OPTIMASI KONDISI PCR UNTUK ANALISIS VARIASI GENETIK MIKROSATELIT VARIAN JATI ARBORETUM DENGAN MENGGUNAKAN PEWARNAAN PERAK
Jati merupakan salah satu spesies perkebunan yang menjanjikan di daerah tropis, maka perakitan calon tanaman jati unggul
diperlukan. Untuk itu dilakukan penelitian dengan tujuann menganalisis variasi genetik varian jati arboretum dengan penanda
mikrosatelit. Sebelum melakukan analisis variasi genetik, terlebih dahulu dilakukan Optimasi kondisi PCR untuk berlangsungnya proses
amplifikasi DNA sampel. PCR peka terhadap sejumlah parameter meliputi: magnesium klorida, DNA templat, konsentrasi primer, da n
suhu annealing selama amplifikasi. Kondisi reaksi dioptimalkan sebelum amplifikasi guna menghindari produk amplifikasi tidak spesifik
seperti ukuran pita DNA yang heterogen. Di samping itu penggunaan pewarnaan perak juga perlu diperhatikan meliputi, komponenkomponen larutan yang digunakan, tahapan-tahapan pewarnaannya harus benar-benar dilakukan sesuai petunjuk dan butuh
keterampilan serta kehatia-hatian. Hal ini dilakukan untuk memperoleh pola pita DNA yang baik sehingga memudahkan analisis
selanjutnya dalam hal ini variasi genetik.

EFFORTS TO PROMOTE CRITICAL THINKING THROUGH COOPERATIVE LEARNING TYPE THINK TALK WRITE ASSIGNMENT BASED ON BASIC GENETICS
This study aims to encourage critical thinking skills of students in learning
basic genetics. To answer these objectives the research conducted on students of
Biology Education class of 2011, amounting to 89 people. Design of experiments
using the One-Shot Case Study. The treatment of the application of cooperative
learning model type Think Talk Write (TTW) based assignments carried out during
one semester. The results of the study after analysis showed that students' critical
thinking skills and a good knowledge of genetic categorized reached more than 60%.
Based on this it can be concluded that the application of cooperative learning model
type Think Talk Write (TTW) based assignments can encourage students' critical
thinking skills in basic genetics course
The Effectiveness of Online Learning in terms of Biology Learning Achievement of Class XII Mathematics and Natural Sciences at SMA a Negeri 1 Paciran
The purpose of this research was to describe the effectiveness of online learning in the Covid-19 era interms of the biology learning achievement of class XII MIPA students of SMA Negeri 1 Paciran. Thisresearch design uses a descriptive quantitative research approach. The subjects of this study were takenby purposive sampling technique with a total of 159 students from five class XII MIPA at SMA Negeri 1Paciran. Research instrument with documentation and summative tests. The research data collectiontechnique used school documentation and the year-end assessment test (PAT) which was carried out byschools online with the google form application. The research data were analyzed by means of the test,then compared with the standard measure of the effectiveness of the Ministry of Home Affairs Researchand Development Agency. The results showed that the biology learning achievement average of class XIIMIPA students reached 72.91. If the mean is compared with the standard measure of effectiveness, it isknown that the level of achievement of the predicate is quite effective. So online learning carried out atSMA Negeri 1 Paciran is quite effective in terms of obtaining biology learning achievement for studentsof class XII MIPA
VALIDITAS PETUNJUK PRAKTIKUM ELEKTRONIK UNTUK MENINGKATKAN KEMAMPUAN BERPIKIR KRITIS SISWA MELALUI PROBLEM BASED LEARNING
Pendekataan pembelajaran yang dapat meningkatkan kemampuan berpikir kritis siswa dapat melalui Problem Based Learning. Tujuan dalam penelitian ini adalah untuk: 1)mendesain petunjuk praktikum elektronik untuk meningkatkan kemampuan berpikir kritis siswa melalui problem based learning yang valid dan 2) mendeskripsikan validitas petunjuk praktikum elektronik untuk meningkatkan kemampuan berpikir kritis siswa melalui problem based learning yang didesain. Model pengembangan yang digunakan dalam penelitian ini adalah Model pengembangan ADDIE, yang meliputi 5 tahapan yaitu, analisis (analysis), desain (design), validitas (developmnet), implementasi (implementation), dan evaluasi (evaluation). Namun dalam penelitian ini hanya sampai validitas karena keterbatasan dalam penelitian. Kriteria penilaian validitas meliputi kelayakan isi, sistematika, kegrafisan, dan bahasa. Data validitas petunjuk praktikum elektonik ini diperoleh dari hasil penilaian validator. Hasil penelitian bahwa rata rata yang diproleh pada aspek kelayakan isi sebesar 77,78%, rata rata pada aspek kelayakan sitematika sebesar 75%, rata rata pada aspek kelayakan kegrafisan sebesar 80%, rata rata pada aspek kelayakan bahasa sebesar 75%. Presentase keseluruhan dari Petunjuk praktikum elektronik untuk meningkatkan kemampuan berpikir kritis siswa melalui problem based learning adalah 77% dengan kategori valid karena presentase nilai skor di atas 70%. Dengan demikian dapat disimpulkan bahwa petunjuk praktikum elektronik untuk meningkatkan kemampuan berpikir kritis siswa melalui model Problem Based Learning yang didesain telah memenuhi kriteria kevalida
