From compressibility to structural investigation of sodium dodecyl sulphate — Part 1: Powder and tablet physico-chemical characteristics
As a part of a study on detergent tablets, investigations were carried out to elucidate the compression behavior of a powdered surfactant, sodium dodecyl sulphate (SDS), based on a comparison with the main component of the formulation, i.e. the chorine provider (DCCNa). The compacted SDS exhibited poor cohesion as well as delayed dissolution whatever the compression pressure. The microscopic observations and the mercury porosimetry measurements both demonstrated that a residual porosity existed in the tablets but the dissolution times were always long. A modification of SDS in contact with water, forming a structure like a gel, probably occurred, inducing the closing of the pores and thereby limiting the water intrusion into the tablets

Staphylococcus aureus gene expression in a rat model of infective endocarditis
Background: Diabetes mellitus is a frequent underlying comorbidity in patients with Staphylococcus aureus endocarditis, and it represents a risk factor for complications and a negative outcome. The pathogenesis of staphylococcal endocardial infections in diabetic hosts has been poorly characterized, and little is known about S. aureus gene expression in endocardial vegetations. Methods: We utilized a rat model of experimental S. aureus endocarditis to compare the pathogenesis of staphylococcal infection in diabetic and nondiabetic hosts and to study the global S. aureus transcriptome in endocardial vegetations in vivo. Results: Diabetic rats had higher levels of bacteremia and larger endocardial vegetations than nondiabetic control animals. Microarray analyses revealed that 61 S. aureus genes were upregulated in diabetic rats, and the majority of these bacterial genes were involved in amino acid and carbohydrate metabolism. When bacterial gene expression in vivo (diabetic or nondiabetic endocardial vegetations) was compared to in vitro growth conditions, higher in vivo expression of genes encoding toxins and proteases was observed. Additionally, genes involved in the production of adhesins, capsular polysaccharide, and siderophores, as well as in amino acid and carbohydrate transport and metabolism, were upregulated in endocardial vegetations. To test the contribution of selected upregulated genes to the pathogenesis of staphylococcal endocarditis, isogenic deletion mutants were utilized. A mutant defective in production of the siderophore staphyloferrin B was attenuated in the endocarditis model, whereas the virulence of a surface adhesin (ΔsdrCDE) mutant was similar to that of the parental S. aureus strain. Conclusions: Our results emphasize the relevance of diabetes mellitus as a risk factor for infectious endocarditis and provide a basis for understanding gene expression during staphylococcal infections in vivo. The pea branching RMS2 gene encodes the PsAFB4/5 auxin receptor and is involved in an auxin-strigolactone regulation loop
Strigolactones (SLs) are well known for their role in repressing shoot branching. In pea,
increased transcript levels of SL biosynthesis genes are observed in stems of highly
branched SL deficient (ramosus1 (rms1) and rms5) and SL response (rms3 and rms4)
mutants indicative of negative feedback control. In contrast, the highly branched rms2
mutant has reduced transcript levels of SL biosynthesis genes. Grafting studies and hormone
quantification led to a model where RMS2 mediates a shoot-to-root feedback signal
that regulates both SL biosynthesis gene transcript levels and xylem sap levels of cytokinin
exported from roots. Here we cloned RMS2 using synteny with Medicago truncatula and
demonstrated that it encodes a putative auxin receptor of the AFB4/5 clade. Phenotypes
similar to rms2 were found in Arabidopsis afb4/5 mutants, including increased shoot branching,
low expression of SL biosynthesis genes and high auxin levels in stems. Moreover,
afb4/5 and rms2 display a specific resistance to the herbicide picloram. Yeast-two-hybrid
experiments supported the hypothesis that the RMS2 protein functions as an auxin receptor.
SL root feeding using hydroponics repressed auxin levels in stems and down-regulated transcript
levels of auxin biosynthesis genes within one hour. This auxin down-regulation was
also observed in plants treated with the polar auxin transport inhibitor NPA. Together these
data suggest a homeostatic feedback loop in which auxin up-regulates SL synthesis in an
RMS2-dependent manner and SL down-regulates auxin synthesis in an RMS3 and RMS4-
dependent manner
Flow-mediated-paradoxical vasoconstriction is independently associated with asymptomatic myocardial ischemia and coronary artery disease in type 2 diabetic patients.
International audienceBACKGROUND: To investigate whether flow-mediated dilation (FMD) impairment, which precedes overt atherosclerosis, is associated with silent myocardial ischemia (SMI) and asymptomatic coronary artery disease (CAD) in type 2 diabetes. METHODS: Forearm FMD was measured by ultrasonography in 25 healthy control, 30 non-diabetic overweight or obese patients and 118 asymptomatic type 2 diabetic patients with a high cardiovascular risk profile. SMI (abnormal stress myocardial scintiscan and/or stress dobutamine echocardiogram) and CAD (coronary angiography in the patients with SMI) were assessed in the diabetic cohort. RESULTS: FMD was lower in diabetic patients (median 0.61% (upper limits of first and third quartiles -1.22;3.2)) than in healthy controls (3.95% (1.43;5.25), p < 0.01) and overweight/obese patients (4.25% (1.74;5.56), p < 0.01). SMI was present in 60 diabetic patients, including 21 subjects with CAD. FMD was lower in patients with SMI than in those without (0.12% (-2.3;1.58) vs 1.64% (0;3.69), p < 0.01), with a higher prevalence of paradoxical vasoconstriction (50.0% vs 29.3%, p < 0.05). FMD was also lower in patients with than without CAD (-1.22% (-2.5;1) vs 1.13% (-0.4;3.28), p < 0.01; paradoxical vasoconstriction 61.9% vs 34.4%, p < 0.05). Logistic regression analyses considering the parameters predicting SMI or CAD in univariate analyses with a p value <0.10 showed that paradoxical vasoconstriction (odds ratio 2.7 [95% confidence interval 1.2-5.9], p < 0.05) and nephropathy (OR 2.6 [1.2-5.7], p < 0.05) were independently associated with SMI; and only paradoxical vasoconstriction (OR 3.1 [1.2-8.2], p < 0.05) with CAD. The negative predictive value of paradoxical vasoconstriction to detect CAD was 88.7%. CONCLUSIONS: In diabetic patients, FMD was independently associated with SMI and asymptomatic CAD.Trial registration: Trial registration number NCT00685984

Investigations were carried out to elucidate the compression behavior of a powdered surfactant, sodium dodecyl sulphate (SDS), based on a comparison with the main component of a detergent formulation, i.e. the chorine provider (DCCNa). The energetic analysis based on the compression cycles highlighted a lower compressibility of SDS compared with DCCNa, especially due to its worse packing ability, larger
elasticity and bad cohesion ability. Also, it pointed out that the pycnometric density seemed to be overrun under pressure whereas a residual porosity had been evidenced in the expanded tablets. DSC/DTA analysis, Raman spectroscopy as well as powder X-ray diffraction refuted the hypothesis of a physico-chemical transformation of SDS under pressure. This was in accordance with the morphology of the SDS particles, quite unchanged after compression. The pycnometric density measurements have been improved; firstly, it allowed to properly express the compaction
ratio of the ejected SDS tablets, and secondly, it led to conclude to a reversible intrinsic compressibility for pressures higher than 50 MPa, explained by the predominant elastic behavior of SDS
