    Genome-wide investigation and expression analysis of OSCA gene family in response to abiotic stress in alfalfa

    Alfalfa is an excellent leguminous forage crop that is widely cultivated worldwide, but its yield and quality are often affected by drought and soil salinization. Hyperosmolality-gated calcium-permeable channel (OSCA) proteins are hyperosmotic calcium ion (Ca2+) receptors that play an essential role in regulating plant growth, development, and abiotic stress responses. However, no systematic analysis of the OSCA gene family has been conducted in alfalfa. In this study, a total of 14 OSCA genes were identified from the alfalfa genome and classified into three groups based on their sequence composition and phylogenetic relationships. Gene structure, conserved motifs and functional domain prediction showed that all MsOSCA genes had the same functional domain DUF221. Cis-acting element analysis showed that MsOSCA genes had many cis-regulatory elements in response to abiotic or biotic stresses and hormones. Tissue expression pattern analysis demonstrated that the MsOSCA genes had tissue-specific expression; for example, MsOSCA12 was only expressed in roots and leaves but not in stem and petiole tissues. Furthermore, RT–qPCR results indicated that the expression of MsOSCA genes was induced by abiotic stress (drought and salt) and hormones (JA, SA, and ABA). In particular, the expression levels of MsOSCA3, MsOSCA5, MsOSCA12 and MsOSCA13 were significantly increased under drought and salt stress, and MsOSCA7, MsOSCA10, MsOSCA12 and MsOSCA13 genes exhibited significant upregulation under plant hormone treatments, indicating that these genes play a positive role in drought, salt and hormone responses. Subcellular localization results showed that the MsOSCA3 protein was localized on the plasma membrane. This study provides a basis for understanding the biological information and further functional analysis of the MsOSCA gene family and provides candidate genes for stress resistance breeding in alfalfa

    Sirt1 Deletion Leads to Enhanced Inflammation and Aggravates Endotoxin-Induced Acute Kidney Injury

    Bacterial endotoxin has been known to induce excessive inflammatory responses and acute kidney injury. In the present study, we used a mouse model of endotoxemia to investigate the role of Sirt1 in inflammatory kidney injury. We examined molecular and cellular responses in inducible Sirt1 knockout (Sirt1-/-) mice and wild type littermates (Sirt1+/+) in lipopolysaccharide (LPS)-induced kidney injury. Our studies demonstrated that Sirt1 deletion caused aggravated kidney injury, which was associated with increased inflammatory responses including elevated pro-inflammatory cytokine production, and increased ICAM-1 and VCAM-1 expression. Inflammatory signaling such as STAT3/ERK phosphorylation and NF-κB activation was markedly elevated in kidney tissues of Sirt1 knockout mice after LPS challenge. The results indicate that Sirt1 is protective against LPS-induced acute kidney injury by suppressing kidney inflammation and down-regulating inflammatory signaling

    Equilibrium between Road Traffic Congestion and Low-Carbon Economy: A Case Study from Beijing, China

    China has allocated low-carbon targets into all regions and trades, and road traffic also has its own emission reduction targets. Congestion may increase carbon emissions from road traffic. It is worthwhile to study whether it is possible to achieve the goal of road traffic reduction by controlling congestion that is, to achieve the equilibrium between traffic congestion and a low-carbon economy. The innovation of this paper is mainly reflected in the innovative topic selection, the introduction of a traffic index, and the establishment of the first traffic congestion and low-carbon economic equilibrium model. First, the relevant calculation method of the traffic index is introduced, and the traffic index is used to quantify the traffic congestion degree. Using the traffic index, GDP, and road passenger traffic volume, a nonlinear regression model of road traffic carbon emissions is constructed. Then, the calculation method of the carbon emission intensity of road traffic in the region is proposed. The equilibrium model of traffic congestion and a low-carbon economy is constructed to look for the degree of road traffic congestion that may occur under the permitted carbon emission intensity. Taking Beijing, where electric vehicles account for less than 3% of the total vehicles, as an example, it is difficult to achieve the equilibrium target between road traffic congestion and a low-carbon economy by alleviating traffic congestion in 2020. If the target of traffic carbon emission reduction in 2020 is adjusted from 40%&ndash 45% to 19.7% based on 2005, the equilibrium will be achieved. A negative correlation between road traffic carbon emissions and the reciprocal of the traffic index (1/TI) is found after eliminating the effects of GDP and PTV (road passenger traffic volume). As the traffic index decreases by units, the carbon emission reduction accelerates. The results show that carbon reduction targets cannot be simply allocated to various industries. The results of the research on the degree of the impact of traffic congestion on carbon emissions can be used as a basis for carbon reduction decisions of the traffic sector. The research method of this paper can provide a reference for the study of the equilibrium of traffic congestion and a low-carbon economy in other regions. Document type: Articl

