    Genetic diversity of Cornigliese sheep breed using STR markers

    To preserve sheep biodiversity is an important issue as the most productive breeds are becoming dependent on intensive management and breeders are abandoning the traditional breeds. The Cornigliese sheep is a local breed from Parma Apennine. In the last centuries it was crossed with Merino and Bergamasca breeds. The aim of this work is to investigate the genetic variability in the Cornigliese breed using 27 microsatellite markers. A total of 144 individual blood samples were randomly collected from the following breeds: Cornigliese, Bergamasca, Appenninica and Spanish Merino. The Palmera breed was used as out-group. A total of 344 alleles were detected. The average expected heterozygosity ranged from a maximum of 0.77 in Appenninica to a minimum of 0.68 in Bergamasca. In general,Cornigliese showed heterozygosity estimates less than expected. The Reynolds weighted genetic distance among the breeds highlighted a clear cluster, including Cornigliese and Bergamasca. As expected, they appeared in close neighbourhood in the tree because of the historic crossbreeding practices between these two breeds. Results of STRUCTURE analyses show that the four studied breeds were discriminated into separate clusters, even if Cornigliese has some individual related to Bergamasca and Spanish Merino. This work confirmed the historical information about the origin of Cornigliese breed which reports gene flow among these breeds. The genetic parameters show a moderate value of inbreeding in Cornigliese and therefore this breed should be monitored due the low number of individuals that compose it

    Serum IgG against Simian Virus 40 antigens are hampered by high levels of sHLA-G in patients affected by inflammatory neurological diseases, as multiple sclerosis

    Background: Many investigators detected the simian polyomavirus SV40 footprints in human brain tumors and neurologic diseases and recently it has been indicated that SV40 seems to be associated with multiple sclerosis (MS) disease. Interestingly, SV40 interacts with human leukocyte antigen (HLA) class I molecules for cell entry. HLA class I antigens, in particular non-classical HLA-G molecules, characterized by an immune-regulatory function, are involved in MS disease, and the levels of these molecules are modified according with the disease status. Objective: We investigated in serum samples, from Italian patients affected by MS, other inflammatory diseases (OIND), non-inflammatory neurological diseases (NIND) and healthy subjects (HS), SV40-antibody and soluble sHLA-G and the association between SV40-prevalence and sHLA-G levels. Methods: ELISA tests were used for SV40-antibodies detection and sHLA-G quantitation in serum samples. Results: The presence of SV40 antibodies was observed in 6 % of patients affected by MS (N = 4/63), 10 % of OIND (N = 8/77) and 15 % of NIND (N = 9/59), which is suggestive of a lower prevalence in respect to HS (22 %, N = 18/83). MS patients are characterized by higher sHLA-G serum levels (13.9 \ub1 0.9 ng/ml; mean \ub1 St. Error) in comparison with OIND (6.7 \ub1 0.8 ng/ml), NIND (2.9 \ub1 0.4 ng/ml) and HS (2.6 \ub1 0.7 ng/ml) subjects. Interestingly, we observed an inverse correlation between SV40 antibody prevalence and sHLA-G serum levels in MS patients. Conclusion: The data obtained showed a low prevalence of SV40 antibodies in MS patients. These results seems to be due to a generalized status of inability to counteract SV40 infection via antibody production. In particular, we hypothesize that SV40 immune-inhibitory direct effect and the presence of high levels of the immune-inhibitory HLA-G molecules could co-operate in impairing B lymphocyte activation towards SV40 specific peptides

    Fecal microbiota characterization of an Italian local horse breed

    The Bardigiano horse is a traditional native Italian breed with a rich history and peculiar characteristics. Local breeds are proven to have unique genetic traits developed over generations to adapt to defined geographical regions and/or conditions. The specific microbial communities that coexist within these animals are unraveled by studying their microbiota, which permits a further step in the characterization of local heritage. This work aimed to characterize Bardigiano horse fecal microbiota composition. The data obtained were then compared with published data of a mix of athlete breeds to evaluate potential differences among local and specialized breeds. The study involved 11 Bardigiano mares between 3 and 4 years of age, from which stool was sampled for the study. Samples were processed for 16S rRNA sequencing. Data obtained were analyzed and plotted using R, RStudio, and FastTree software. The samples analyzed were similar to what literature has reported on horses of other breeds and attitudes at higher taxonomic levels (from phylum to genera). While at lower taxonomic levels, the difference was more marked highlighting specific families found in the Bardigiano breed only. Weight, province of origin, and breeding sites significantly affected microbiota composition (p-value ≤0.02, p-value ≤0.04, and p-value ≤0.05, respectively). The comparison with athlete breed showed a significant difference confirming that animal and environmental factors are crucial in determining fecal microbiota composition (p-value <0.001). Understanding the microbiota composition in local breeds like the Bardigiano horse is crucial for preserving biodiversity, managing animal health, and promoting sustainable farming practices

    The Se-75 internal bioavailability was investigated in microalgae, mussels and rats as biological experimental models. The Se-75 accumulation from freshwater to microalgae [Scenedesmus obliquus (Turpin) Kutzing], from freshwater to mussels (Unio mancus Lamark) and, finally, per os to rats (Rattus norvegicus Berkenhout) was followed using Se-75-labelled selenite looking at Se-75 uptake, retention, intracellular distribution and binding with cellular biocomplexes. After exposure to 10, 50 and 500 mu g Se L-1, the microalgae showed an inhibitory effect on population growth only at the highest concentration. Mussels exposed to 105 mu g Se L-1 showed an accumulation of the element with time in all tissues. Intracellularly, Se was present in all subcellular fractions, especially in the cytosol. Rats were treated via oral administration with 5 mu g Se rat(-1). After 24 h, liver and kidney showed the highest Se concentration
