    SVR Rates of HCV-infected population under PEG-IFN-α/R treatment in Northwest China

    BACKGROUND: Chronic HCV Patients taking PEG-IFN-α/R from different ethnic groups have different probabilities of reaching a sustained viral response (SVR). There are many influence factors, such as HCV genotype, IL-28B single-nucleotide polymorphisms (SNP), Fibrosis 4 index (FIB-4), and aspartate aminotransferase-to-platelet ratio index (APRI) score. But the baseline factors in relation to treatment outcome was still not much clear. METHODS: We evaluated data from 231 chronic HCV patients with or without liver fibrosis and their antiviral efficacy after treatment with pegylated interferon plus ribavirin (PEG-IFN-α/R) for 24–48 weeks. IL-28B SNP and HCV genotypes were analyzed with genome sequencing using pyrosequencing. RESULTS: Sustained viral response (SVR) rates of patients with HCV 1b and 2a genotypes were 52.25% (58/111) and 75.28% (67/89) (P < 0.01). SVR rates of patients with IL-28B rs8099917 TT, rs12979860 CC and rs12980275 AA were 92.41% (25/27), 92.86% (26/28) and 88.89% (24/27) separately. We found that SVR rates in HCV 1b and 2a patients were only 31.0 and 39.4% if their FIB-4 > 3.25. In addition, when their APRI > 2, only 30.3% of HCV 1b patients and 50.2% of HCV 2a patients could obtain SVR. CONCLUSIONS: There were high proportion of HCV genotype 1b and 2a in Northwest China. In both HCV 1b and 2a genotypes, patients with protective-genotype of IL-28B were more likely to obtain SVR. However, those with significant fibrosis or cirrhosis were less likely, no matter their genotype. Combined factors of HCV genotype, IL-28B genotype, FIB-4 and ARPI may indicate high prediction and clinical value regarding treatment with PEG-IFN-α/R and prognostic evaluation of chronic hepatitis C patients

    Novel <em>acsF</em> gene primers revealed a diverse phototrophic bacterial population, including <em>Gemmatimonadetes</em>, in Lake Taihu (China)

    UNLABELLED: Anoxygenic phototrophs represent an environmentally important and phylogenetically diverse group of organisms. They harvest light using bacteriochlorophyll-containing reaction centers. Recently, a novel phototrophic bacterium, Gemmatimonas phototrophica, belonging to a rarely studied phylum, Gemmatimonadetes, was isolated from a freshwater lake in the Gobi Desert. To obtain more information about the environmental distribution of phototrophic Gemmatimonadetes, we collected microbial samples from the water column, upper sediment, and deeper anoxic sediment of Lake Taihu, China. MiSeq sequencing of the 16S rRNA, pufM, and bchY genes was carried out to assess the diversity of local phototrophic communities. In addition, we designed new degenerate primers of aerobic cyclase gene acsF, which serves as a convenient marker for both phototrophic Gemmatimonadetes and phototrophic Proteobacteria Our results showed that most of the phototrophic species in Lake Taihu belong to Alpha- and Betaproteobacteria Sequences of green sulfur and green nonsulfur bacteria (phototrophic Chlorobi and Chloroflexi, respectively) were found in the sediment. Using the newly designed primers, we identified a diverse community of phototrophic Gemmatimonadetes forming 30 operational taxonomic units. These species represented 10.5 and 17.3% of the acsF reads in the upper semiaerobic sediment and anoxic sediment, whereas their abundance in the water column was &lt;1%.IMPORTANCE: Photosynthesis is one of the most fundamental biological processes on Earth. Recently, the presence of photosynthetic reaction centers has been reported from a rarely studied bacterial phylum, Gemmatimonadetes, but almost nothing is known about the diversity and environmental distribution of these organisms. The newly designed acsF primers were used to identify phototrophic Gemmatimonadetes from planktonic and sediment samples collected in Lake Taihu, China. The Gemmatimonadetes sequences were found mostly in the upper sediments, documenting the preference of Gemmatimonadetes for semiaerobic conditions. Our results also show that the phototrophic Gemmatimonadetes present in Lake Taihu were relatively diverse, encompassing 30 operational taxonomic units.</p

