Snapshot 3D tracking of insulin granules in live cells
Rapid and accurate volumetric imaging remains a challenge, yet has the
potential to enhance understanding of cell function. We developed and used a
multifocal microscope (MFM) for 3D snapshot imaging to allow 3D tracking of
insulin granules labeled with mCherry in MIN6 cells. MFM employs a special
diffractive optical element (DOE) to simultaneously image multiple focal
planes. This simultaneous acquisition of information determines the 3D location
of single objects at a speed only limited by the frame rate of array detector .
We validated the accuracy of MFM imaging and tracking with fluorescence beads;
the 3D positions and trajectories of single fluorescence beads can be
determined accurately over a wide range of spatial and temporal scales. The 3D
positions and trajectories of single insulin granules in a 3.2 micro meter deep
volume were determined with imaging processing that combines 3D decovolution,
shift correction, and finally tracking using the Imaris software package. We
find that the motion of the granules is super-diffusive, but less so in 3D than
2D for cells grown on coverslip surfaces, suggesting an anisotropy in the
cytoskeleton (e.g. microtubules and action)
Developmental course of autistic social impairment in males
BACKGROUND: Recent research has suggested that autistic social impairment (ASI) is continuously distributed in nature, and that subtle autistic-like social impairments aggregate in the family members of children with pervasive developmental disorders (PDDs). This study examined the longitudinal course of quantitatively-characterized ASI in 3 to 18 year old boys with and without PDD. METHODS: We obtained assessments of 95 epidemiologically ascertained male-male twin pairs and a clinical sample of 95 affected children using the Social Responsiveness Scale (SRS), at two time points, spaced 1–5 years apart. Longitudinal course was examined as a function of age, familial loading for PDD, and autistic severity at baseline. RESULTS: Inter-individual variation in SRS scores was highly preserved over time, with test-retest correlation of 0.90 for the entire sample. SRS scores exhibited modest general improvement over the study period; individual trajectories varied as a function of severity at baseline and were highly familial. CONCLUSION: Quantitative measurements of ASI reflect heritable trait-like characteristics. Such measurements can serve as reliable indices of phenotypic severity for genetic and neurobiologic studies, and have potential utility for ascertaining incremental response to intervention
(Between the Streets) In Worcester : Redefining Professional Education in Community Development to Cultivate Empathy Through a Community Theatrical Framework
This research paper presents an alternative form of conducing Community Development Research. It highlights the gaps that currently exist in professional education with the community development and planning program at Clark University. The research paper employs a theatrical framework to encourage practitioners to ask more illuminating questions that informs the ‘human work’ that sometimes gets overlooked. In order to be authentic in the field of Community Development, practitioners need to be in touch with a less scientifically rational side of themselves, to truly embrace the complexities of the human condition. Drawing from my personal experiences, I wrote a play based on my field observations, conversations and interactions that I’ve had during my time in the program. This will hopefully provide both students and practitioners an alternative way to see their world and their encounters with people around them. By engaging with a theatrical framework, it is my hope that both students and practitioners also discover new ways of conducing research, and new ways of Being, both inside and outside the classroom
ABCDs of Triggerfish in Pemba and Misali Island: Abundance, Biodiversity, Behaviour, Cultural Significance and Distribution
This study takes a socioecological approach to understanding triggerfish (Balistidae) in Pemba Island, using Misali Island reefs as a sample ecological site. Two reef sites on Misali Island and three of four districts in Pemba were observed during the study. The abundance of Balistipus undulatus at one of two study sites is close to the maximum carrying capacity of the reef. Overall observed biodiversity was low. Behavioural trends point to distinct species specific defence strategies and species aggression taking different forms. Triggerfishes cultural significance was primarily as a food fish, though it is not highly sought after. With polarised responses for both catch and as a food, the fishery pressure on them was assessed to be moderate. Balistidae abundance, especially B. undulatus could be a good reef health and fishery monitoring metric. This study samples triggerfish ecology, finding interactions and samples which produce a holistic idea of triggerfish in Pemba

Abstract Primary ciliary dyskinesia (PCD) is a chronic suppurative airways disease that is usually recessively inherited and has marked clinical phenotypic heterogeneity. Classic symptoms include neonatal respiratory distress, chronic rhinitis since early childhood, chronic otitis media, recurrent airway infections leading to bronchiectasis, chronic sinusitis, laterality defects with and without congenital heart disease including abnormal situs in approximately 50% of the cases, and male infertility. Lung function deteriorates progressively from childhood throughout life. ‘Better Experimental Approaches to Treat Primary Ciliary Dyskinesia’ (BEAT-PCD) is a network of scientists and clinicians coordinating research from basic science through to clinical care with the intention of developing treatments and diagnostics that lead to improved long-term outcomes for patients. BEAT-PCD activities are supported by EU funded COST Action (BM1407). The third BEAT-PCD conference and fourth PCD training school were held jointly in February 2018 in Lisbon, Portugal. Presentations and workshops focussed on advancing the knowledge and skills relating to PCD in: basic science, epidemiology, diagnostic testing, clinical management and clinical trials. The multidisciplinary conference provided an interactive platform for exchanging ideas through a program of lectures, poster presentations, breakout sessions and workshops. Three working groups met to plan consensus statements. Progress with BEAT-PCD projects was shared and new collaborations were fostered. In this report, we summarize the meeting, highlighting developments made during the meeting
Pan-Cancer Analysis of lncRNA Regulation Supports Their Targeting of Cancer Genes in Each Tumor Context
Long noncoding RNAs (lncRNAs) are commonly dys-regulated in tumors, but only a handful are known toplay pathophysiological roles in cancer. We inferredlncRNAs that dysregulate cancer pathways, onco-genes, and tumor suppressors (cancer genes) bymodeling their effects on the activity of transcriptionfactors, RNA-binding proteins, and microRNAs in5,185 TCGA tumors and 1,019 ENCODE assays.Our predictions included hundreds of candidateonco- and tumor-suppressor lncRNAs (cancerlncRNAs) whose somatic alterations account for thedysregulation of dozens of cancer genes and path-ways in each of 14 tumor contexts. To demonstrateproof of concept, we showed that perturbations tar-geting OIP5-AS1 (an inferred tumor suppressor) andTUG1 and WT1-AS (inferred onco-lncRNAs) dysre-gulated cancer genes and altered proliferation ofbreast and gynecologic cancer cells. Our analysis in-dicates that, although most lncRNAs are dysregu-lated in a tumor-specific manner, some, includingOIP5-AS1, TUG1, NEAT1, MEG3, and TSIX, synergis-tically dysregulate cancer pathways in multiple tumorcontexts
