Estimating mechanical properties of cellular solid materials from additive manufacturing processes
the inherent limitations of the AM process. This research aims to develop a mechanical property-estimation procedure for additively manufactured cellular materials by considering the effects of AM processes. In order to clearly understand the AM process, related parameters are categorized into four groups: (a) Design and Manufacturing process parameters (DMP), (b) Layer deposition parameters (LDP), (c) Structural element parameters (SEP), and (d) Cellular material properties (CMP). Three transformations are defined among these groups. Firstly, the functional relationship between DMPs and LDPs is established based on process-modeling simulation. The variation in LDPs due to manufacturing instabilities is quantified in the form of a stochastic distribution. Next, an as-fabricated voxel modeling approach is developed for describing the propagation of geometrical degradation from LDPs to SEPs. The effective values of SEPs are determined based on semi-rigid joint frame element formulation. Finally, a discrete homogenization approach is implemented with the semi-rigid elements to integrate the effects of AM processes into the mechanical XVI property estimation procedure. The estimation framework developed in this research can be applied to analyze the performance of additively manufactured cellular materials and help to design of cellular materials.Ph.D
Increased Immunoendocrine Cells in Intestinal Mucosa of Postinfectious Irritable Bowel Syndrome Patients 3 Years after Acute Shigella Infection - An Observation in a Small Case Control Study
PURPOSE: Postinfectiously irritable bowel syndrome (PI-IBS) develops in 3-30% of individuals with bacterial gastroenteritis. Recent studies demonstrated increases in inflammatory components in gut mucosa of PI-IBS patients even after complete resolution of infection. We aimed to investigate histological changes in colon and rectum of PI-IBS subjects after long term period of infection.
MATERIALS AND METHODS: We recruited PI-IBS subjects who had been diagnosed IBS after complete resolution of enteritis caused by shigellosis outbreak 3 years earlier. We compared unmatched four groups, PI-IBS (n = 4), non PI-IBS (n = 7), D-IBS (n = 7, diarrhea predominant type) and healthy controls (n = 10). All of them underwent colonoscopic biopsy at three areas, including descending colon (DC), sigmoid colon (SC) and rectum, which were assessed for 5-hydroxytryptamine (5-HT)/peptide YY (PYY)-containing enterochromaffin (EC) cell, intraepithelial (IEL) and lamina propria T lymphocyte (CD3), CD8 lymphocytes, mast cells and CD68/calprotectin+ macrophages.
RESULTS: All subjects had no structural or gross abnormalities at colonoscopy. In PI-IBS, 5-HT containing EC cells, PYY containing EC cells, IELs, CD3 lymphocytes, CD8 lymphocytes, mast cells, and CD68 + macrophages were increased compared to control (p < 0.05). In D-IBS, PYY containing EC cells, IELs, and CD3 lymphocytes were increased compared to control (p < 0.05). In PI-IBS, 5-HT containing EC cells tended to increase and PYY containing EC cells, CD8 lymphocytes, mast cells, and CD68+ macrophages were increased compared to non PI-IBS (p < 0.05). Calprotectin + marcrophages were decreased in PI-IBS, non PI-IBS and IBS compared to control.
CONCLUSION: The immunoendocrine cells were sporadically increased in PI-IBS, non PI-IBS and D-IBS compared with control. Our findings in a very small number of patients suggest that mucosal inflammation may play a role in long-term PI-IBS, and that other sub-groups of IBS and larger scale studies are needed to confirm this observation.ope

Discovery of Novel Sources of Vitamin B12 in Traditional Korean Foods from Nutritional Surveys of Centenarians
Human longevity can be explained by a variety of factors, among them, nutritional factor would play an important role. In our study of Korean centenarians for their longevity, the apparent nutritional imbalance in the traditional semi-vegetarian diet raised a special attention, especially on vitamin B12 status, supplied by animal foods. Interestingly, we found that the prevalence of vitamin B12 deficient Korean centenarians was not higher compared with those from Western nations with animal-oriented traditional foods. We assumed that there might be some unveiled sources for vitamin B12 in the Korean traditional foods. Screening of vitamin B12 contents has revealed that some traditional soybean-fermented foods, such as Doenjang and Chunggukjang, and seaweeds contain considerable amounts of vitamin B12. Taken together, it can be summarized that the traditional foods, especially of fermentation, might be evaluated for compensation of the nutritional imbalance in the vegetable-oriented dietary pattern by supplying vitamin B12, resulting in maintenance of health status
UVB Induces HIF-1α-Dependent TSLP Expression via the JNK and ERK Pathways