    Fabrication of micro-scale radiation shielding structures using tungsten nanoink through electrohydrodynamic inkjet printing

    Electronics components used in space and strategic missions are exposed to harsh radiation environments, which could cause operational malfunction of the system through lattice displacement or ionization effects. One potential solution is to use tungsten as radiation shielding. Tungsten is a very effective material in shielding electronic components and manufacturing gratings for x-ray imaging. However, intrinsic properties of tungsten (e.g. density, chemical/thermal inertness and hardness) post a significant challenge of fabricating the material into micro-scale and delicate structures, especially in electronic device fabrication. To address the problem, we designed a new tungsten nanoink and developed a straightforward approach to create tungsten micro-structures by 3D printing. Various microstructures down to 10 µm resolution have been patterned and fabricated by electrohydrodynamic inkjet (e-jet) printing using tungsten nanoink. By optimizing process parameters (voltage modality) and materials properties (ink formulation), the dimension and morphology of the structures can be precisely controlled. An AC-modulated voltage was employed during the e-jet printing process to make the patterns much more controllable and stable. Multi-layer tungsten lines were characterized by x-ray imaging and exhibited excellent absorption of x-ray radiation. With the same thickness, printed lines showed nearly 1/3 absorptivity of x-ray radiation of bulk tungsten, leading to significant radiation attenuation effectiveness. Tungsten nanoink is a new material used in e-jet printing that has not been reported in the literature to the best of authors\u27 knowledge. The study establishes a new methodology of manufacturing micro-nano scale shielding components for electronic devices and rapid prototyping of gratings and collimators in radiography for medical and inspection applications. The research also provides practical guidance to fabricate high melting-point metals via nanoink and micro/nano scale 3D printing

    Steady-state bifurcation and regularity of nonlinear Burgers equation with mean value constraint

    In this paper, we focus on the steady-state bifurcation problem of the nonlinear Burgers equation within a bounded domain, considering both homogeneous Dirichlet boundary conditions and homogeneous Neumann boundary conditions with a mean value constraint. Unlike previous studies, we develop an enhanced turbulence model by incorporating nonlinear higher-order terms (such as u2 u^2 and u3 u^3 ) and linear source terms λu \lambda u into the one-dimensional Burgers equation. Our steady-state bifurcation analysis establishes for the first time how the coupled forward energy cascade and inverse energy transfer mechanisms collectively govern the dynamics of initial flow instability. By combining the spectral theorem for a linear compact operator with the normalized Lyapunov–Schmidt reduction method and the implicit function theorem, we derive the complete criterion for the critical bifurcation condition, the explicit form of the bifurcation solution, and its regularity

    The Nucleated Poly(L-lactic acid): The Role of a Phenylacetic Hydrazide Derivative

    The melt-crystallization, cold-crystallization, melting behaviors, thermal stability, and optical properties of poly(L-lactic acid) (PLLA) nucleated by a phenylacetic hydrazide derivative (DAPH) were investigated. The melt-crystallization confirmed DAPH’s heterogeneous nucleation role in improving PLLA’s poor crystallization capability, concurrently DAPH loading, cooling rate and final melting temperature were three key factors affecting PLLA’s melt-crystallization behaviors. The results from cold-crystallization indicated that the addition of DAPH could accelerate PLLA’s cold-crystallization via cold-crystallization peak’s shift toward the lower temperature side as DAPH loading increased. Through analysis of PLLA/DAPH’s multiple melting behaviors after melt-crystallization, it was found that DAPH loading, the previous melt-crystallization and heating rate determined PLLA/DAPH’s melting processes after melt-crystallization; and PLLA/DAPH’s melting processes after isothermal crystallization depended on the previous crystallization temperature. A drop in thermal decomposition temperature for 5 wt.% mass loss suggested that pure PLLA has better thermal stability compared with PLLA/DAPH. Additionally, the introduction of DAPH could not improve PLLA’s transparency, in contrast, the haze of PLLA was increased greatly owing to the existence of DAPH