    A functional anatomical shift from the lateral frontal pole to dorsolateral prefrontal cortex in emotion action control underpins elevated levels of anxiety: partial replication and generalization of Bramson et al., 2023

    Background: Emotion control represents a promising intervention target for mental disorders. In a recent study Bramson et al. (2023) demonstrate a functional-anatomical shift from the lateral frontal pole (FPl) to the dorsolateral prefrontal cortex (DLPFC) in anxious individuals during emotional action control. However, findings of neuroimaging experiments are often limited regarding generalizability and reproducibility. The present study examined the robustness of the reported functional shift across samples, cultures and paradigms. Methods: We capitalized on large-scale task fMRI data (n = 250 participants) using an affective linguistic Go/NoGo paradigm to examine the anxiety-related shift between FPl and DLPFC during emotional action control. Additionally, context-dependent functional connectivity analyses were employed to examine anxiety-related differences and associations on the network level. Results: Non-anxious individuals engaged the left FPl while highly anxious individuals specifically recruited the DLPFC, but non-significant between-group differences were found (see also Bramson et al.). The secondary analyses revealed moderate evidence for the absence of left FPl activation in the high-anxious as well as for left DLPFC activation in the non-anxious group. Additionally, trait anxiety scores were positively correlated with left DLPFC activity but negatively correlated with left FPl activity across groups. Furthermore, we found a context-specific connectivity shift between the subgenual anterior cingulate cortex (sgACC) with the FPl and DLPFC specifically in highly anxious individuals. Conclusion: The results partially confirmed the anxiety-related shift as reported by Bramson and colleagues across paradigms and samples. The findings provide further support for the functional shift in anxiety and can inform target-based interventions of persistent emotional control deficits in anxiety disorders.published_or_final_versio

    Opposing associations of Internet Use Disorder symptom domains with structural and functional organization of the striatum: A dimensional neuroimaging approach

    Background: Accumulating evidence suggests brain structural and functional alterations in Internet Use Disorder (IUD). However, conclusions are strongly limited due to the retrospective case-control design of the studies, small samples, and the focus on general rather than symptom-specific approaches. Methods: We here employed a dimensional multi-methodical MRI-neuroimaging design in a final sample of n 5 203 subjects to examine associations between levels of IUD and its symptom-dimensions (loss of control/time management, craving/social problems) with brain structure, resting state and taskbased (pain empathy, affective go/no-go) brain function. Results: Although the present sample covered the entire range of IUD, including normal, problematic as well as pathological levels, general IUD symptom load was not associated with brain structural or functional alterations. However, the symptom-dimensions exhibited opposing associations with the intrinsic and structural organization of the brain, such that loss of control/time management exhibited negative associations with intrinsic striatal networks and hippocampal volume, while craving/social problems exhibited a positive association with intrinsic striatal networks and caudate volume. Conclusions: Our findings provided the first evidence for IUD symptom-domain specific associations with progressive alterations in the intrinsic structural and functional organization of the brain, particularly of striatal systems involved in reward, habitual and cognitive control processes

    Soil microbial networks’ complexity as a primary driver of multifunctionality in photovoltaic power plants in the northwest region of China

    IntroductionExploiting photovoltaic power generation as a novel source of clean energy has become increasingly common in recent times. Nevertheless, the impact of photovoltaic power plants (PVs) on soil microbial activity and several functions is unclear.MethodsThe present investigation aims to collect soil samples from photovoltaic power plants in arid and semi-arid regions with different years of construction, determine the physicochemical properties of the soil, and employ high-throughput sequencing to obtain 16S rRNA and ITS genes from the PV. This approach examines the community composition of bacteria and fungi in plant soils. This dataset is adopted to explore the role of soil physicochemical characteristics and climatic factors in the variousness and complexness of the network of soil microbial communities in PVs.ResultsThe findings reveal that soil physicochemical properties exhibit a gradual increase over time, with bacterial and fungal diversity showing a corresponding gradual increase and reaching a maximum over a period of 5–10 years. Furthermore, it is observed that the topological properties of the microbial network underwent significant changes driven by microbial diversity. Bacterial and fungal diversity as well as network complexity also display positive and negative correlations, respectively. A positive and significant correlation is detected between the bacterial network complexity and the soil multifunctionality, whereas a substantial negative correlation is observed between the fungal network complexity and the soil multifunctionality.DiscussionIn conclusion the environment is able to directly regulate soil microbial diversity, thereby affecting network complexity and driving soil multifunctionality. Such discoveries are aimed to have crucial ecological implications for predicting environmental-soil-microbial effects on soil multifunctionality in photovoltaic zones

