (DBS) therapy is used for managing symptoms associated with a growing number of neurological disorders. One of the primary challenges with delivering this therapy, however, continues to be accurate neurosurgical targeting of the DBS lead electrodes and post-operative programming of the stimulation settings. Two approaches for addressing targeting have been advanced in recent years. These include novel DBS lead designs with more electrodes and computational models that can predict cellular modulation during DBS. Here, we developed a personalized computational modeling framework to (1) thoroughly investigate the electrode design parameter space for current and future DBS array designs, (2) generate and evaluate machine learning feature sets for semi-automated programming of DBS arrays, (3) study the influence of model parameters in predicting behavioral and electrophysiological outcomes of DBS in a preclinical animal model of Parkinsonā€™s disease, and (4) evaluate feasibility of a novel endovascular targeting approach to delivering DBS therapy in humans. These studies show how independent current controlled stimulation with advanced machine learning algorithms can negate the need for highly dense electrode arrays to shift, steer, and sculpt regions of modulation within the brain. Additionally, these studies show that while advanced and personalized computational models of DBS can predict many of the behavioral and electrophysiological outcomes of DBS, there are remaining inconsistencies that suggest there are additional physiological mechanisms of DBS that are not yet well understood. Finally, the results show how computational models can be beneficial for prospective development of novel approaches to neuromodulation prior to large-scale preclinical and clinical studies

    In the last year there has been increasing interest and investment into developing devices to interact with the central nervous system, in particular developing a robust brain-computer interface (BCI). In this article, we review the most recent research advances and the current host of engineering and neurological challenges that must be overcome for clinical application. In particular, space limitations, isolation of targeted structures, replacement of probes following failure, delivery of nanomaterials and processing and understanding recorded data. Neural engineering has developed greatly over the past half-century, which has allowed for the development of better neural recording techniques and clinical translation of neural interfaces. Implementation of general purpose BCIs face a number of constraints arising from engineering, computational, ethical and neuroscientific factors that still have to be addressed. Electronics have become orders of magnitude smaller and computationally faster than neurons, however there is much work to be done in decoding the neural circuits. New interest and funding from the non-medical community may be a welcome catalyst for focused research and development; playing an important role in future advancements in the neuroscience community
