    Photoreactivation of Lethal Damage Induced in Hamster X Xenopus Hybrid Cells and Their Parentals by UV Light

    A85 Xenopus cells that exhibited a high level of photoreactivation (PR) and V79B2 hamster cells that exhibited little PR were fused to produce the V79B2 x A85 cell line — a hybrid line which possessed a relatively stable karyotype, with most cells containing the entire V79B2 and A85 genomes. UV and UV plus PR fluence-survival relations were then determined and compared for the hybrid and parental lines in a first attempt to elucidate interactions of the parental PR mechanisms in the hybrid. It was anticipated that the A85 genome in the hybrid would produce PR enzyme in sufficient concentration and of such a nature as to efficiently PR UV-induced lethal damage in both A85 and V79B2 DNA, and little difference would be observed in the levels of PR exhibited by the V79B2 x A85 and A85 lines. To the contrary, the level of PR observed for the hybrid was substantially below that observed for the A85 line. To assist in the interpretation of this unexpected observation, three additional preliminary studies were carried out: 1) Comparison of the optimum PR schemes for the A85 and hybrid lines, 2) examination of relations between the PR and dark UV repair mechanisms possessed by these lines, and 3) comparison of the levels of PR of chromatid deletions induced by UV in selected V79B2 and A85 chromosomes of the hybrid. The results suggested that the relatively low level of PR manifested by the hybrid cells was a consequence of their inability to efficiently PR pyrimidine dimers induced by UV in V79B2 DNA

    Household saving

    Household decisions on whether to save or spend play a key role in the outlook for aggregate demand. A range of factors could help to explain the fall in the household saving ratio over the period 1995 to 2007. Declines in long-term real interest rates, looser credit conditions, rising asset values and greater macroeconomic stability are all likely to have reduced the incentive or the need to save. Lower household saving was also offset to some extent by higher corporate saving. Since 2007, the financial crisis and subsequent recession have unwound some of these factors and may continue to lead to a rise in household saving.

    Nanowire and core-shell-structures on flexible Mo Foil for CdTe solar cell applications

    CdTe films, nanowires, film-nanowire combinations and CdS-CdTe core-shell structures have been fabricated in a preliminary survey of growth methods that will generate structures for PV applications. Selectivity between film, nanowire and film plus nanowire growth was achieved by varying the pressure of N2 gas present during Au-catalysed VLS growth of CdTe, on either Mo or Si substrates. Metamorphic growth of CdTe nanowires on sputtered CdTe films, deposited on glass substrates, was demonstrated. Coating of CdTe nanowires with CBD CdS gave conformal coverage whereas coating with MOCVD (Cd,Zn)S yielded highly crystallographic dendritic growth on the wires

    Purinergic receptor mediated calcium signalling in urothelial cells

    Non-neuronal ATP released from the urothelium in response to bladder stretch is a key modulator of bladder mechanosensation. Whilst non-neuronal ATP acts on the underlying bladder afferent nerves to facilitate sensation, there is also the potential for ATP to act in an autocrine manner, modulating urothelial cell function. The aim of this study was to systematically characterise the functional response of primary mouse urothelial cells (PMUCs) to ATP. PMUCs isolated from male mice (14-16 weeks) were used for live-cell fluorescent calcium imaging and qRT-PCR to determine the expression profile of P2X and P2Y receptors. The majority of PMUCs (74-92%) responded to ATP (1 μM-1 mM), as indicted by an increase in intracellular calcium (iCa2+). PMUCs exhibited dose-dependent responses to ATP (10 nM-1 mM) in both calcium containing (2 mM, EC50 = 3.49 ± 0.77 μM) or calcium free (0 mM, EC50 = 9.5 ± 1.5 μM) buffers. However, maximum iCa2+ responses to ATP were significantly attenuated upon repetitive applications in calcium containing but not in calcium free buffer. qRT-PCR revealed expression of P2X1-6, and P2Y1-2, P2Y4, P2Y6, P2Y11-14, but not P2X7 in PMUCs. These findings suggest the major component of ATP induced increases in iCa2+ are mediated via the liberation of calcium from intracellular stores, implicating functional P2Y receptors that are ubiquitously expressed on PMUCs.Russell Chess-Williams, Donna J.     Profiling micro-organic contaminants in groundwater using multi-level piezometers