Analysis of Environmental Conditions and Diversity of Fisherman Catches as Indicators of Sea Water Pollution at The Beach of Karangmangu Village
The habit of disposing of household waste by Karangmangu villagers will affect the environmental conditions of the surrounding beaches, especially for the marine life under the sea that fish are frequently captured by fishermen. Under these conditions, it is necessary to do research that aims to: 1) analyze the condition of the coastal environment of Karangmangu Village based on the provisions of water quality and 2) analyze the diversity of fish caught by fishermen on the beach of Karangmangu Village. This type of research is a descriptive study. Data collection using the observation method. The data obtained were in the form of environmental conditions, abundance of fish species, diversity, and dominance index. The results of the study 1) found trash along the coast in the form of household waste, 2) an abundance of fish species of 105 individuals/m2 in the form of Decapterus macrosoma, Rastralliger kanagurta, Selar crumenophthalmus L, and Euthynnus affinis. The results of data analysis on the condition of the coastal environment of Karangmangu Village experience light pollution, the diversity index value is 0.30 which means low, the dominance index is 0.07 which means low. Based on the results of data analysis, it can be concluded that the condition of Karangmangu waters is classified as unstable, but it is still able to support the life of fish caught by fishermen, even though the value of diversity and dominance is low
Identificaton of Sago (Metroxylon sp) In Pelang Tourism, Merakurak District, Tuban Regency
Pelang Tourism is a natural tourist spot located in Pelang Hamlet, Tuban Regency, East Java. Local people take advantage of the Pelang river and the surrounding Metroxylon park as natural attractions. Many human activities affect the environment, one of which is logging Metroxylon and land use activities to make food stalls or souvenir shops. These activities have an impact on changing the characteristics of the Rainbow Tourism area so that it has an impact on the decline in population and biodiversity in the area, one of which is Metroxylon. The high population of Metroxylon is a source of germplasm in tourist areas. Based on this, it is necessary to do research. The purpose of this research is to study the diversity of Metroxylon in Pelang Tourism. This study uses descriptive quantitative research methods with observation data collection techniques. Based on the research results, there are 762 families of Metroxylon Arecaceae consisting of Metroxylon rumphii Martius, Metroxylon sago Rottb, Metroxylon microcanthum Mart. Based on the results of data analysis obtained Metroxylon Diversity Value of 0.9692 which means low. Metroxylon dominance value of 0.4213 means low. Based on the Diversity and Domination Index, it shows that the diversity of Metroxylon in Pelang Tourism is low, meaning that the procurement of tourist attractions has affected the diversity of Metroxylon. Therefore, it is necessary to maintain the diversity and population of Metroxylon in Pelang Tourism so that the germplasm source is not destroyed
Utilization Of Various Banana Stems (Musa) As A Medium for Growing Mustard Plants (Brassica juncea.)
Banana stems are parts that have not been utilized optimally, it will be discarded or collected in a place as waste and allowed to rot. The content of banana stems includes potassium, phosphorus and nitrogen which are good for supporting plant growth. Based on these contents, research was carried out using banana stalks as a growing medium for mustard greens (Brassica juncea). The objectives to be achieved in this study were to determine the effect of giving banana stems as a planting medium on the growth of mustard greens. This type of research is experimental, while the experimental design used was a completely randomized design with 3 treatments, namely kepok banana, raja sereh banana and biru banana with 6 repetitions. Data collection techniques by means of observation and measurement. The data that had been collected were analyzed statistically using the analysis of variance test using the SPSS application. From the results of statistical analysis, it shows that giving different kinds of banana stems has no difference in the growth of mustard greens. However, the provision of different types of banana stems can affect the growth of mustard greens where the growth can be increased than the control data because the growth rate of mustard greens is better than the control growth
