    cellSTORM - Cost-effective Super-Resolution on a Cellphone using dSTORM

    Expensive scientific camera hardware is amongst the main cost factors in modern, high-performance microscopes. Recent technological advantages have, however, yielded consumer-grade camera devices that can provide surprisingly good performance. The camera sensors of smartphones in particular have benefited of this development. Combined with computing power and due to their ubiquity, smartphones provide a fantastic opportunity for "imaging on a budget". Here we show that a consumer cellphone is capable even of optical super-resolution imaging by (direct) Stochastic Optical Reconstruction Microscopy (dSTORM), achieving optical resolution better than 80 nm. In addition to the use of standard reconstruction algorithms, we investigated an approach by a trained image-to-image generative adversarial network (GAN). This not only serves as a versatile technique to reconstruct video sequences under conditions where traditional algorithms provide sub-optimal localization performance, but also allows processing directly on the smartphone. We believe that "cellSTORM" paves the way for affordable super-resolution microscopy suitable for research and education, expanding access to cutting edge research to a large community

    AirCode: Unobtrusive Physical Tags for Digital Fabrication

    We present AirCode, a technique that allows the user to tag physically fabricated objects with given information. An AirCode tag consists of a group of carefully designed air pockets placed beneath the object surface. These air pockets are easily produced during the fabrication process of the object, without any additional material or postprocessing. Meanwhile, the air pockets affect only the scattering light transport under the surface, and thus are hard to notice to our naked eyes. But, by using a computational imaging method, the tags become detectable. We present a tool that automates the design of air pockets for the user to encode information. AirCode system also allows the user to retrieve the information from captured images via a robust decoding algorithm. We demonstrate our tagging technique with applications for metadata embedding, robotic grasping, as well as conveying object affordances.Comment: ACM UIST 2017 Technical Paper

    The ePetri dish, an on-chip cell imaging platform based on subpixel perspective sweeping microscopy (SPSM)

    We report a chip-scale lensless wide-field-of-view microscopy imaging technique, subpixel perspective sweeping microscopy, which can render microscopy images of growing or confluent cell cultures autonomously. We demonstrate that this technology can be used to build smart Petri dish platforms, termed ePetri, for cell culture experiments. This technique leverages the recent broad and cheap availability of high performance image sensor chips to provide a low-cost and automated microscopy solution. Unlike the two major classes of lensless microscopy methods, optofluidic microscopy and digital in-line holography microscopy, this new approach is fully capable of working with cell cultures or any samples in which cells may be contiguously connected. With our prototype, we demonstrate the ability to image samples of area 6 mm × 4 mm at 660-nm resolution. As a further demonstration, we showed that the method can be applied to image color stained cell culture sample and to image and track cell culture growth directly within an incubator. Finally, we showed that this method can track embryonic stem cell differentiations over the entire sensor surface. Smart Petri dish based on this technology can significantly streamline and improve cell culture experiments by cutting down on human labor and contamination risks

    Includes bibliographical references (leaves 75-80).This media dissertation researches the cell phone's actual and potential role as a multimedia tool of communication amongst South African youth and looks at how it can be used to platform youth stories for media and advertising. The youth's connection to the cell phone has come to mean so much more than its actual technological functions. This media dissertation investigates the cell phone phenomenon amongst the youth of today, by looking at both local and international trends, with a more intimate focus on the current trends amongst the South African youth. It will look at the sociology of the cell phone and the culture surrounding it. It will then consider new technology and how the cell phone's role may also be a tool for leapfrogging in South Africa. It is also important to consider the negative connotations that arise with the cell phone's infiltration amongst the youth.Through analysis of recent research about the cell phone's impact on the youth here and internationally, two opposing media directions are identified: the cell phone as a tool in marketing and advertising; and the cell phone as a tool in investigative journalism. By analyzing two different areas, this media dissertation creates a broad and holistic understanding of the cell phone's potential functions through a strong literature review. Firstly, the cell phone's function in marketing and advertising will be analyzed. This media dissertation will take into account the youth market in South Africa through a case study of one of South Africa's strategic consultancy companies: Instant Grass. Through a close look at the youth market now, it will be possible to create a greater understanding of the current trends and how to capitalize on these trends. In terms of marketing and advertising, this media dissertation then discusses an advertising exercise with a youth group studying Film and Media Studies at the University of Cape Town. Secondly, the cell phone in terms of media and investigative journalism will be analysed through fieldwork done with etv's 3rd Degree. This media dissertation looks at how the cell phone could be used as a tool for youth stories by looking at the parameters involved in creating investigative stories. This chapter also takes a look at the issue of citizen journalism in the digital publishing world today and the rate at which cell phone technology is spurring this development on
