    Bile Salt Resistance and Antimicrobial Activity of Lactic Acid Bacteria

    Cilj ovog zavr┼ínog rada bio je ispitati prisutnostfragmenatabsh gena, koji kodira za hidrolazu ┼żu─Źnih soli, kod bakterija mlije─Źne kiseline Lactobacillus helveticus M92, L. brevis D6, L. brevis ZG1, L. brevis SF9B, L. brevis SF15B i L. plantarum D13. Naime, bakterije aktivno┼í─çu enzima hidrolaze ┼żu─Źnih soli mogu utjecati na regulaciju koncentracije kolesterola, a ujedno taj enzim mo┼że biti odgovoran za pre┼żivljavanje probioti─Źkih sojeva u gastrointestinalnom traktu gdje izme─Ĺu ostalog, postoje nepovoljni uvjeti zbog ┼żu─Źnih soli. PCR reakcijom su fragmenti bsh gena potvr─Ĺeni kod sojeva Lactobacillus plantarum D13, L. brevis D6 i L. brevis SF9B. Probioti─Źke bakterije tako─Ĺer imaju va┼żnu ulogu u inhibiciji rasta potencijalno patogenih mikroorganizama, a to antimikrobno djelovanje je posljedica biosinteze raznih ekstracelularnih metabolita, me─Ĺu kojima su i bakteriocini. Stoga je u ovom radu dodatno ispitivana antimikrobna aktivnost soja Lactobacillus plantarum D13, metodom s dvostrukim slojem agara. Bakteriocinska aktivnost ispitivanog soja je inducirana kokultivacijom s test-mikroorganizmima L. monocytogenes ATCC 19111 i St. aureus 3048.Dodatkom ┼żu─Źnih soli, kao dodatnih stresnih okoli┼ínih ─Źimbenika za bakterije mlije─Źne kiseline, dodatno je potaknuto bakteriocinsko djelovanje prema test-mikroorganizmu St. aureus 3048.The aim of this study was to examine presence of bsh gene, encoding for bile salt hydrolase (BSH), in lactic acid bacteria strains Lactobacillus helveticus M92, L. brevis D6, L. brevis ZG1, L. brevis SF9B, L. brevis SF15B andL. plantarum D13. Probiotic bacteria can be involved in regulation of cholesterol level by BSH activity. At the same time, BSH actvity could be responsible for survival of probiotic strains in stressful gastrointestinal tract conditions, caused, amoung other, by bile salts. According to results of PCR reactions, bsh gene fragments were detected in Lactobacillus plantarum D13, L. brevis D6 and L. brevis SF9B strains. In addition, lactic acid bacteria have an important role in inhibition of growth of potentially pathogenic microorganisms. That antimicrobial activity is result of biosynthesis of various extracellular compounds,including bacteriocins. Hence, antimicrobial activity of Lactobacillus plantarum D13 strain was also examined by agar-spot-agar method. Bacteriocin activity was induced by cocultivation wtih test-microorganisms L. monocytogenes ATCC 19111 andSt. aureus 3048. Addition of bile salts, as additional stressful environmental condition, has furhter induced bacteriocin activity towards test-microorganism St. aureus 3048

    A method for real-time detection of human fall from video

    In this paper we present a method for real-time detection of human fall from video for support of elderly people living alone in their homes. The detection algorithm has four steps: background estimation, extraction of moving objects, motion feature extraction, and fall detection. The detection is based on features that quantify dynamics of human motion and body orientation. The algorithms are implemented in C++ using the OpenCV library. The method is tested using a single camera and 20 test video recordings showing typical fall scenarios and regular household behaviour. The experimental results show 90% of human fall detection accuracy

