    Grâce àl’accessibilité de plus en plus grande d’importants, corpus oraux, l’analyse de phénomènes d’hésitation, que nous désignerons, par l’expression « marqueurs du travail de formulation », a retenu, l’attention de nombreux chercheurs et a été appréhendée sous divers, angles, souvent d’inspiration cognitiviste. Pour ce qui est du français, il, existe une vaste litteérature consacrée au travail de formulation chez des, locuteurs natifs (voir entre autres la bibliographie de Candea 2000), mais, pour les apprenants de français L2 les recherches sont moins nombreuses, et elles se caractérisent souvent par une visée large, par exemple, concernant l’organisation discursive et la capacité de créer un discours, plus ou moins fluide (voir entre autres les travaux de Hilton et de Towell).,L’objectif de notre étude sera plus pointue : nous examinerons les, marqueurs du travail de formulation produits dans le discours oral, d’apprenants néerlandophones du français langue seconde (FL2) afin de, cerner les processus d’appropriation de la morphologie verbale, plus, précisément de la conjugaison et de l’accord sujet-verbe àla 3ème personne, de l’indicatif présent et du passé composé. Nous présenterons d’abord, brièvement les résultats de recherches antérieures, pour ensuite esquisser, notre méthodologie et interpréter les résultats obtenus

    On the Rationality of Network Development: the Case of the Belgian Highway Network

    The development of transport networks has been explained, predicted and planned using a variety of methodological approaches. These range from narrative historical accounts to the application of models borrowed from the natural sciences, the latter being predominant in the field of transport economics. Probably the most remarkable example is the mimicking of highway networks by slime mould in Petri dishes. The aim of this paper is to examine and compare methods used to hypothesise on and explain the development of transport networks, and we specifically focus on methods that emphasise topology over topography, relations over form (in line with the work of Gabriel Dupuy and others). Belgium was chosen as case because the topology of Belgium’s highway network is considered by some as one of the most ‘rational’ in the world, although its form and materiality are often qualified as ‘chaotic’, or indeed ‘irrational’. The quantitative analysis of the development of this network reported in the present paper indicates that this supposed rationality is seldom followed and seemingly ‘irrational’ parameters literally deviate its growth. Therefore, the quantitative part is complemented by a historical analysis which focuses more on the role, and indeed rationality, of historical actors and the wider institutional context. Material provided by this case study supplies fuel for discussion about broader issues, in particular the underlying ideological and political-economic claims associated with a particular methodological approach. This is especially relevant given the fact that models used to predict past transport investments are also employed to evaluate future investments in infrastructure. Quantitative approaches generally attribute a central role to the concept of demand, and thus degrees of ‘rationality’ are in fact linked to ideas of consumer democracy where individual demand guides investment decisions. In contrast, interpretations of a less deterministic nature emphasise the degrees of freedom of political actors/choice. We conclude that the views held by actors of the past, present and future of transport networks are relevant for democratic debates on transport policy since the metaphors and models used are not value-neutral

    A role for human N-alpha acetyltransferase 30 (Naa30) in maintaining mitochondrial integrity

    N-terminal acetylation (Nt-acetylation) by N-terminal acetyltransferases (NATs) is one of the most common protein modifications in eukaryotes. The NatC complex represents one of three major NATs of which the substrate profile remains largely unexplored. Here, we defined the in vivo human NatC Nt-acetylome on a proteome-wide scale by combining knockdown of its catalytic subunit Naa30 with positional proteomics. We identified 46 human NatC substrates, expanding our current knowledge on the substrate repertoire of NatC which now includes proteins harboring Met-Leu, Met-Ile, Met-Phe, Met-Trp, Met-Val, Met-Met, Met-His and Met-Lys N termini. Upon Naa30 depletion the expression levels of several organellar proteins were found reduced, in particular mitochondrial proteins, some of which were found to be NatC substrates. Interestingly, knockdown of Naa30 induced the loss of mitochondrial membrane potential and fragmentation of mitochondria. In conclusion, NatC N-tacetylates a large variety of proteins and is essential for mitochondrial integrity and function

