    Hippocampal structural plasticity accompanies the resulting contextual fear memory following stress and fear conditioning

    The present research investigated the resulting contextual fear memory and structural plasticity changes in the dorsal hippocampus (DH) following stress and fear conditioning. This combination enhanced fear retention and increased the number of total and mature dendritic spines in DH. Intra-basolateral amygdala (BLA) infusion of midazolam prior to stress prevented both the enhancement of fear retention and an increase in the density of total and mature dendritic spines in DH. These findings emphasize the role of the stress-induced attenuation of GABAergic neurotransmission in BLA in the promoting influence of stress on fear memory and on synaptic remodeling in DH. In conclusion, the structural remodeling in DH accompanied the facilitated fear memory following a combination of fear conditioning and stressful stimulation.Fil: Giachero, Marcelo.     A BDNF sensitive mechanism is involved in the fear memory resulting from the interaction between stress and the retrieval of an established trace

    The present study investigates the fear memory resulting from the interaction of a stressful experience and the retrieval of an established fear memory trace. Such a combination enhanced both fear expression and fear retention in adult Wistar rats. Likewise, midazolam intra-basolateral amygdala (BLA) infusion prior to stress attenuated the enhancement of fear memory thus suggesting the involvement of a stress-induced reduction of the GABAergic transmission in BLA in the stress-induced enhancing effect. It has been suggested that, unlike the immediate-early gene Zif268 which is related to the reconsolidation process, the expression of hippocampal brain-derived neurotrophic factor (BDNF) is highly correlated with consolidation. We therefore evaluate the relative contribution of these two neurobiological processes to the fear memory resulting from the above-mentioned interaction. Intra-dorsal hippocampus (DH) infusions of either the antisense Zif268 or the inhibitor of the protein degradation (Clasto-Lactacystin β-Lactone), suggested to be involved in the retrieval-dependent destabilization process, did not affect the resulting contextual memory. In contrast, the knockdown of hippocampal BDNF mitigated the stress-induced facilitating influence on fear retention. In addition, the retrieval experience elevated BDNF level in DH at 60 min after recall exclusively in stressed animals. These findings suggest the involvement of a hippocampal BDNF sensitive mechanism in the stress-promoting influence on the fear memory following retrieval.Fil: Giachero, Marcelo.     The effect of Midazolam and Propranolol on fear memory reconsolidation in ethanol-withdrawn rats: Influence of D-cycloserine

    Background: Withdrawal from chronic ethanol facilitates the formation of contextual fear memory and delays the onset to extinction, with its retrieval promoting an increase in ethanol consumption. Consequently, manipulations aimed to reduce these aversive memories, may be beneficial in the treatment of alcohol discontinuation symptoms. Related to this, pharmacological memory reconsolidation blockade has received greater attention due to its therapeutic potential. Methods: Here, we examined the effect of post-reactivation amnestic treatments such as Midazolam (MDZ, 3 mg/kg i.p) and Propranolol (PROP, 5 mg/kg i.p) on contextual fear memory reconsolidation in ethanol- withdrawn (ETOH) rats. Next, we examined whether the activation of N-methyl-D-aspartate (NMDA) receptors induced by d-cycloserine (DCS, 5 mg/kg i.p., a NMDA partial agonist) before memory reactivation can facilitate the disruptive effect of PROP and MDZ on fear memory in ETOH rats. Results: We observed a resistance to the disruptive effect of both MDZ and PROP following memory reactivation. Although intra-basolateral amygdala (BLA; 1.25 ug/side) and systemic PROP administration attenuated fear memory in DCS pre-treated ETOH rats, DCS/MDZ treatment did not affect memory in these animals. Finally, a decrease of both total and surface protein expression of the α1 GABAA receptor (GABAA-R) subunit in BLA was found in the ETOH rats. Conclusions: Ethanol withdrawal facilitated the formation of fear memory resistant to labilization post-reactivation. DCS administration promoted the disruptive effect of PROP on memory reconsolidation in ETOH rats. The resistance to MDZ's disruptive effect on fear memory reconsolidation may be, at least in part, associated with changes in the GABAA-R composition induced by chronic ethanol administration/withdrawal.Fil: Ortiz, Vanesa.     Polymeric near-infrared absorbing dendritic nanogels for efficient in vivo photothermal cancer therapy

    In recent years, several near-infrared light absorbing inorganic nanomaterials have been developed for photothermal therapy. However, their biological fate after injection limits their clinical utilization. In this work, we developed a novel polymeric near-infrared light absorbing material based on a biocompatible thermoresponsive nanogel that is semi-interpenetrated with polyaniline, a conjugated polymer with strong near-infrared absorbance. This polymeric nanocomposite generates heat after being irradiated by NIR light, thereby inducing a local hyperthermia that is used for photothermal cancer therapy in vitro and in vivo

    GIS without GPS: new opportunities in technology and survey research to link people and place

    This paper presents innovative ways to relate survey data to GIS maps, thereby making the connection of people and place more accessible for the research community. Based on data from rural areas in the Brazilian Amazon, we describe a successful effort to sample households while linking farm-level data to property boundaries, these boundaries generated from subjects’ interpretations of satellite images on a computer screen. The sampling framework is based on legislation requiring farmers to report to a government agency in a four-week period, and the farmers’ input allows for a more efficient means of identifying property boundaries as compared to GPS. We show that the resulting sampling is statistically representative. We discuss the potential of this association of institutional design and low-cost methods of data collection to allow for more cost-effective generation of spatial data and of geospatial analysis

