    Cloning, expression, and localization of a rat brain high-affinity glycine transporter

    A cDNA clone encoding a glycine transporter has been isolated from rat brain by a combined PCR and plaque-hybridization strategy. mRNA synthesized from this clone (designated GLYT1) directs the expression of sodium-and chloride-dependent, high-affinity uptake of [3H]glycine by Xenopus oocytes. [3H]Glycine transport mediated by clone GLYT1 is blocked by sarcosine but is not blocked by methylaminoisobutyric acid or L-alanine, a substrate specificity similar to that described for a previously identified glycine-uptake system called system Gly. In situ hybridization reveals that GLYT1 is prominently expressed in the cervical spinal cord and brainstem, two regions of the central nervous system where glycine is a putative neurotransmitter. GLYT1 is also strongly expressed in the cerebellum and olfactory bulb and is expressed at lower levels in other brain regions. The open reading frame of the GLYT1 cDNA predicts a protein containing 633 amino acids with a molecular mass of ≈70 kDa. The primary structure and hydropathicity profile of GLYT1 protein reveal that this protein is a member of the sodium- and chloride-dependent superfamily of transporters that utilize neurotransmitters and related substances as substrates

    Extractability and mobility of mercury from agricultural soils surrounding industrial and mining contaminated areas

    This study focussed on a comparison of the extractability of mercury in soils with two different contamination sources (a chlor-alkali plant and mining activities) and on the evaluation of the influence of specific soil properties on the behaviour of the contaminant. The method applied here did not target the identification of individual species, but instead provided information concerning the mobility of mercury species in soil. Mercury fractions were classified as mobile, semi-mobile and non-mobile. The fractionation study revealed that in all samples mercury was mainly present in the semi-mobile phase (between 63 and 97%). The highest mercury mobility (2.7 mg kg-1) was found in soils from the industrial area. Mining soils exhibited higher percentage of non-mobile mercury, up to 35%, due to their elevated sulfur content. Results of factor analysis indicate that the presence of mercury in the mobile phase could be related to manganese and aluminum soil contents. A positive relation between mercury in the semi-mobile fraction and the aluminium content was also observed. By contrary, organic matter and sulfur contents contributed to mercury retention in the soil matrix reducing the mobility of the metal. Despite known limitations of sequential extraction procedures, the methodology applied in this study for the fractionation of mercury in contaminated soil samples provided relevant information on mercury's relative mobility

    The Effect of Olanzapine on Craving and Alcohol Consumption

    Previous studies have indicated that olanzapine decreases craving after a priming dose of alcohol, that craving after a priming dose of alcohol is greater among individuals with the seven-repeat allele of the DRD4 variable number of tandem repeats (VNTR) polymorphism, and that the effect of olanzapine (a D2/D4 antagonist) is more pronounced among individuals with this allele. The present study tested the hypothesis that olanzapine may be differentially effective at reducing cue-elicited craving and differentially effective as a treatment for alcohol dependence over the course of a 12-week, randomized, placebo-controlled trial among individuals with and without the seven-repeat allele. Participants who met DSM IV criteria for alcohol dependence were randomly assigned to receive olanzapine (5 mg) or a placebo over the course of the trial. After 2 weeks of treatment, participants completed a cue reactivity assessment. The results suggested that participants who were homozygous or heterozygous for the seven (or longer)-repeat allele of the DRD4 VNTR responded to olanzapine with reductions in cue-elicited craving as well as reductions in alcohol consumption over the course of the 12-week trial, whereas individuals with the shorter alleles did not respond favorably to olanzapine

    A novel two-step sequential bioaccessibility test for potentially toxic elements in inhaled particulate matter transported into the gastrointestinal tract by mucociliary clearance

    A novel two-step sequential extraction has been developed to assess the bioaccessibility of As, Cd, Cr, Cu, Fe, Mn, Ni, Pb, and Zn in airborne particulate matter following inhalation and transport into the human gastrointestinal tract by mucociliary clearance. A new artificial mucus fluid (AMF) was used to determine the bioaccessible potentially toxic element (PTE) fraction in the upper airways, in sequence with the simplified bioaccessibility extraction test (SBET) or the stomach phase of the unified bioaccessibility method (gastric fluid only) (UBMG). Filter dynamic measurement system TX40 filters smeared with soil reference material (BGS RM 102) were used as test samples. Analysis was performed by ICP-MS. Comparison between results obtained for soil alone and when the soil was supported on TX40 filters indicated that the presence of the substrate did not affect the extraction efficiency, although a large Zn blank was detected. The sequential AMF→SBET extraction liberated similar amounts of Fe, Mn, Ni and Zn to the SBET alone; but significantly less Cd; and significantly more As, Cr, Cu and Pb. The sequential AMF→UBMG extraction liberated similar amounts of Cd, Cr, Mn and Zn to the UBMG alone, but significantly less As, Fe and Ni; and significantly more Cu and Pb. Enhanced extractability was due to the greater quantities of exchangeable ions and complexing agents present. Adoption of a two-step sequential extraction (AMF followed by either the SBET or the UBMG) is recommended because it is more representative of biological conditions and avoids overestimation or underestimation of bioaccessible PTE concentrations

