Insects Inhabiting the Burrows of the Ozark Pocket Gopher in Arkansas

Pocket gopher burrows provide a stable environment for a variety of inquilines; hence this mammal is regarded as a keystone species. Most of the arthropods inhabiting pocket gopher burrows are restricted to this microhabitat. As part of a planned state-wide biotic survey of insects inhabiting this unusual microhabitat, we have focused our initial sampling efforts on the Ozark pocket gopher (Geomys bursarius ozarkensis; Geomyidae). In 2004 and 2005, pitfall traps were established in pocket gopher burrows and in 2007 and 2008, nests and associated chambers were excavated. Retrieved samples contained scarab, histerid, and rove beetles, cave crickets and anthomyiid flies. The histerids consisted of five species, one of which is undescribed, and all of which are new state records. A total of five species of scarab beetles were collected and all of these represent new state records. Two species of cave crickets were collected; one appears to be undescribed, and the other is essentially a Great Plains species and represents a considerable range extension. AbstractTissue resident macrophages have vital homeostatic roles in many tissues but their roles are less well defined in the heart. The present study aimed to identify the density, polarisation status and distribution of macrophages in the healthy murine heart and to investigate their ability to respond to immune challenge. Histological analysis of hearts from CSF-1 receptor (csf1-GFP; MacGreen) and CX3CR1 (Cx3cr1GFP/+) reporter mice revealed a sparse population of GFP positive macrophages that were evenly distributed throughout the left and right ventricular free walls and septum. F4/80+CD11b+ cardiac macrophages, sorted from myocardial homogenates, were able to phagocytose fluorescent beads in vitro and expressed markers typical of both ‘M1’ (IL-1β, TNF and CCR2) and ‘M2’ activation (Ym1, Arg 1, RELMα and IL-10), suggesting no specific polarisation in healthy myocardium. Exposure to Th2 challenge by infection of mice with helminth parasites Schistosoma mansoni, or Heligmosomoides polygyrus, resulted in an increase in cardiac macrophage density, adoption of a stellate morphology and increased expression of Ym1, RELMα and CD206 (mannose receptor), indicative of ‘M2’ polarisation. This was dependent on recruitment of Ly6ChighCCR2+ monocytes and was accompanied by an increase in collagen content.In conclusion, in the healthy heart resident macrophages are relatively sparse and have a phagocytic role. Following Th2 challenge this population expands due to monocyte recruitment and adopts an ‘M2’ phenotype associated with increased tissue fibrosis

Comparative analysis of antibodies to SARS-CoV-2 between asymptomatic and convalescent patients

Summary: The SARS-CoV-2 viral pandemic has induced a global health crisis, which requires more in-depth investigation into immunological responses to develop effective treatments and vaccines. To understand protective immunity against COVID-19, we screened over 60,000 asymptomatic individuals in the Southeastern United States for IgG antibody positivity against the viral Spike protein, and approximately 3% were positive. Of these 3%, individuals with the highest anti-S or anti-RBD IgG level showed a strong correlation with inhibition of ACE2 binding and cross-reactivity against non-SARS-CoV-2 coronavirus S-proteins. We also analyzed samples from 94 SARS-CoV-2 patients and compared them with those of asymptomatic individuals. SARS-CoV-2 symptomatic patients had decreased antibody responses, ACE2 binding inhibition, and antibody cross-reactivity. Our study shows that healthy individuals can mount robust immune responses against SARS-CoV-2 without symptoms. Furthermore, IgG antibody responses against S and RBD may correlate with high inhibition of ACE2 binding in individuals tested for SARS-CoV-2 infection or post vaccination
