TakeCARE, a Video Bystander Program to Help Prevent Sexual Violence on College Campuses: Results of Two Randomized, Controlled Trials
Objective: The present research reports on two randomized controlled trials evaluating TakeCARE, a video bystander program designed to help prevent sexual violence on college campuses. Method: In Study 1, students were recruited from psychology courses at two universities. In Study 2, first-year students were recruited from a required course at one university. In both studies, students were randomly assigned to view one of two videos: TakeCARE or a control video on study skills. Just before viewing the videos, students completed measures of bystander behavior toward friends and ratings of self-efficacy for performing such behaviors. The efficacy measure was administered again after the video, and both the bystander behavior measure and the efficacy measure were administered at either one (Study 1) or two (Study 2) months later. Results: In both studies, students who viewed TakeCARE, compared to students who viewed the control video, reported engaging in more bystander behavior toward friends and greater feelings of efficacy for performing such behavior. In Study 1, feelings of efficacy mediated effects of TakeCARE on bystander behavior; this result did not emerge in Study 2. Conclusions: This research demonstrates that TakeCARE, a video bystander program, can positively influence bystander behavior toward friends. Given its potential to be easily distributed to an entire campus community, TakeCARE might be an effective addition to campus efforts to prevent sexual violence
Viral complementation allows HIV-1 replication without integration
<p>Abstract</p> <p>Background</p> <p>The integration of HIV-1 DNA into cellular chromatin is required for high levels of viral gene expression and for the production of new virions. However, the majority of HIV-1 DNA remains unintegrated and is generally considered a replicative dead-end. A limited amount of early gene expression from unintegrated DNA has been reported, but viral replication does not proceed further in cells which contain only unintegrated DNA. Multiple infection of cells is common, and cells that are productively infected with an integrated provirus frequently also contain unintegrated HIV-1 DNA. Here we examine the influence of an integrated provirus on unintegrated HIV-1 DNA (uDNA).</p> <p>Results</p> <p>We employed reporter viruses and quantitative real time PCR to examine gene expression and virus replication during coinfection with integrating and non-integrating HIV-1. Most cells which contained only uDNA displayed no detected expression from fluorescent reporter genes inserted into early (Rev-independent) and late (Rev-dependent) locations in the HIV-1 genome. Coinfection with an integrated provirus resulted in a several fold increase in the number of cells displaying uDNA early gene expression and efficiently drove uDNA into late gene expression. We found that coinfection generates virions which package and deliver uDNA-derived genomes into cells; in this way uDNA completes its replication cycle by viral complementation. uDNA-derived genomes undergo recombination with the integrated provirus-derived genomes during second round infection.</p> <p>Conclusion</p> <p>This novel mode of retroviral replication allows survival of viruses which would otherwise be lost because of a failure to integrate, amplifies the effective amount of cellular coinfection, increases the replicating HIV-1 gene pool, and enhances the opportunity for diversification through errors of polymerization and recombination.</p
Centers of Mass and Rotational Kinematics for the Relativistic N-Body Problem in the Rest-Frame Instant Form
In the Wigner-covariant rest-frame instant form of dynamics it is possible to
develop a relativistic kinematics for the N-body problem. The Wigner
hyperplanes define the intrinsic rest frame and realize the separation of the
center-of-mass. Three notions of {\it external} relativistic center of mass can
be defined only in terms of the {\it external} Poincar\'e group realization.
Inside the Wigner hyperplane, an {\it internal} unfaithful realization of the
Poincar\'e group is defined. The three concepts of {\it internal} center of
mass weakly {\it coincide} and are eliminated by the rest-frame conditions. An
adapted canonical basis of relative variables is found. The invariant mass is
the Hamiltonian for the relative motions. In this framework we can introduce
the same {\it dynamical body frames}, {\it orientation-shape} variables, {\it
spin frame} and {\it canonical spin bases} for the rotational kinematics

