    Cloning and expression analysis of a Petunia hybrida flower specific mitotic-like cyclin

    AbstractA cyclin cDNA clone (Pethy;CycB1;1) was isolated from a Petunia hybrida ovary specific cDNA library. Sequence comparison revealed that Pethy;CYCB1;1 protein is highly homologous to mitotic B1 cyclins. Northern analysis and in situ hybridisation experiments showed that its expression is developmentally regulated and restricted to flower organs. We have attempted to define some of the cell division patterns which contribute to shaping each floral organ by analysing Pethy;CycB1;1 expression on Petunia flower sections. While in sepals, epidermis and parenchyma cell division patterns were comparable, there were two distinct cell division patterns in petals. In the epidermis, Pethy;CYCB1;1 expression was found both at the petal tip and along epidermis, whereas in the parenchyma only at the petal tips. In reproductive organs cell divisions were detected only in sporophytic tissues. No signals were detected inside meiotic cells

    Depositi carbonatici infrapleistocenici di tipo foramol in sistemi di scarpata (Salento – Italia meridionale)

    Along the eastern Salento coast (Southern Italy), Cretaceous to Quaternary carbonates crop out on a 100 m high escarpment that connects a wide and relatively flat area to the Otranto Strait (Ionian Sea). Along the escarpment, faulted and tilted Cretaceous and Eocene limestones record deposition in shallow-marine and marginal carbonate environments. Younger carbonates disconformably overlie this substratum and they formed when the region was partially submerged. Accordingly, Priabonian to Messinian carbonates which crop out along the escarpment were interpreted as slope deposits linked to reef systems developed on the margin of the top area, while lower Pleistocene carbonates cropping out at the base of the same escarpment were considered coastal in origin and formed during the uplift of the region. Stratigraphic and sedimentologic studies carried out on these lower Pleistocene carbonates demonstrate that also these deposits should be considered as slope deposits. The studied carbonates are foramol-type ones and discontinuously crop out along the lower part of the escarpment. They show a variable thickness (from a few meters up to several tens) and often exhibit a fan morphology; they correspond to small isolated bodies (up to a few km2 wide) developed in some indentations of the escarpment, and are composed of coarse skeletal grains mainly deposited via slumpings or grain flows. Successions are characterized by long clinobeds cut by slump scars downward passing to gullies; backsets, slumps and chaotic deposits fill these erosional features. Clinobeds are alternatively made up of molechfor and rodhalgal facies which respectively record resedimentation by grain flows of shallow marine carbonates developed on top of the region and along-slope carbonate production. Depositional systems correspond to small shallow-marine aprons whose discontinuous distribution along the escarpment (inside indentations) should be considered an original feature. The idea that these deposits record the Sicilian sea-level along the escarpment should be abandoned as they are not coastal deposits. The studied deposits should be correlated to the Calcarenite di Gravina Formation which in other areas of the Apulian Foreland (which the Salento region belongs to) are considered as the deposits linked to the subsidence induced by the estward migration of the south- Apennines orogenic system. The Quaternary uplift of the Salento region should be begun after the deposition of the studied slope deposits, as indicated also by a series of middle-upper Pleistocene marine terraces which start at heights higher than those ones of the studied deposits

    “Chama a Roda” na Escola Família Agrícola de Santa Cruz do Sul: o círculo de cultura reinventado na pedagogia da alternância

    Este é um artigo feito a várias mãos e que tem como objetivo apresentar algumas reflexões sobre o processo de ensino-aprendizagem existentes na Escola Família Agrícola de Santa Cruz do Sul (EFASC) a partir da sua particular experiência e da recepção da Pedagogia da Alternância à luz da epistemologia freiriana. Trazemos falas de educandos/as e educadores/as dessa escola que são decorrentes do processo de sistematização de informações que, no presente trabalho, recebem tratamento, percepções e indagações que são frutos do envolvimento no cotidiano escolar. Faz-se um percurso de constituição histórica dessa pedagogia e a sua recepção na EFASC seguindo para a reflexão sobre a relação ensino-aprendizagem apontando para uma educação contextualizada na região do Vale do Rio Pardo, Rio Grande do Sul. A roda se apresenta como espaço pedagógico dialógico que contextualiza e problematiza as relações humanas, sociais e produtivas dos povos do campo buscando a transformação da realidade. Entende-se que: cada sujeito é único e é valorizado em sua inteireza; a Pedagogia da Alternância se fundamenta nos componentes do “vivido-pensado” na busca pela transformação; cada sujeito, implicado no processo alternante, educa-se em comunhão; ensinam e aprendem em sua diversidade, nunca como desiguais – (re)conhecendo-se como sujeitos do campo; e educar-se significa “ler crítica e criativamente o seu próprio mundo”. Dessa forma, os instrumentos pedagógicos construídos pela/na EFASC reinventam a Pedagogia da Alternância ao apoiar-se em Paulo Freire. Superam, com isso, a caracterização da roda como sendo um simples método. E, ao fazê-lo, assumem a existência de um legado tanto metodológico quanto epistemológico

    Free-range rearing density for male and female Milanino chickens: carcass yield and qualitative meat traits