BLE koriste iBeaconi za slanje identifikatora koji kompatibilna aplikacija mo┼że pretvoriti u lokaciju ili pokrenuti odre─Ĺenu akciju. Spree je e-commerce okvir otvorenog koda koje pru┼ża osnovnu funkcionalnost Internet trgovine i podlogu za jednostavno nadogra─Ĺivanje. U radu je opisan razvoj hibridne mobilne aplikacije za kupnju posredstvom Interneta. Za izradu aplikacije kori┼íteni su programski okviri Apache Cordova i Ionic koji se zasniva na AngularJS-u. Implementirana aplikacija pru┼ża funkcionalnost pregledavanja proizvoda Internet trgovine, stvaranja narud┼żbe te odre─Ĺivanje lokacije najbli┼żeg iBeacon ure─Ĺaja i dohva─çanja pripadaju─çe kategorije proizvoda.Bluetooth low energy is a network technology which compared to Classic Bluetooth provides reduced power consumption and cost while maintaining a similar communication range. BLE is used by iBeacon to transmit identifier picked up by compatible app that can be turned into a physical location or trigger an action on the device. Spree is an open source e-commerce solution that offers basic webshop features and it is easy to add any new funcionality. This thesis describes development of a hybrid smartphone application for online shopping. Frameworks used for application development are Apache Cordova and Ionic which is based on AngularJS. Application can be used for assortment browsing, making an order, determining the closest iBeacon and displaying appropriate product category

    Naj─Źe┼í─çi proizvodi industrijske konoplje (Cannabis sativa L.) su bra┼íno i ulje koje se proizvodi hladnim pre┼íanjem sjemenki. Prilikom tog postupka kao nusproizvod zaostaje uljna poga─Źa konoplje koja je bogata proteinima. Enzimskom hidrolizom proteina konoplje dobivaju se peptidi iznimnog biolo┼íkog potencijala te se zato koriste u proizvodnji hrane i farmaceutika. Zato je cilj ovog rada bio ispitati antiproliferacijsko djelovanje proteinskih hidrolizata uljne poga─Źe konoplje pripremljenih s tri razli─Źita enzima (alkalaza, neutraza i protamex) in vitro na tumorskoj HeLa i netumorskoj HaCaT stani─Źnoj liniji, protektivni u─Źinak na HaCaT stanicama u kojima je induciran oksidativni stres te induciranje stani─Źne smrti u HeLa stanicama. Dobiveni rezultati pokazuju kako svi pripremljeni hidrolizati i izolat posjeduju antiproliferacijsku aktivnost. HeLa stanice su osjetljivije na u─Źinak hidrolizata te je najja─Źi antiproliferativni u─Źinak imao hidrolizat dobiven pomo─çu enzima alkalaze. Dokazan je i antioksidativni u─Źinak hidrolizata pripravljenog enzimom neutrazom na HaCaT stanice u kojima je induciran oksidativni stres. Tako─Ĺer, dokazano je i kako hidrolizat dobiven pomo─çu enzima alkalaze inducira stani─Źnu smrt u HeLa stanicama.The most common products of industrial hempseed (Cannabis sativa L.) are flour and oil, which is produced by cold pressing of seeds. Byproduct of this process is hempseed oilcake that is rich in protein. Product of enzymatic hydrolysis of hemp proteins are peptides with biological potential and therefore are used in the production of food and pharmaceuticals. In this thesis the antiproliferative effects of hempseed protein isolate and hydrolysates prepared using three proteases (Alcalase, Neutrase and Protamex) were tested in vitro on cancer (HeLa) and normal (HaCaT) cell lines. Protective effects were evaluated on H2O2-induced oxidative stress in HaCaT cells. In addition, it was tested whether hempseed protein hydrolysate induces cell death in HeLa cells. The obtained results showed that hempseed protein isolate and hydrolysates exhibit antiproliferative effects. The strongest antiproliferative effect was observed in hydrolysates obtained by Alcalase. The study has confirmed the protective effects of hydrolysate obtained by Neutrase during induced oxidative stress in HaCaT cell line. Furthermore, it is confirmed that hydrolysate obtained by Alcalase induces cell death in HeLa cell line
