Analysis of benzo[c] phenanthridine alkaloids in Eschscholtzia californica cell culture using HPLC-DAD and HPLC-ESI-MS/MS
Effective HPLC-DAD and HPLC-ESI-MS/MS methods have been developed for the analysis of eight benzo[c] phenanthridine alkaloids (sanguinarine, chelirubine, macarpine, chelerythrine, dihydrosanguinarine, dihydrochelirubine, dihydromacarpine and dihydrochelerythrine), which are important metabolites in Eschscholtzia californica cell culture. By adopting a ternary gradient pump system, the dihydro-form alkaloids hardly separable from each other could be successfully separated, and all the target alkaloids could be simultaneously quantified with the LOD values of 0.01-0.79 mu g/mL and the LOQ values of 0.03-3.59 mu g/mL. This HPLC-DAD method was further confirmed by HPLC-ESI-MS/MS system in multiple reaction monitoring mode. Each separated HPLC peak was identified as the target alkaloid, showing its relevant ionized molecule and selected fragment ion. By applying the established method, alkaloid production during the E. californica cell culture could be successfully monitored and some valuable information on its metabolism could be deduced.11Ysciescopu

Lactational coumestrol exposure increases ovarian apoptosis in adult rats
This study is the first to examine the increased apoptosis in the adult rat ovary after lactational exposure to coumestrol (COU), a potent phytoestrogen. Lactating dams were gavaged at doses of 0.01, 0.1, 1, and 10 mg/kg COU during the lactation period and the reproductive effects of female pups were investigated in young adults. Rats were sacrificed at postnatal days (PND) 81–84. Ovarian weights were reduced significantly at 0.1 and 1.0 mg/kg COU. The reduction in the ovarian weight occurred in parallel with an increase in the apoptosis at PND 135–140. A marked dose-dependent increase in the expressions of active caspase-3 and -7 was observed in ovarian granulosa cells. Immunostaining for active caspase-3 and the TUNEL staining of apoptotic cells were also increased in ovaries exposed to COU in a dose-dependent manner. Discutindo a educação ambiental no cotidiano escolar: desenvolvimento de projetos na escola formação inicial e continuada de professores
A presente pesquisa buscou discutir como a Educação Ambiental (EA) vem sendo trabalhada, no Ensino Fundamental e como os docentes desta escola compreendem e vem inserindo a EA no cotidiano escolar., em uma escola estadual do município de Tangará da Serra/MT, Brasil. Para tanto, realizou-se entrevistas com os professores que fazem parte de um projeto interdisciplinar de EA na escola pesquisada. Verificou-se que o projeto da escola não vem conseguindo alcançar os objetivos propostos por: desconhecimento do mesmo, pelos professores; formação deficiente dos professores, não entendimento da EA como processo de ensino-aprendizagem, falta de recursos didáticos, planejamento inadequado das atividades. A partir dessa constatação, procurou-se debater a impossibilidade de tratar do tema fora do trabalho interdisciplinar, bem como, e principalmente, a importância de um estudo mais aprofundado de EA, vinculando teoria e prática, tanto na formação docente, como em projetos escolares, a fim de fugir do tradicional vínculo “EA e ecologia, lixo e horta”.Facultad de Humanidades y Ciencias de la Educació
Determination of sodium bis(2-ethylhexyl)sulfosuccinate (AOT) surfactant with liquid chromatography: Comparative study of evaporative light scattering detector, ultraviolet detector and conductivity detector
This work presents comparison of performance of ultraviolet (UV) detector, conductivity detector (CD) and evaporative light scattering detector (ELSD) in terms of quantitative analysis of AOT (sodium bis(2-ethylhexyl)sulfosuccinate) using liquid chromatography. The employed chromatographic condition, including an acetonitrile/water (45:55, v/v) isocratic eluent system, is suitable for the three different detectors, and the figures of merits obtained by building up calibration plots are compared. The sensitivities of the detectors are in the order of ELSD ≈ CD >> UV detector. The linear range for quantification of AOT depends on the type of detector: the lower limits are in the order of UV detector (207 ㎍ mL-1) < CD (310 ㎍ mL-1) << ELSD (930 ㎍ mL-1), while the upper limits are 3720 ㎍ mL-1 for all the detectors (the maximum concentration of injected standard solution). The detection limits are 155 ㎍ mL-1 for ELSD, 78 ㎍ mL-1 for UV detector and 13 ㎍ mL-1 for CD, respectively. The figures of merit for each detector could be a guideline in choosing a detector in quantization of AOT. Furthermore, application of the chromatographic method to two commercial products is demonstrated
Bioethanol production from mannitol by a newly isolated bacterium, Enterobacter sp. JMP3
In this study a new bacterium capable of growing on brown seaweed Laminaria japonica, Enterobacter sp. JMP3 was isolated from the gut of turban shell, Batillus cornutus. In anaerobic condition, it produced high yields of ethanol (1.15 mol-EtOH mol-mannitol(-1)) as well as organic acids from mannitol, the major carbohydrate component of L. japonica. Based on carbon distribution and metabolic flux analysis, it was revealed that mannitol was more favorable than glucose for ethanol production due to their different redox states. This indicates that L japonica is one of the promising feedstock for bioethanol production. Additionally, the mannitol dehydrogenation pathway in Enterobacter sp. JMP3 was examined and verified. Finally, an attempt was made to explore the possibility of controlling ethanol production by altering the redox potential via addition of external NADH in mannitol fermentation.
