SMCR8 negatively regulates AKT and MTORC1 signaling to modulate lysosome biogenesis and tissue homeostasis
The intronic hexanucleotide expansion in the C9orf72 gene is one of the leading causes of frontotemporal lobar degeneration (FTLD) and amyotrophic lateral sclerosis (ALS), two devastating neurodegenerative diseases. C9orf72 forms a heterodimer with SMCR8 (Smith-Magenis syndrome chromosome region, candidate 8) protein. However, the physiological function of SMCR8 remains to be characterized. Here we report that ablation of SMCR8 in mice results in splenomegaly with autoimmune phenotypes similar to that of C9orf72 deficiency. Furthermore, SMCR8 loss leads to a drastic decrease of C9orf72 protein levels. Many proteins involved in the macroautophagy-lysosome pathways are downregulated upon SMCR8 loss due to elevated activation of MTORC1 and AKT, which also leads to increased spine density in the Smcr8 knockout neurons. In summary, our studies demonstrate a key role of SMCR8 in regulating MTORC1 and AKT signaling and tissue homeostasis. Křesťanské hodnoty v životě dospívající mládeže Diplomová práce Diplomová práce Křesťanské hodnoty v životě dospívající mládeže je koncipována jako pracovní materiál pro terapeuty, kněze a další pomáhající profese. Vycházím ze svých zkušeností poradenské praxe v oblasti osobnostního rozvoje a řešení konfliktních situací. Zaměřuji se na problematiku dospívajících a jejich potřeby zvládnout své osobní problémy, vyplývající z disharmonických rodinných vztahů a z různorodých negativních podnětů současné společnosti. Cílem práce je zviditelnit různorodost vlivů na utváření názorů dospívajících a současně nabídnout způsob, jak vytipovat a vybrat vhodné morální hodnoty křesťanského etosu, pro potřeby podpůrného rozhovoru s dospívajícím. V první kapitole vymezuji pojmy hodnota, morální princip a morální norma. Dále navrhuji základní kriteria, která musí být splněna, aby navrhované hodnoty byly přijaty. Téma doplňuji o pohledy hodnotové filosofie, křesťanské antropologie a morální teologie. V závěru kapitoly se zamýšlím nad morálním jednáním člověka ve vztahu ke svobodě a dobru. Ve druhé kapitole shrnuji poznatky současné psychologie o významu a funkci rodiny a jak ovlivňuje své jednotlivé členy. Následně uvádím, jaký význam rodině přikládá křesťanství. V třetí kapitole se zabývám vývojem..

The study aimed to explore the autoantibody profiles and the correlation of autoantibodies with the disease severity and the prognosis of the coronavirus disease 2019 (COVID-19) patients.MethodsThree hundred thirty-seven hospitalized COVID-19 patients from 6th to 23rd January 2023 were enrolled. Logistic and Cox regression analyses were used to analyze the risk factors for the patient’s disease severity and outcome. The association between Anti-extractable nuclear antigen antibody (ENA) positivity and the prognosis of COVID-19 patients was analyzed using Kaplan–Meier survival curves.Results137 of COVID-19 patients were detected positive for antinuclear antibody (ANA), 61 had positive results for ENA, and 38 were positive for ANA and ENA. ANA positivity rate was higher in non-severe illness group (p = 0.032). COVID-19 patients who died during hospitalization had a high rate of ENA positivity than convalescent patients (p = 0.002). Multivariate logistic regression showed that ANA positivity was a protective factor for the disease severity of COVID-19. Multivariate Cox regression analysis revealed that ENA positivity, white blood cells count (WBC), aspartate aminotransferase (AST), Creatinine (CREA), and CRP were independent risk factors for the outcome of COVID-19 patients, and that COVID-19 patients with ENA positivity had a lower cumulative survival rate (p = 0.002).ConclusionA spectrum of autoantibodies were expressed in COVID-19 patients, among which ANA and ENA positivity was associated with the severity and prognosis of COVID-19. Therefore, autoantibodies may help to assess the disease severity and prognosis of COVID-19 patients.</p