    Exploring park–people conflicts in Colombia through a social lens

    Natural resource-related conflicts between local communities and nation states can be extremely destructive. Worldwide, interest is growing in gaining a better understanding of why and how these conflicts originate, particularly in protected areas inhabited by local communities. The literature on local attitudes towards and perceptions of park conservation and park-people conflicts is quite extensive. Studies have examined the socioeconomic and geographical determinants of attitudes to protected areas. However, the role of such determinants in the experience of park-people conflicts has received considerably less attention. Drawing on 601 interviews with people living in or near 15 Colombian national protected areas (NPAs), we examine the socioeconomic and geographical variables that are most influential in people's experience of conflict related to restricted access to natural resources. We find that the experience of this type of conflict is largely explained by the NPA where a person resides, pursuit of productive activities within the NPA, previous employment in NPA administration, gender and ethnicity. We recommend implementing socially inclusive conservation strategies for conflict prevention and resolution in Colombia's NPAs, whereby both women and men from different ethnic groups are engaged in design and implementation

    PLecDom: a program for identification and analysis of plant lectin domains

    PLecDom is a program for detection of Plant Lectin Domains in a polypeptide or EST sequence, followed by a classification of the identified domains into known families. The web server is a collection of plant lectin domain families represented by alignments and profile Hidden Markov Models. PLecDom was developed after a rigorous analysis of evolutionary relationships between available sequences of lectin domains with known specificities. Users can test their sequences for potential lectin domains, catalog the identified domains into broad substrate classes, estimate the extent of divergence of new domains with existing homologs, extract domain boundaries and examine flanking sequences for further analysis. The high prediction accuracy of PLecDom combined with the ease with which it handles large scale input, enabled us to apply the program to protein and EST data from 48 plant genome-sequencing projects in various stages of completion. Our results represent a significant enrichment of the currently annotated plant lectins, and highlight potential targets for biochemical characterization. The search algorithm requires input in fasta format and is designed to process simultaneous connection requests from multiple users, such that huge sets of input sequences can be scanned in a matter of seconds.     A Saccharomyces cerevisiae model reveals in vivo functional impairment of the Ogden syndrome N-Terminal Acetyltransferase NAA10 Ser37Pro Mutant

    N-terminal acetylation (Nt-acetylation) occurs on the majority of eukaryotic proteins and is catalyzed by N-terminal acetyltransferases (NATs). Nt-acetylation is increasingly recognized as a vital modification with functional implications ranging from protein degradation to protein localization. Although early genetic studies in yeast demonstrated that NAT-deletion strains displayed a variety of phenotypes, only recently, the first human genetic disorder caused by a mutation in a NAT gene was reported; boys diagnosed with the X-linked Ogden syndrome harbor a p. Ser37Pro (S37P) mutation in the gene encoding Naa10, the catalytic subunit of the NatA complex, and suffer from global developmental delays and lethality during infancy. Here, we describe a Saccharomyces cerevisiae model developed by introducing the human wild-type or mutant NatA complex into yeast lacking NatA (NatA-Delta). The wild-type human NatA complex phenotypically complemented the NatA-Delta strain, whereas only a partial rescue was observed for the Ogden mutant NatA complex suggesting that hNaa10 S37P is only partially functional in vivo. Immunoprecipitation experiments revealed a reduced subunit complexation for the mutant hNatA S37P next to a reduced in vitro catalytic activity. We performed quantitative Nt-acetylome analyses on a control yeast strain (yNatA), a yeast NatA deletion strain (yNatA-Delta), a yeast NatA deletion strain expressing wild-type human NatA (hNatA), and a yeast NatA deletion strain expressing mutant human NatA (hNatA S37P). Interestingly, a generally reduced degree of Nt-acetylation was observed among a large group of NatA substrates in the yeast expressing mutant hNatA as compared with yeast expressing wild-type hNatA. Combined, these data provide strong support for the functional impairment of hNaa10 S37P in vivo and suggest that reduced Nt-acetylation of one or more target substrates contributes to the pathogenesis of the Ogden syndrome. Comparative analysis between human and yeast NatA also provided new insights into the co-evolution of the NatA complexes and their substrates. For instance, (Met-) Ala-N termini are more prevalent in the human proteome as compared with the yeast proteome, and hNatA displays a preference toward these N termini as compared with yNatA