    Regulation of High-Temperature Stress Response by Small RNAs

    Temperature extremes constitute one of the most common environmental stresses that adversely affect the growth and development of plants. Transcriptional regulation of temperature stress responses, particularly involving protein-coding gene networks, has been intensively studied in recent years. High-throughput sequencing technologies enabled the detection of a great number of small RNAs that have been found to change during and following temperature stress. The precise molecular action of some of these has been elucidated in detail. In the present chapter, we summarize the current understanding of small RNA-mediated modulation of high- temperature stress-regulatory pathways including basal stress responses, acclimation, and thermo-memory. We gather evidence that suggests that small RNA network changes, involving multiple upregulated and downregulated small RNAs, balance the trade-off between growth/development and stress responses, in order to ensure successful adaptation. We highlight specific characteristics of small RNA-based tem- perature stress regulation in crop plants. Finally, we explore the perspectives of the use of small RNAs in breeding to improve stress tolerance, which may be relevant for agriculture in the near future

    Atrasentan and renal events in patients with type 2 diabetes and chronic kidney disease (SONAR): a double-blind, randomised, placebo-controlled trial

    Background: Short-term treatment for people with type 2 diabetes using a low dose of the selective endothelin A receptor antagonist atrasentan reduces albuminuria without causing significant sodium retention. We report the long-term effects of treatment with atrasentan on major renal outcomes. Methods: We did this double-blind, randomised, placebo-controlled trial at 689 sites in 41 countries. We enrolled adults aged 18–85 years with type 2 diabetes, estimated glomerular filtration rate (eGFR)25–75 mL/min per 1·73 m 2 of body surface area, and a urine albumin-to-creatinine ratio (UACR)of 300–5000 mg/g who had received maximum labelled or tolerated renin–angiotensin system inhibition for at least 4 weeks. Participants were given atrasentan 0·75 mg orally daily during an enrichment period before random group assignment. Those with a UACR decrease of at least 30% with no substantial fluid retention during the enrichment period (responders)were included in the double-blind treatment period. Responders were randomly assigned to receive either atrasentan 0·75 mg orally daily or placebo. All patients and investigators were masked to treatment assignment. The primary endpoint was a composite of doubling of serum creatinine (sustained for ≥30 days)or end-stage kidney disease (eGFR <15 mL/min per 1·73 m 2 sustained for ≥90 days, chronic dialysis for ≥90 days, kidney transplantation, or death from kidney failure)in the intention-to-treat population of all responders. Safety was assessed in all patients who received at least one dose of their assigned study treatment. The study is registered with ClinicalTrials.gov, number NCT01858532. Findings: Between May 17, 2013, and July 13, 2017, 11 087 patients were screened; 5117 entered the enrichment period, and 4711 completed the enrichment period. Of these, 2648 patients were responders and were randomly assigned to the atrasentan group (n=1325)or placebo group (n=1323). Median follow-up was 2·2 years (IQR 1·4–2·9). 79 (6·0%)of 1325 patients in the atrasentan group and 105 (7·9%)of 1323 in the placebo group had a primary composite renal endpoint event (hazard ratio [HR]0·65 [95% CI 0·49–0·88]; p=0·0047). Fluid retention and anaemia adverse events, which have been previously attributed to endothelin receptor antagonists, were more frequent in the atrasentan group than in the placebo group. Hospital admission for heart failure occurred in 47 (3·5%)of 1325 patients in the atrasentan group and 34 (2·6%)of 1323 patients in the placebo group (HR 1·33 [95% CI 0·85–2·07]; p=0·208). 58 (4·4%)patients in the atrasentan group and 52 (3·9%)in the placebo group died (HR 1·09 [95% CI 0·75–1·59]; p=0·65). Interpretation: Atrasentan reduced the risk of renal events in patients with diabetes and chronic kidney disease who were selected to optimise efficacy and safety. These data support a potential role for selective endothelin receptor antagonists in protecting renal function in patients with type 2 diabetes at high risk of developing end-stage kidney disease.     Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    The main difficulty of credit risk evaluation is to evaluate borrowers' willingness of repayment, which is a subjective factor depending on the thoughts and ideas of borrowers. Text description is a kind of human behavior which reflects the mental process of writers. The authors identify the characteristics of borrowers from their text descriptions and further use them to evaluate the credit risk of loans. Experimental results show that: (1) textual information is a good choice when traditional financial information is missing. The authors can achieve similar accuracy using only textual information as traditional methods which use financial information and credit information from the third party. (2) Textual information is a good complementary information source to traditional financial information sources. Using textual information can improve the performance of credit risk evaluation system when combined with traditional financial information. </jats:p