    The presence of micro-organic pollutants, including ‘emerging contaminants’ within groundwater is of increasing interest. Robust protocols are required to minimise the introduction of contamination during the sampling process. Below we discuss the sampling protocols used to reduce inputs of plasticisers during the sampling process, as well as the techniques used to characterise the distribution of micro-organic pollutants in the subsurface. In this study multi-level piezometers installed into the Sherwood Sandstone aquifer in the suburban area of Doncaster, Yorkshire, were used to look at the changes in micropollutants with depth and lithology. Two different methods of pumping groundwaters were employed, and sampling campaigns were undertaken in February and July 2014, under contrasting hydrological conditions

    Evaluating an online support package delivered within a disability unemployment service: study protocol for a randomised controlled feasibility study

    Background Mental health problems such as anxiety and depression are known to be higher in those who are unemployed. Cognitive behavioural therapy (CBT) is a recognised support for people with such problems and can improve the ability of people to get back to work.<p></p> Methods/design Participants with symptoms of low mood will be recruited from the disability employment service, Remploy. Participants will receive either immediate or delayed access to an online CBT-based life skills intervention, the “Living Life” package. The primary end point will be at 3 months when the delayed group will be offered the intervention. This feasibility study will test the trial design and assess recruitment, retention, acceptability and adherence, as well as providing efficacy data.<p></p> Discussion The study will inform the design and sample size for a future full randomised controlled trial (RCT) which will be carried out to determine the effectiveness of the online package in improving mood and employment status.<p></p&gt

    Attitudes of Paramedic Students towards Specific Medical Conditions: A Four-Year Study

    Purpose: The purpose of this study was to determine the attitude of first year students entering a paramedic course over 4 consecutive yearly intakes toward patients with intellectual disability, substance abuse, attempted suicide, and acute mental illness. Method: The students’ attitude towards four medical conditions commonly encountered in the out-of-hospital setting was assessed using the Medical Condition Regard Scale (MCRS). The MCRS score ranges from 11 to 66. Results: 230 students in Victoria, Australia, participated in the study; 66% were female, and 76% of all students \u3c 21 years of age. Students showed the most negative attitudes towards substance abuse, mean 35(SD+6.6), and the most positive attitude toward intellectual disability, mean 38.2(SD+6.3). Students in 2008 cohort displayed significantly higher regard for all medical conditions (pConclusions: The study findings suggest that these students in Victoria, Australia, entering an undergraduate paramedic or paramedic/nursing course have a relatively poor attitude towards the four reviewed medical conditions, particularly substance abuse

    Fit for purpose? Fitting ontological security studies 'into' the discipline of International Relations : towards a vernacular turn

    The performance of International Relations (IR) scholarship – as in all scholarship – acts to close and police the boundaries of the discipline in ways that reflect power–knowledge relations. This has led to the development of two strands of work in ontological security studies in IR, which divide on questions of ontological choice and the nature of the deployment of the concept of dread. Neither strand is intellectually superior to the other and both are internally heterogeneous. That there are two strands, however, is the product of the performance of IR scholarship, and the two strands themselves perform distinct roles. One allows ontological security studies to engage with the ‘mainstream’ in IR; the other allows ‘international’ elements of ontological security to engage with the social sciences more generally. Ironically, both can be read as symptoms of the discipline’s issues with its own ontological (in)security. We reflect on these intellectual dynamics and their implications and prompt a new departure by connecting ontological security studies in IR with the emerging interdisciplinary fields of the ‘vernacular’ and ‘everyday’ via the mutual interest in biographical narratives of the self and the work that they do politically

    There and back again: detecting regularity in human encounter communities

    Detecting communities that recur over time is a challenging problem due to the potential sparsity of encounter events at an individual scale and inherent uncertainty in human behavior. Existing methods for community detection in mobile human encounter networks ignore the presence of temporal patterns that lead to periodic components in the network. Daily and weekly routine are prevalent in human behavior and can serve as rich context for applications that rely on person-to-person encounters, such as mobile routing protocols and intelligent digital personal assistants. In this article, we present the design, implementation, and evaluation of an approach to decentralized periodic community detection that is robust to uncertainty and computationally efficient. This alternative approach has a novel periodicity detection method inspired by a neural synchrony measure used in the field of neurophysiology. We evaluate our approach and investigate human periodic encounter patterns using empirical datasets of inferred and direct-sensed encounters