    Actitud del residente hacia el turismo y el visitante : factores determinantes en el turismo y excursionismo rural-cultural

    [email protected] [email protected] la literatura sobre turismo se han planteado diversos modelos con el fin de analizar los efectos directos y/o indirectos de los impactos percibidos del turismo y la valoración de dichos efectos realizada por los residentes en el destino turístico. El objetivo del estudio es analizar la actitud de los residentes en un destino turístico de naturaleza rural-cultural respecto a los visitantes, ya sean turistas o excursionistas en el destino objeto de estudio. Asimismo, se examinan los factores susceptibles de influir en la formación de dicha actitud y su potencial uso en el desarrollo turístico del destino. Como resultado de nuestro estudio, se observan actitudes hacia el turismo significativamente diferentes en función del grado de desarrollo local y del uso de recursos turísticos por parte de los residentes.In the literature on tourism several models have been proposed in order to analyze the direct and/or indirect effects of the perceived impacts of tourism and the assessment of such effects by the resident community in the tourist destination. The objective of this study is to analyze resident attitude in a rural-cultural destination towards visitors, either tourists or day-trippers in the tourism destination. Furthermore, we analyze the potential factors likely to influence on attitude formation and its potential use in the development of the tourist destination. As a result of our study, we observe significantly different attitudes towards tourism depending on degree of local development and use of tourism resources by residents

    Estudio comparativo de desempeño del lenguaje de niños normales, nacidos asfixiados y nacidos prematuros mediante el uso de una escala de hitos tempranos del Lenguaje

    62 p.El presente trabajo, desarrollado sobre la práctica clínica del autor, plantea la necesidad de establecer qué ocurre al comparar el desempeño de tres grupos de niños usando un instrumento que evalúa los hitos tempranos del lenguaje. Estos grupos, que conforman la muestra del trabajo son: un grupo de prematuros de muy bajo peso al nacer, un grupo de asfixiados, y un grupo de niños normales, que se controlan en atención primaria. Los tres grupos son relevantes a la realidad nacional. La sobrevida de los nacidos prematuros con muy bajo peso es cada vez mayor gracias al desarrollo tecnológico, y su desarrollo de lenguaje ha sido escasamente estudiado en nuestro medio. Por otra parte, aunque exista atención profesional del parto, un grupo reducido de niños de embarazo de término tendrá asfixia al nacer, lo que justifica un programa de seguimiento formal de ellos. Por último, el grupo de niños normales, controlados a nivel de Atención Primaria en salud, es evaluado gruesamente en relación a su desarrollo de lenguaje, utilizando instrumentos no confiables. En el manejo de este grupo mayoritario, no existe una postura de consenso en relación a qué intervenciones son adecuadas cuando se pesquisan desviaciones en el desarrollo de lenguaje. Hasta este trabajo, no existen reportes nacionales que den cuenta de la situación de desarrollo temprano del lenguaje y de su valor predictivo, en estos grupos en particular./ Abstract:The present work, developed over the author practice, state the need of establish what happens to confront the performance on three groups of children using an instrument that assess early language milestones. These groups that conforms the sample are: a group prematurely born, an asphyxia group and a normal group that are controlled in primary care. All of these groups are relevant to Chilean reality. Prematurely born survival is greater thanks technologic development, and its language development has been poorly studied in our country. Otherwise, although there were medical cares on delivery, some children of term will have asphyxia when they are born, what justify formal follow up of them. Finally, the normal group which is controlled in primary care is assessed grossly in language development using instruments without reliability. In the management of this group there is no agreement in what interventions are adequate when deviations are detected in language development.     Chitosan-g-oligo(epsilon-caprolactone) polymeric micelles: microwave-assisted synthesis and physicochemical and cytocompatibility characterization

    With the aim to produce mucoadhesive polymeric micelles for drug administration by mucosal routes, chitosan-g-oligo(epsilon-caprolactone) copolymers were synthesized by the microwave-assisted ring-opening polymerization of epsilon-caprolactone using chitosan as the macroinitiator and methanesulfonic acid as the solvent, catalyst and protecting group of the amine moieties. The reaction was conducted under very mild conditions and was completed within 10 min with a monomer conversion above 90%. The grafting of oligo(epsilon-caprolactone) blocks to the free hydroxyl groups of chitosan was confirmed by ATR/FT-IR, 1H- and 13C-NMR, WAXD and thermal analysis (TGA/DSC). The molecular weight of the synthetic hybrid copolymers was determined by GPC and MALDI-ToF mass spectrometry. Polymeric micelles obtained by the solvent diffusion/evaporation method showed a spherical shape (TEM and AFM) with sizes between 111 and 154 nm and highly positive zeta potential (>+50 mV) (DLS). In addition, they displayed good cell compatibility in the human lung adenocarcinoma epithelial line, A549, and were readily up-taken by the cervical cancer cell line, HeLa. Results from the encapsulation of the antituberculosis drug, rifampicin, showed that the micelles had better performance than other nanocarriers previously investigated (e.g., cyclodextrins). Moreover, the micelles conserved the mucoadhesiveness displayed by pristine chitosan and are expected to transiently open tight cell junctions and lead to more prolonged residence times in mucosal tissues and greater drug bioavailability.Fil: Glisoni, Romina Julieta.
