    Guanabenz Acetate Induces Endoplasmic Reticulum Stress–Related Cell Death in Hepatocellular Carcinoma Cells

    Background Development of chemotherapeutics for the treatment of advanced hepatocellular carcinoma (HCC) has been lagging. Screening of candidate therapeutic agents by using patient-derived preclinical models may facilitate drug discovery for HCC patients. Methods Four primary cultured HCC cells from surgically resected tumor tissues and six HCC cell lines were used for high-throughput screening of 252 drugs from the Prestwick Chemical Library. The efficacy and mechanisms of action of the candidate anti-cancer drug were analyzed via cell viability, cell cycle assays, and western blotting. Results Guanabenz acetate, which has been used as an antihypertensive drug, was screened as a candidate anti-cancer agent for HCC through a drug sensitivity assay by using the primary cultured HCC cells and HCC cell lines. Guanabenz acetate reduced HCC cell viability through apoptosis and autophagy. This occurred via inhibition of growth arrest and DNA damage-inducible protein 34, increased phosphorylation of eukaryotic initiation factor 2α, increased activating transcription factor 4, and cell cycle arrest. Conclusions Guanabenz acetate induces endoplasmic reticulum stress–related cell death in HCC and may be repositioned as an anti-cancer therapeutic agent for HCC patients

    A single nucleotide polymorphism in CAPN1 associated with marbling score in Korean cattle

    <p>Abstract</p> <p>Background</p> <p>Marbling score (MS) is the major quantitative trait that affects carcass quality in beef cattle. In this study, we examined the association between genetic polymorphisms of the micromolar calcium-activated neutral protease gene (micro-calpain, <it>CAPN1</it>) and carcass traits in Korean cattle (also known as Hanwoo).</p> <p>Results</p> <p>By direct DNA sequencing in 24 unrelated Korean cattle, we identified 39 sequence variants within exons and their flanking regions in <it>CAPN1</it>. Among them, 12 common polymorphic sites were selected for genotyping in the beef cattle (<it>n </it>= 421). Statistical analysis revealed that a polymorphism in the 3'UTR (<it>c.2151*479C>T</it>) showed significant association with MS (<it>P</it><sup><it>cor</it>. </sup>= 0.02).</p> <p>Conclusion</p> <p>Our findings suggest that polymorphisms in <it>CAPN1 </it>might be one of the important genetic factors involved in carcass quality in beef cattle, although it could be false positive association.</p

    Complications of Teeth Affected by Molar-Incisor Malformation and Pathogenesis According to Microbiome Analysis

    A molar-incisor malformation (MIM) is a recently reported dental anomaly that causes premature loss of the first molar with severe dentoalveolar infection. However, there has been no research on the pathogenesis yet. The aim of this study was to report the clinical process of MIMs and investigate the pathogenesis by conducting a microbiome analysis. An eight-year-old girl was diagnosed with MIM and after two years, four permanent first molars were sequentially extracted due to severe dentoalveolar infection. We recorded the patient`s clinical progress and collected oral microbiome samples from the extracted teeth with MIM and sound teeth as controls. The sites of microbiome sampling were represented by five habitats in two groups. Group (1) was the perio group: ① supragingival plaque, ② subgingival plaque, and ③ a pical abscess; and group (2) was the endo group: ④ coronal pulp chamber and ⑤ root canal. The perio group was composed predominantly of genera Streptococcus, Veilonella, and Leptotrichia. Spirochetes appeared in one sample from a severe periodontal abscess. Aggregatibacter actinomyces were not identified. In the endo groups, pulp necrosis was observed in all MIM and the genera Peptostreptococcus and Parvimonas predominated. In conclusion, MIM teeth caused localized tooth-related periodontitis with pulp necrosis rather than localized juvenile periodontitis, resulting in a poor prognosis, and timely extraction is highly recommended