    Oral bioaccessibility tests to measure potentially toxic elements in inhalable particulate matter collected during routine air quality monitoring

    The simplified bioaccessibility extraction test (SBET) and the stomach phase of the unified bioaccessibility method (UBM) have been modified for use in assessing the bioaccessibility of potentially toxic elements in samples of PM10 collected during routine air quality monitoring. Arsenic, Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn were measured by inductively coupled plasma mass spectrometry in extracts of synthetic PM samples prepared by loading candidate soil reference material BGS102 onto TX40 (Teflon-coated borosilicate) filters widely used in standard commercial tapered element oscillating microbalance/filter dynamics measurement system (FDMS) ambient particulate monitors. Analysis of blanks revealed two important sources of contamination that had to be controlled in order to achieve reproducible results. The syringe filters used in the SBET released Cu and Zn into sample extracts during filtration and had to be washed with 80 mL of 0.4 M glycine at pH 1.5 immediately prior to use, whilst the FDMS filters used to collect PM10 were found to contain sufficient extractable Zn (ca. 3 μg per filter) to almost double the concentration released by the soil. The latter is a consequence of the use of Zn in filter manufacture and so could not be eliminated, but was successfully overcome by means of blank-subtraction. A ten-fold miniaturisation of the SBET and six-fold miniaturisation of the UBM allowed 0.1 g samples to be processed, with analyte recoveries generally within ±10% of those obtained when the conventional procedures were used. Comparison between results obtained when the modified procedures were applied to soil alone, and when soil was loaded onto FDMS filters, indicated that the presence of the filter had no effect on extraction efficiency, except for Fe, provided blank-correction was performed. Results obtained for As, Cd and Pb when the modified UBM was applied to BGS102 on FDMS filters were 4.40 ± 0.04, 0.224 ± 0.002 and 17.3 ± 0.8 mg kg-1, respectively (n = 3), all within recommended ranges

    An online survey of Australian physicians reported practice with the off-label use of nebulised frusemide

    BACKGROUND: Off-label prescribing is common in palliative care. Despite inconsistent reports of the benefit of nebulised frusemide for breathlessness, its use continues to be reported. METHODS: An online survey was emailed to 249 members of the Australian and New Zealand Society of Palliative Medicine to estimate the use of nebulised frusemide for breathlessness by Australian physicians involved in palliative care in the previous 12 months. RESULTS: There were 52/249 (21%) respondents to the survey. The majority (44/52; 85%) had not prescribed nebulised frusemide in the previous 12 months. The most common (18/44; 43%) reason for not prescribing nebulised frusemide was a belief that there was not enough evidence to support its use. Whilst only a few respondents (8/52; 15%) reported having used nebulised frusemide, all that had used it thought there was at least some benefit in relieving breathlessness. CONCLUSION: This report adds to the series of case studies reporting some benefit from nebulised frusemide in relieving breathlessnes