Cooperation between STAT3 and c-Jun Suppresses Fas Transcription
Decreased Fas expression during tumor progression often results in a loss of Fas-ligand (FasL)-mediated apoptosis. Human and mouse melanoma exhibit an inverse correlation between the degree of Fas cell surface expression, tumorigenicity, and metastatic capacity. The expression of dominant negative Stat3 or c-Jun in melanoma cells efficiently increased Fas expression and sensitized cells to FasL-induced apoptosis. Stat3+/− as well as c-Jun−/− cells exhibited increased Fas cell surface expression and higher sensitivity to FasL-mediated apoptosis. Suppression of Fas expression by Stat3 and c-Jun is uncoupled from Stat3-mediated transcriptional activation. Our findings indicate that Stat3 oncogenic activities could also be mediated through its cooperation with c-Jun, resulting in downregulation of Fas surface expression, which is implicated in the tumor's ability to resist therapy and metastasize
The Dielectric Constant of Ionic Solutions: A Field-Theory Approach
We study the variation of the dielectric response of a dielectric liquid
(e.g. water) when a salt is added to the solution. Employing field-theoretical
methods we expand the Gibbs free-energy to first order in a loop expansion and
calculate self-consistently the dielectric constant. We predict analytically
the dielectric decrement which depends on the ionic strength in a complex way.
Furthermore, a qualitative description of the hydration shell is found and is
characterized by a single length scale. Our prediction fits rather well a large
range of concentrations for different salts using only one fit parameter
related to the size of ions and dipoles.Comment: 5 pages, 2 figure
Universal features in the growth dynamics of complex organizations
We analyze the fluctuations in the gross domestic product (GDP) of 152
countries for the period 1950--1992. We find that (i) the distribution of
annual growth rates for countries of a given GDP decays with ``fatter'' tails
than for a Gaussian, and (ii) the width of the distribution scales as a power
law of GDP with a scaling exponent . Both findings are in
surprising agreement with results on firm growth. These results are consistent
with the hypothesis that the evolution of organizations with complex structure

The Ultra-Potent and Selective TLR8 Agonist VTX-294 Activates Human Newborn and Adult Leukocytes
Background: Newborns display distinct immune responses that contribute to susceptibility to infection and reduced vaccine responses. Toll-like receptor (TLR) agonists may serve as vaccine adjuvants, when given individually or in combination, but responses of neonatal leukocytes to many TLR agonists are diminished. TLR8 agonists are more effective than other TLR agonists in activating human neonatal leukocytes in vitro, but little is known about whether different TLR8 agonists may distinctly activate neonatal leukocytes. We characterized the in vitro immuno-stimulatory activities of a novel benzazepine TLR8 agonist, VTX-294, in comparison to imidazoquinolines that activate TLR8 (R-848; (TLR7/8) CL075; (TLR8/7)), with respect to activation of human newborn and adult leukocytes. Effects of VTX-294 and R-848 in combination with monophosphoryl lipid A (MPLA; TLR4) were also assessed. Methods: TLR agonist specificity was assessed using TLR-transfected HEK293 cells expressing a NF-κB reporter gene. TLR agonist-induced cytokine production was measured in human newborn cord and adult peripheral blood using ELISA and multiplex assays. Newborn and adult monocytes were differentiated into monocyte-derived dendritic cells (MoDCs) and TLR agonist-induced activation assessed by cytokine production (ELISA) and co-stimulatory molecule expression (flow cytometry). Results: VTX-294 was ∼100x more active on TLR8- than TLR7-transfected HEK cells (EC50, ∼50 nM vs. ∼5700 nM). VTX-294-induced TNF and IL-1β production were comparable in newborn cord and adult peripheral blood, while VTX-294 was ∼ 1 log more potent in inducing TNF and IL-1β production than MPLA, R848 or CL075. Combination of VTX-294 and MPLA induced greater blood TNF and IL-1β responses than combination of R-848 and MPLA. VTX-294 also potently induced expression of cytokines and co-stimulatory molecules HLA-DR and CD86 in human newborn MoDCs. Conclusions: VTX-294 is a novel ultra-potent TLR8 agonist that activates newborn and adult leukocytes and is a candidate vaccine adjuvant in both early life and adulthood

According to the importance of the embodiment of robots in the sense of presence, the influence of robots in communication culture is anticipated.
