An assessment of the causes of the errors in the 2015 UK General Election opinion polls
The opinion polls undertaken prior to the 2015 UK General Election under-estimated the Conservative lead over Labour by an average of 7 percentage points. This collective failure led politicians and commentators to question the validity and utility of political polling and raised concerns regarding a broader public loss of confidence in survey research. In this paper, we assess the likely causes of the 2015 polling errors. We begin by setting out a formal account of the statistical methodology and assumptions required for valid estimation of party vote shares using quota sampling. We then describe the current approach of polling organisations for estimating sampling variability and suggest a new method based on bootstrap re-sampling. Next, we use poll micro-data to assess the plausibility of different explanations of the polling errors. Our conclusion is that the primary cause of the polling errors in 2015 was unrepresentative sampling

Finally, the expressions of novel DEGs were validated in Chinese PanNET tissues by RT-qPCR.ResultsFive new DEGs (ABCC8, PCSK2, IL13RA2, KLKB1, and PART1) and one confirmed DEG-ISL1 were identified. The mutation counts of DEGs increased with the tumor grade increasing from G1 to G3, and PanNET patients present vascular invasion or are deceased. These DEG expression patterns in PanNETs are quite different from that of pancreatic ductal adenocarcinoma and are related to A–D–M (ATRX–DAXX–MEN1) mutation. ABCC8 and KLKB1 are co-occurrence with the A–D–M axis in PanNETs. Importantly, patients with DEG mutations have a lower survival rate. RT-qPCR verification results of KLKB1 (P ConclusionFive novel common DEGs identified are related to the development and prognosis of PanNETs and may serve as specific biomarkers and therapeutic targets.</p

miR-199a-3p plays an anti-tumorigenic role in lung adenocarcinoma by suppressing anterior gradient 2
Previous studies have explored the association between protein-coding genes and microRNAs (miRNAs) in lung adenocarcinoma (LUAD). However, the influence of the miR-199a-3p/anterior gradient 2 (AGR2) axis in LUAD has not yet been fully explored. Therefore, this study aimed to examine the underlying roles of AGR2 and miR-199a-3p in the development of LUAD. The expression levels of miR-199a-3p and AGR2 in LUAD tissues and cells were detected via quantitative reverse transcription-polymerase chain reaction (qRT-PCR). A luciferase assay was also performed to identify the interaction between AGR2 and miR-199a-3p. Moreover, the cell counting kit 8 (CCK-8), 5ʹ-bromo-2ʹ-deoxyuridine (BrdU), and adhesion assays were used along with flow cytometry to verify the malignancy of LUAD in vitro, while a xenograft tumor assay was performed to confirm the tumor growth in vitro. The findings showed a decrease in the expression of miR-199a-3p in LUAD. Additionally, miR-199a-3p overexpression inhibited the growth of LUAD cells in vitro and in vivo, while elevating the apoptosis rate of the cells. AGR2 knockdown had the same effect in the cells as that of miR-199a-3p overexpression. It was also found that miR-199a-3p directly targeted AGR2 in LUAD cells to suppress tumorigenesis. In conclusion, this study suggests that miR-199a-3p plays an anti-tumorigenic role in LUAD by targeting AGR2. Moreover, our study provides insights into the development of novel therapeutic targets for the treatment of LUAD.</p
Structure and Optical Property Prediction of 2D Plasmonic Photonic Crystals Fabricated by Shadow Sphere Lithography
Shadow sphere lithography (SSL) is a powerful and large-scale
fabrication
method to produce two-dimensional (2D) plasmonic photonic crystals
and three-dimensional metamaterials. Practically, one of the biggest
challenges for SSL-based fabrications is that it is hard to accurately
predict the physical properties of the fabricated nanostructures if
the structures were only modeled by the geometric shadowing effect.
A Monte Carlo (MC) simulation is developed to show that the dynamic
shadowing effect due to the accumulation of materials on the template
as well as the thin-film growth mechanism plays a key role in determining
the structure details. For a one-to-three step-based SSL fabrication,
the nanostructures predicted by MC match very well with those produced
experimentally, and the plasmonic properties predicted by these MC-simulated
structures are also consistent with the features obtained experimentally,
both qualitative and semi-quantitative. This study indicates a possible
solution to use MC simulation and numerical calculation to guide the
design of the plasmonic photonic crystals and metamaterials based
on SSL for optic applications

BackgroundNeuroinflammation and neuronal injury have been reported to be associated with the development of postoperative delirium in both preclinical and clinical settings. This study aimed to investigate the potential correlation between biomarkers of neurofilament light chain and glial fibrillary acidic protein and emergence and postoperative delirium in elderly patients undergoing surgery.MethodsPatients who developed emergence delirium (n = 30) and postoperative delirium (n = 32), along with their matched controls, were enrolled after obtaining ethics approval and written informed consent. Delirium was assessed using the Confusion Assessment Method for the Intensive Care Unit or Confusion Assessment Method scale, and blood samples were collected before and after surgery for plasma neurofilament light chain and glial fibrillary acidic protein measurements using a single-molecule array.ResultsThe study found that in patients with emergence delirium, the increase in plasma neurofilament light chain protein levels during surgery was significantly higher than in non-delirium patients (P = 0.002). Additionally, in patients with postoperative delirium, both the increase in plasma neurofilament light chain protein levels (P ConclusionOur findings suggest that emergence delirium is linked to elevated levels of neurofilament light chain, a biomarker of axonal injury, during surgery. Furthermore, in addition to axonal injury, postoperative delirium was also associated with an increase in glial fibrillary acidic protein, a marker of astrocyte activation.</p

Cadmium (Cd) is a harmful heavy metal that is risky for plant growth and human health. The zinc-finger transcription factor ZAT10 is highly conserved with ZAT6 and ZAT12, which are involved in Cd tolerance in plants. However, the definite function of ZAT10 in Cd tolerance remains uncertain. Here, we demonstrated that ZAT10 negatively regulated Cd uptake and enhanced Cd detoxification in Arabidopsis. The expression of ZAT10 in plants is induced by Cd treatment. The zat10 mutant plants exhibited a greater sensitivity to Cd stress and accumulated more Cd in both shoot and root. Further investigations revealed that ZAT10 repressed the transcriptional activity of IRT1, which encodes a key metal transporter involved in Cd uptake. Meanwhile, ZAT10 positively regulated four heavy metal detoxification-related genes: NAS1, NAS2, IRT2, and MTP3. We further found that ZAT10 interacts with FIT, but their regulatory relationship is still unclear. In addition, ZAT10 directly bound to its own promoter and repressed its transcription as a negative feedback regulation. Collectively, our findings provided new insights into the dual functions of ZAT10 on Cd uptake and detoxification in plants and pointed to ZAT10 as a potential gene resource for Cd tolerance improvement in plants.</p