    Inventory of Pacific Research at Victoria University of Wellington 1999-2005

    This inventory seeks to document the range of research interests and activities taking place on Pacific topics at Victoria University of Wellington (VUW). It has emerged out of a collaboration between the Office of Research and Postgraduate Studies, and the Programme in Pacific Studies. The inventory aims to provide an informed basis for the strategic development of Pacific research and teaching at VUW, through the newly established unit, Va'aomana Pasifika. By "Pacific" we mean the most inhabited portions of the Pacific basin. At times, reference may be made to "Pacific people": by this we mean people who trace descent to andtor are citizens of any of the territories commonly understood to be part of the Pacific (i.e., Melanesia, Micronesia, Polynesia). Alternative terms that may appear in the inventory include: "Pasifika", "Pasefika", "Pasifiki". These are respectively generic Polynesian, and specifically Samoan and Tongan variations on the word "Pacific". The terms "Asia-Pacific" or "Asia Pacific" arise occasionally: we understand that the terms do not always coincide with our emphasis on the Pacific basin and Pacific peoples, and for the most part privilege nations and peoples on the "Pacific Rim". However, we are also aware that some permutations of "Asia-Pacific" at W are more conscientiously engaged with the Pacific basin. We have allowed for the benefit of the doubt in most cases. In this inventory, distinction is made between academic staff who are "Pacific by ethnicity" and academic staff who are "Pacific by interest". Academic staff who are actively engaged in research on Pacific topics may not trace their ancestry to Pacific nations, while academic staff who are "Pacific by ethnicity" may not necessarily be conducting research on Pacific topics. This inventory does not include exclusively Maori research. Inventories of Maori research activity and services at W have already been undertaken through the office of Toiahurei, the Pro Vice Chancellor Maori at VUW. Finally, this research inventory is limited to the period 1999-2005 because of the restrictions of our staffing and research resources. While it would have been useful to have a more extensive inventory, perhaps reaching back 10 or even 30 years, this was beyond our means at the time and could be a future project. We know that we may not have captured all the Pacific research carried out during this period. We apologise for any omissions and welcome such information to add to and assist grow this inventory data base

    Overview and challenges of mercury fractionation and speciation in soils

    Analytical procedures to assess mercury speciation in soils still lack consensus. This article presents an overview of the mercury fractionation and speciation procedures used in soils. Mercury fractionation is the most common approach despite the operational definition of the fractions. Application of single extraction procedures that target the water-soluble, exchangeable and acid-soluble fractions and the application of EPA method 3200 for mercury sequential extraction are examined in soils with different physicochemical characteristics. A step forward in mercury speciation is thermo-desorption, a useful tool to rapidly obtain needed information about contaminated soils. The advantages and limitations of these procedures are compared; the importance of soils’ physicochemical characteristics highlighted. Criteria to be considered when choosing a suitable method are given - assessing total mercury concentration, soil physicochemical characteristics, environmental conditions, and legislation. It is recommended that the interpretation of results is done wisely, to correctly support decisions concerning intervention strategies at contaminated sites

    Microstructural Characterization of LENS\u3csup\u3eTM\u3c/sup\u3e Ti-6Al-4V: Investigating the Effects of Process Variables Across Multiple Deposit Geometries

    Laser based additive manufacturing of Ti-6Al-4V components is under consideration for aerospace applications. The mechanical properties of the finished components depend on their microstructure. Process mapping compares process variables such as heat source power, heat source travel speed, material feed rate, part preheat temperature and feature geometry to process outcomes such as microstructure, melt pool geometry and residual stresses. In this work, the microstructure of two-dimensional pads, multilayer pads, thin walls, and structural components at the steady state location was observed. A method for measuring β grain widths that allows for the calculation of standard deviations, confidence intervals, and variances in grain size was developed. This represents an improvement over the commonly used line-intercept method. The method was used to compare variability of β grain widths across different part geometries. It was found that thin wall parts have the highest β width variability and that the width of the β grains varies more towards the top of multi-layered samples than towards the bottom. Experimental results for α and β grain size across multiple deposit geometries are presented that offer new insight into the effect of process variables on microstructure. β grain widths are also compared for different deposit geometries with the same power, velocity, and feed rate. Single layer pad geometries were found to have the smallest β grain widths, multi-layer pads had larger β grain widths, and thin wall samples had the largest β grain widths. Trends in α width with Vickers hardness were also considered in the context of thermal gradient measurements. Hardness maps were created for the structural component samples. Optical microscopy was used to observe a layering effect in structural component samples. It was found that light and dark bands had different Vickers microhardness values

    Closing the Food Gap in Adams County: A Proposal for Comprehensive Solutions through Community Action

    Today, in Adams County, we have two food systems. The 20% who live in poverty survive on food stamps, the food pantry, church donations, and trips to discount food vendors. Those with more financial stability could choose from an abundance of healthy, sustainably produced, local and international foods. We have come to accept these two food systems as the norm without critically analyzing how it is affecting individuals and the health of our communities. In addition to reducing the disparity and closing the food gap, this initiative is focused on building our local economy and educating our community about nutrition and the benefit of local foods in order to promote a more sustainable social, environmental and economic future. This paper begins an investigation of community food security in Adams County and makes recommendations to start the process of finding stronger community solutions. The data represented here is drawn from community discussions and programmatic statistics. It is an initial assessment to be followed up by a more comprehensive study and analysis