    SUMMARY The Milanino is a heavy Italian chicken breed and represents an important genetic resource for alternative production systems. In order to support its promotion in the market according to consumer expectations on healthy nutrition and animal welfare, this trial aims to study the slaughter performance and the meat quality in male and female Milanino chickens kept at different rearing density in a separate-sex free-range system. A total of 140 birds (70M:70F) were reared in outdoor pens from 75 to 235 d of life according to the following experimental groups (35 birds/group): M2) males in 2 m2/bird; F2) females in 2 m2/bird; M10) males in 10 m2/bird; F10) females in 10 m2/bird. At 235 d of age, 6 birds per group were slaughtered. Slaughter performance and meat quality were assessed. The Milanino chicken is characterized by high carcass weight and carcass yield among local chicken breeds, and a rearing density of 10 m2/bird is recommended for male birds to obtain heavier carcasses. Milanino meat appears bright and intensely colored, and it is characterized by high protein and low fat contents compared with the standard broiler meat. Total lipids of Milanino meat are characterized by a healthy fatty acid composition, corresponding to a high PUFA/SFA ratio. Milanino breast meat is a good supply of nutraceutical PUFA with a positive low n-6/n-3 PUFA ratio. The ability of the Milanino breed either to synthesize or to transfer to tissue a high quantity of PUFA relevant for human health could be a key factor for its economic valorization

    Primary DNA damage and genetic polymorphisms for CYP1A1, EPHX and GSTM1 in workers at a graphite electrode manufacturing plant

    <p>Abstract</p> <p>Background</p> <p>The results of a cross-sectional study aimed to evaluate whether genetic polymorphisms (biomarkers of susceptibility) for <it>CYP1A1</it>, <it>EPHX </it>and <it>GSTM1 </it>genes that affect polycyclic aromatic hydrocarbons (PAH) activation and detoxification might influence the extent of primary DNA damage (biomarker of biologically effective dose) in PAH exposed workers are presented. PAH-exposure of the study populations was assessed by determining the concentration of 1-hydroxypyrene (1OHP) in urine samples (biomarker of exposure dose).</p> <p>Methods</p> <p>The exposed group consisted of workers (n = 109) at a graphite electrode manufacturing plant, occupationally exposed to PAH. Urinary 1OHP was measured by HPLC. Primary DNA damage was evaluated by the alkaline comet assay in peripheral blood leukocytes. Genetic polymorphisms for <it>CYP1A1</it>, <it>EPHX</it> and <it>GSTM1</it> were determined by PCR or PCR/RFLP analysis.</p> <p>Results</p> <p>1OHP and primary DNA damage were significantly higher in electrode workers compared to reference subjects. Moreover, categorization of subjects as normal or outlier highlighted an increased genotoxic risk OR = 2.59 (CI95% 1.32–5.05) associated to exposure to PAH. Polymorphisms in <it>EPHX</it> exons 3 and 4 was associated to higher urinary concentrations of 1OHP, whereas none of the genotypes analyzed (<it>CYP1A1</it>, <it>EPHX</it>, and <it>GSTM1</it>) had any significant influence on primary DNA damage as evaluated by the comet assay.</p> <p>Conclusion</p> <p>The outcomes of the present study show that molecular epidemiology approaches (i.e. cross-sectional studies of genotoxicity biomarkers) can play a role in identifying common genetic risk factors, also attempting to associate the effects with measured exposure data. Moreover, categorization of subjects as normal or outlier allowed the evaluation of the association between occupational exposure to PAH and DNA damage highlighting an increased genotoxic risk.</p

    3-hydroxy-L-kynurenamine is an immunomodulatory biogenic amine

    Tryptophan catabolism is a major metabolic pathway utilized by several professional and non-professional antigen presenting cells to maintain immunological tolerance. Here we report that 3-hydroxy-l-kynurenamine (3-HKA) is a biogenic amine produced via an alternative pathway of tryptophan metabolism. In vitro, 3-HKA has an anti-inflammatory profile by inhibiting the IFN-gamma mediated STAT1/NF-kappa Beta pathway in both mouse and human dendritic cells (DCs) with a consequent decrease in the release of pro-inflammatory chemokines and cytokines, most notably TNF, IL-6, and IL12p70. 3-HKA has protective effects in an experimental mouse model of psoriasis by decreasing skin thickness, erythema, scaling and fissuring, reducing TNF, IL-1 beta, IFN-gamma, and IL-17 production, and inhibiting generation of effector CD8(+) T cells. Similarly, in a mouse model of nephrotoxic nephritis, besides reducing inflammatory cytokines, 3-HKA improves proteinuria and serum urea nitrogen, overall ameliorating immune-mediated glomerulonephritis and renal dysfunction. Overall, we propose that this biogenic amine is a crucial component of tryptophan-mediated immune tolerance. 3-hydroxy-L-kynurenamine (3-HKA) is a metabolite deriving from a lateral pathway of tryptophan catabolism. Here the authors identify 3-HKA as a biogenic amine and show it has anti-inflammatory properties that can protect mice against psoriasis and nephrotoxic nephritis.Peer reviewe