    Impact of the National Reimbursement Drug List Negotiation Policy on Accessibility of Anticancer Drugs in China: An Interrupted Time Series Study

    Objective: Since 2016, the Chinese government has been regularly implementing the National Reimbursement Drug List Negotiation (NRDLN) to improve the accessibility of drugs. In the second round of NRDLN in July 2017, 18 anticancer drugs were included. This study analyzed the impact of the NRDLN on the accessibility of these 18 anticancer drugs in China. Methods: National hospital procurement data were collected from 2015 to 2019. As measurements of drug accessibility, monthly average of drug availability or defined daily dose cost (DDDc) was calculated. Interrupted time series (ITS) analysis was employed to evaluate the impact of NRDLN on drug accessibility. Multilevel growth curve models were estimated for different drug categories, regions or levels of hospitals. Results: The overall availability of 18 anticancer drugs increased from about 10.5% in 2015 to slightly over 30% in 2019. The average DDDc dropped from 527.93 CNY in 2015 to 401.87 CNY in 2019, with a reduction of 23.88%. The implementation of NRDLN was associated with higher availability and lower costs for all 18 anticancer drugs. We found an increasing level in monthly drug availability (β2 = 2.1126), which ascended more sharply after the implementation of NRDLN (β3 = 0.3656). There was a decreasing level in DDDc before July 2017 (β2 = −108.7213), together with a significant decline in the slope associated with the implementation of NRDLN (β3 = −4.8332). Compared to Traditional Chinese Medicines, the availability of Western Medicines was higher and increased at a higher rate (β3 = 0.4165 vs. 0.1108). Drug availability experienced a larger instant and slope increase in western China compared to other regions, and in secondary hospitals than tertiary hospitals. Nevertheless, regional and hospital-level difference in the effect of NRDLN on DDDc were less evident. Conclusion: The implementation of NRDLN improves the availability and reduces the cost of some anticancer drugs in China. It contributes to promoting accessibility of anticancer drugs, as well as relieving regional or hospital-level disparities. However, there are still challenges to benefit more patients sufficiently and equally. It requires more policy efforts and collaborative policy combination

    Transcriptome analysis of the hepatopancreas from the Litopenaeus vannamei infected with different flagellum types of Vibrio alginolyticus strains

    Vibrio alginolyticus, one of the prevalently harmful Vibrio species found in the ocean, causes significant economic damage in the shrimp farming industry. Its flagellum serves as a crucial virulence factor in the invasion of host organisms. However, the processes of bacteria flagella recognition and activation of the downstream immune system in shrimp remain unclear. To enhance comprehension of this, a ΔflhG strain was created by in-frame deletion of the flhG gene in V. alginolyticus strain HN08155. Then we utilized the transcriptome analysis to examine the different immune responses in Litopenaeus vannamei hepatopancreas after being infected with the wild type and the mutant strains. The results showed that the ΔflhG strain, unlike the wild type, lost its ability to regulate flagella numbers negatively and displayed multiple flagella. When infected with the hyperflagella-type strain, the RNA-seq revealed the upregulation of several immune-related genes in the shrimp hepatopancreas. Notably, two C-type lectins (CTLs), namely galactose-specific lectin nattectin and macrophage mannose receptor 1, and the TNF receptor-associated factor (TRAF) 6 gene were upregulated significantly. These findings suggested that C-type lectins were potentially involved in flagella recognition in shrimp and the immune system was activated through the TRAF6 pathway after flagella detection by CTLs