Thymic stromal lymphopoietin (TSLP) may have a key role in the initiation and maintenance of allergic inflammatory diseases, including atopic dermatitis. The present study revealed that UVB radiation exposure could induce TSLP expression in human keratinocytes and a human skin equivalent model. In addition, we investigated the regulatory mechanism of UVB-induced TSLP expression in keratinocytes. TSLP expression was upregulated by transfection with pcDNA3–hypoxia-inducible factor (HIF)-1α (P402A and P564A), which stably expresses HIF-1α protein. UVB-induced TSLP induction in keratinocytes was suppressed in the treatment of mitogen-activated protein kinase inhibitors or small interfering RNAs against HIF-1α. The results of chromatin immunoprecipitation assays indicate the direct involvement of HIF-1α in UVB-mediated TSLP induction. Taken together, these findings indicate that UVB exposure may increase TSLP expression through a HIF-1α-dependent mechanism via the c-JUN N-terminal kinase and extracellular signal-regulated kinase pathways in human keratinocytes. Our data showed that UVB-induced TSLP might increase secretion of the T-helper type 2–attracting chemokine (c–c motif) ligand 17 by human dendritic cells. The present study suggests an important role of HIF-1α in UVB-mediated immune response in keratinocytes
Rapid quantification of DNA methylation through dNMP analysis following bisulfite-PCR
We report a novel method for rapid quantification of the degree of DNA methylation of a specific gene. Our method combined bisulfite-mediated PCR and quantification of deoxyribonucleoside monophosphate (dNMP) contents in the PCR product through capillary electrophoresis. A specific bisulfite-PCR product was enzymatically hydrolyzed to dNMP monomers which were quantitatively analyzed through subsequent capillary electrophoresis. PCR following bisulfite treatment converts unmethylated cytosines to thymines while leaving methyl-cytosines unchanged. Then the ratio of cytosine to thymine determined by capillary electrophoresis represents the ratio of methyl-cytosine to cytosine in genomic locus of interest. Pure oligonucleotides with known sequences were processed in parallel as standards for normalization of dNMP peaks in capillary electrophoresis. Sources of quantification uncertainty such as carryovers of dNTPs or primers and incomplete hydrolysis were examined and ruled out. When the method was applied to samples with known methylation levels (by bisulfite-mediated sequencing) as a validation, deviations were within ±5%. After bisulfite-PCR, the analytical procedure can be completed within 1.5 h
Phenylalanine ammonia-lyase expression and pyranocoumarin accumulation in Angelica gigas plantlets exposed to light-emitting diodes
Angelica gigas (Dang Gui) is an important medicinal plant. In this study, we examined the accumulation of pyranocoumarin (decursin and decursinol angelate) and the expression of phenylalanine ammonia-lyase (PAL) in Korean angelica plantlet grown under different light-emitting diodes (LEDs) (red, orange, green, blue, and white). Three weeks after LED exposure (WAE), the transcript levels of phenylalanine ammonia-lyase mRNA in seedlings grown under orange LEDs were 4-, 18-, and 7-fold higher than those in seedlings grown under green, blue, and white LEDs, respectively. The decursinol angelate content was almost double than the decursin content. The highest levels of decursin (3.2 mg/g dry weight) and decursinol angelate (6 mg/g dry weight) were detected in plants grown under orange LEDs, at 2 WAE. Therefore, we suggest that orange LEDs may affect decursin and decursinol angelate accumulation. The findings of this study could help to determine an effective strategy for producing secondary metabolites in A. gigas using LED technology
Attribution of the 2015 record high sea surface temperatures over the central equatorial Pacific and tropical Indian Ocean
This study assessed the anthropogenic contribution to the 2015 record-breaking high sea surface temperatures (SSTs) observed in the central equatorial Pacific and tropical Indian Ocean. Considering a close link between extreme warm events in these regions, we conducted a joint attribution analysis using a fraction of attributable risk approach. Probability of occurrence of such extreme anomalies and long-term trends for the two oceanic regions were compared between CMIP5 multi-model simulations with and without anthropogenic forcing. Results show that the excessive warming in both regions is well beyond the range of natural variability and robustly attributable to human activities due to greenhouse gas increase. We further explored associated mechanisms including the Bjerknes feedback and background anthropogenic warming. It is concluded that background warming was the main contribution to the 2015 extreme SST event over the central equatorial Pacific Ocean on a developing El Niño condition, which in turn induced the extreme SST event over the tropical Indian Ocean through the atmospheric bridge effect.113Ysciescopu
Reconstituting ring-rafts in bud-mimicking topography of model membranes.
During vesicular trafficking and release of enveloped viruses, the budding and fission processes dynamically remodel the donor cell membrane in a protein- or a lipid-mediated manner. In all cases, in addition to the generation or relief of the curvature stress, the buds recruit specific lipids and proteins from the donor membrane through restricted diffusion for the development of a ring-type raft domain of closed topology. Here, by reconstituting the bud topography in a model membrane, we demonstrate the preferential localization of cholesterol- and sphingomyelin-enriched microdomains in the collar band of the bud-neck interfaced with the donor membrane. The geometrical approach to the recapitulation of the dynamic membrane reorganization, resulting from the local radii of curvatures from nanometre-to-micrometre scales, offers important clues for understanding the active roles of the bud topography in the sorting and migration machinery of key signalling proteins involved in membrane budding
