Bone Marrow B cell Precursor Number after Allogeneic Stem Cell Transplantation and GVHD Development

Patients without chronic graft-versus-host disease (cGVHD) have robust B cell reconstitution and are able to maintain B cell homeostasis after allogeneic hematopoietic stem cell transplantation (HSCT). To determine whether B lymphopoiesis differs before cGVHD develops, we examined bone marrow (BM) biopsies for terminal deoxynucleotidyl transferase (TdT) and PAX5 immunostaining early post-HSCTat day 30 when all patients have been shown to have high B cell activating factor (BAFF) levels. We found significantly greater numbers of BM B cell precursors in patients who did not develop cGVHD compared with those who developed cGVHD (median = 44 vs 2 cells/high powered field [hpf]; respectively; P < .001). Importantly, a significant increase in precursor B cells was maintained when patients receiving high-dose steroid therapy were excluded (median = 49 vs 20 cells/hpf; P =.017). Thus, we demonstrate the association of BM B cell production capacity in human GVHD development. Increased BM precursor B cell number may serve to predict good clinical outcome after HSCT

Derisking the Polymorph Landscape: The Complex Polymorphism of Mexiletine Hydrochloride

This work presents an updated solid form discovery approach to the polymorphism of the anti-arrhythmic drug mexiletine hydrochloride, in which experimental and computational techniques are combined to provide a rigorous characterisation of the solid-form landscape of this compound. The resulting solid forms were characterised by powder and single crystal X-ray diffraction, IR spectroscopy, DSC, and 13C solid-state NMR. This approach reveals five solid form types of mexiletine hydrochloride. Forms 1, 2 and 3 are mutually enantiotropically related anhydrous polymorphs, with Form 1 the room temperature stable form, Form 2 the high temperature form and Form 3 is the thermodynamically stable polymorph between 148 °C and 167 °C. The final two forms termed Types A and B comprise two large families of isomorphous channel solvates, including a fourth non-solvated form isostructural to the type A solvates. We report eleven modifications of each solvate, in which a diverse range of solvents are included in the channels, without changing the fundamental structure of the drug framework. These experimental results go hand-in-hand with computational crystal structure prediction (using the AstraZeneca crystal structure prediction approach), which together suggest that it is unlikely further non-solvated forms, at least with Z' = 1, will be discovered under ambient conditions

La laïcité en question

Cet ouvrage inscrit la réflexion sur la laïcité dans la longue durée de l’histoire française et allemande, tout en croisant les approches disciplinaires, afin d’éclairer respectivement chacun des deux modèles dans sa spécificité. Divers éléments entrent en ligne de compte pour ce faire : outre le critère de neutralité réciproque du politique et du religieux, le respect de la liberté religieuse dans ses diverses facettes, la séparation des Églises et de l’État sous ses différentes formes, le statut juridique accordé aux sociétés religieuses, la mise en oeuvre du droit à l’incroyance.
